The economics of microalgal bioproduct commercialization would benefit from increased accumulation of energy storage compounds, such as carbohydrates and lipids, and increased biomass productivity. This thesis explores two adaptive laboratory evolution strategies for improving Scenedesmus obliquus: single UV-mutagenesis and low light cultivation at a high dilution rate to produce cultigens with greater carbohydrate content and iterative UV-mutagenesis and selection under outdoor conditions in a raceway pond at a high dilution rate to increase biomass productivity.

The effort to protect coastal property and infrastructure from storm damage, erosion, and sea level rise has resulted in increased construction of coastal protection structures (CPS) worldwide. Researchers around the globe have found that the marine communities living on CPS differ from those living on natural rock outcroppings in the same area. We conducted a classic disturbance experiment to investigate possible differences in marine organism response and community assembly between natural and human-constructed rocky intertidal habitat along the Massachusetts coast. Two studies on this potential are described in this thesis – one on a locally isolated filamentous yellow-green alga and the other on a planktonic strain genetically improved via selective enrichment.
Polycultures have been viewed as productive, stable, and, in some cases, harvestable by natural bioflocculation. Local native strains might have higher productivity than culture collection strains because they are already adapted to local outdoor conditions. "The elevated toxicity of toxic metals in the presence of nanoparticles (NPs) has raised significant concerns on the NP environmental safety. This research focuses on the impact of environmental conditions, specifically algae, on this type of toxicity. This thesis examines the effect of solids recycling on nitrification, organic carbon removal, and algal-bacterial productivity in bench-scale photobioreactors (PBRs) simulating winter pond conditions in San Luis Obispo, California and a 6-acre raceway system in Delhi, California. Two sets of duplicate photobioreactors operated with or without solids recycling were fed primary clarifier effluent (1oEff) for the first experiment, and facultative pond effluent (FAC) for the second experiment.
In both experiments mean productivity was lower in solids recycled PBRs (3.67 and 2.3 g/m2-day), than in controls without solids recycling (4.15 and 3.9 g/m2-day). When fed …

Disinfection byproducts (DBPs) are formed from the disinfectant (e.g., chlorine) reacting with components of natural organic matter (NOM) in water drawn from surface water supplies, and are considered as the cause of potential serious human health problems. DBP precursors originate in large reservoirs from at least three types of sources: (1) watershed or allochthonous, (2) algal or autochthonous, and (3) bottom sediments or benthic. Cladophora glomerata is a filamentous green alga native to the Great Lakes. However, its nuisance growth in phosphorus rich waters negatively affects the lakes’ aesthetic and water quality. The Great Lakes Cladophora Model (GLCM) v1, developed in 1982, was the first mechanistic model to simulate Cladophora growth basing phosphorus availability and environmental conditions followed by Cladophora Growth Model and GLCM v2. Algal organic matter (AOM) is found in high concentration during algal bloom season in drinking water systems, which is generally categorized into extracellular organic matter (EOM) and intracellular organic matter (IOM). Growth media recycling during algae cultivation is necessary to increase the efficiency and reduce the cost of biofuel production from algae feedstocks. Without recycling media, the cost of algae based biofuel production would be prohibitively high and large scale algae based biofuel production would not be economically viable. Neither the use of algae to clean wastewater, nor the use of photosynthetic organisms to generate biodiesel, are new concepts on their own. By combining these two processes, algal-based wastewater treatment with algal biofuel production, additional benefits can be derived, among which could be a cost-savings. In California the average estimated base production cost per gallon for algal biodiesel is $5.98/gallon. Compared to the adjusted production cost of petroleum-based diesel of $1.53/gallon, biodiesel is not cost-competitive. The high nutrient use in algae cultivation and its recovery and recycling is one of the challenges that may limit the scalability and sustainability of algal biofuels. The present study evaluates the use of Hydrolysate obtained after Flash Hydrolysis (FH) of Scenedesmus at 280 C as a nutrient source for microalgae cultivation. While treating domestic wastewater, algae can be grown and harvested for biofuel production. Water is a vital resource and it is imperative to conserve and reuse as much as possible. Several pilot and lab scale experiments were conducted to further research into a full scale wastewater treatment and biofuel production facility. However, thousands of colonias residents (settlements in the Southwest US that lack access to basic infrastructure) still lack access to clean drinking water. Installations similar to Louisiana sugar mills have been proposed as potential biorefineries for sugarcane, energy cane, and sweet sorghum. The nutrient-rich wastewater generated from the milling process can be used as an algal growth medium. The effectiveness of algae to treat NO2and NO in simulated flue gas was tested using Chlorella vulgaris in photobioreactors (PBRs) using NOxconcentrations between 30 ppm to 780 ppm. Deer Creek Reservoir in Utah has a history of high algae concentrations. Despite recent nutrient reduction efforts, seasonal algae continue to present problems. Cost effective, accurate, and comprehensive monitoring is important to understand the reservoir processes driving this problem and characterizing the algae spatial and temporal distributions are an important part of this effort. Based on results of its Aquatic Species Program (1978-1996), which sought to develop algae-to-liquid fuel technology, the U.S. Department of Energy has suggested that algal wastewater treatment may be incorporated into biodiesel production schemes to reduce the operating costs of both processes. the purpose of the current research was to evaluate the triglycerides produced by wastewater-grown algae for their suitability as a fuel feedstock and to investigate the effectiveness of several solvent mixtures and extraction procedures at recovering lipids from fresh algae.
