Wilsons disease is an autosomal recessive disorder of copper metabolism which results from mutations in the gene encoding for Wilson disease protein (WDP). The cytosolic N-terminus of WDP comprises six metal binding domains each containing a conserved metal binding motif, GMXCXXC. The contribution of these metal binding domains to copper homeostasis is poorly understood. We have cloned, expressed and purified residues 485 to 633 corresponding to N-terminal metal-binding domains 5 and 6 of WDP (WD5-6). Two of the 3 disease causing mutations in the N-terminal metal binding domains of WDP occur one in each of these domains and the two …

A Study Of How Precursor Key Concepts For Organic Chemistry Success Are Understood By General Chemistry Students, Patrick Gerard Meyer

This study examines college student understanding of key concepts that will support future organic chemistry success as determined by university instructors. During four one-hour individual interviews the sixteen subjects attempted to solve general chemistry problems. A think-aloud protocol was used along with a whiteboard where the students could draw and illustrate their ideas. The protocols for the interviews were adapted from the Covalent Structure and Bonding two-tiered multiple choice diagnostic instrument (Peterson, Treagust, & Garnett, 1989) and augmented by the Geometry and Polarity of Moleculessingle-tiered multiple choice instrument (Furió & Calatayud, 1996). A three-fold exercise was conducted to assess the groundwater potentiality in the Eastern Desert (ED) of Egypt. First, a database was generated to host all relevant data sets in a GIS environment for a better understanding of the spatial relationships between these data sets. Co-registered digital mosaics were generated from relevant data sets including remote sensing (e.g., Landsat TM, SIR-C, SRTM, TRMM), geochemical (solute concentrations, O and H stable isotope composition), geological (geologic maps), and hydrological (lithology, depth to water table) data sets. Pitzer's Ion-interaction Model had been widely used in predicting the non-ideal behavior of solution over a wide range of conditions for natural water systems. The model is successful only when there are sufficient parameters available. However, generating these needed parameters requires a great quantity of work and it would be advantageous to be able to predict the Pitzer's parameters for salt systems when there is no data available.

In this dissertation, we present an online problem solving environment for multivariate numerical integration. To build an environment for users with various backgrounds, we address issues of Web interface support for end-users, remote application programming interfaces for distributed system programmers, visualization of multivariate integrand functions for problem analysis, and highly efficient integration methods. Compared to previous implementations, our new load balancing schemes increase the scalability of parallel integration and hence are suitable for large scale computations. Interaction between synchrotron radiation with inner-shell electrons is a fundamental method to study the structure and dynamics of atoms and molecules. This thesis, under the guidance of Dr. Nora Berrah, concentrates on the study of the interaction between soft x-ray photons with Ar atoms and HBr moleculeswith time-of-flight (TOF) electron spectrometers and two-dimensional photoelectron spectroscopy (2DPES). Improved prediction of the transport and bioavailability of metals in contaminated sediments demands a better quantitative understanding of how metals partition between aqueous solutions and mineral phases. This research examines the partitioning of nickel in a contaminated aquifer from Grand Rapids, Ml. Contaminated aquifer sediments were examined using the five step Tessier et al. (1979) operationally defined sequential extraction procedure to determine the solid phase distribution of nickel in the aquifer sediments. Models used to guide remediation efforts at the contaminated site assume that Ni behavior can be predicted based on Ni adsorption onto ferrihydrite (HFO). Although sequential extraction results …

Some hydrocarbon reservoirs have a documented reducing chimney, extending upward to the surface due to diffusion of hydrocarbon volatiles. This effect reportedly reduces the iron in the soil to magnetite and precipitates fine-grained pyrite if there is sufficient sulfur in the system. This study was conducted to test the detectability of the hypothetical chimney, using electrical geophysics in an environment of relatively recent glacial cover, known depth to an underlying petroleum reservoir, and known faulting.

Carbonic anhydrase II (CA II) is a zinc metalloenzyme that catalyzes the reversible hydration of carbon dioxide. The role of this reaction in physiology has made it a target of many therapeutic approaches. Most CA II inhibitors currently known are sulfonamide-based and this is the only functional group incorporated into rationally designed CA II inhibitors. Thioxolone (6-hydroxy-1, 3-benzoxathiol-2-one), is a non-sulfonamide that has been found to have CA II inhibitory activity.

The goal of our work is to gain more insight into the vortex dynamics in YBa2Cu307-8 single crystals with point defects induced by 9MeV proton irradiation. The position of the critical points from the first order melting transition line can be shifted in a controlled manner with the induced disorder. The central hypothesis that nanoscale self-assemblies can provide excellent metal ion recognition has been substantiated by employing acylpyrazolones and trivalent lanthanide metal ions as model systems. Several novel acylpyrazolones and their amphiphilic analogs have been designed, synthesized, and characterized. Their lanthanide metal ion recognition efficacies have been demonstrated through baseline separations of a mixture of light, middle, and heavy lanthanide metal ions by employing them in the aqueous mobile phase of high performance liquid chromatography (HPLC) with octadecylsilanized silica (ODS) as the stationary phase. Translational energy-gain spectroscopy (TES) has been used to investigate state-selective dissociative and non-dissociative single-electron capture processes in low-energy collisions of He2+ ions with O2 , H2 O, CO2 , N2 , and NH3 at impact energies between 100 eV and 1600 eV. The measured energy spectra for theHe2+ -O2 and H2 O collision systems show that the dominant exit channel is due to dissociative transfer ionization (DTI), single-electron captureaccompanied by ionization of the molecular target-ion, at the lowest collision energies. Zinc sulphide quantum dots were synthesized in aqueous media using sodium polyphosphate as the stabilizer. The nanoparticles were 2.5 ± 0.5 nm and 5.0 ± 0.5 nm diameter depending on the synthetic procedure employed and the amount of Mn doped varied from 0.003 to 0.32% (w/w). The nanoparticles were characterized by UV-Vis absorption, luminescence, EPR, near edge x-ray absorption fine structure (NEXAFS), and x-ray photoelectron spectroscopy (XPS), and x-ray diffraction (XRD), and transmission electron microscopy (TEM). Rank-based estimation methods provide alternatives to least squares. Estimators derived via least squares are generally not robust to aberrant observations.Rank-based methods for linear models generalize traditional Wilcoxon procedures in the simple location models and are robust.

In the usual linear model it is assumed that the errors are independent. In the case of repeated measures data several observations are taken on each experimental unit. In the case of longitudinal data the measures are taken on the same subject over time. As such an independence assumption does not seem valid. This research project focused on novel and efficient synthetic approaches of inosamine derivatives (rhizopines). The preparation of specific rhizopines would help elucidate the microbial pathway of rhizopine catabolism and degradation, and determine the role of corresponding genes during nitrogen fixation. We have investigated regio - and stereoselective transformations of myo-inositol derivative 28 utilizing an organostannane as key intermediate towards rhizopine 1a. The synthesis involved several protection and deprotection strategies to control competing reactivities of six hydroxyl groups of myo-inositol. The modern day landscape of Van Buren County Michigan was created by the advance and retreat of glaciers from about 13,000 to 20,000 years ago. The Lake Michigan Lobe of the Laurentide Ice Sheet sculpted southwest Michigan most recently. The sediments deposited by the glacier include diamicton, outwash sediments, and lacustrine deposits. The build up of sediment by the glaciers led to the construction of particular features such as moraines, till plains, and outwash fans. A detailed investigation of selected mineralogical and textural characteristics of glacial deposits was conducted in order to explore the complex relationships between the Lake Michigan and Saginaw Lobe ice-margins and associated depositional regimes during the deglaciation of southwest Michigan. Various analytical methodologies were applied to the characterization of glacial materials, glacial and post-glacial environments in order to infer a chronology of lobate activity.

The human copper-transporting ATPase Wilson's disease protein was one of the first proteins shown to be actively involved in cellular copper homeostasis. The protein includes six N-terminal metal binding domains. The individual capabilities of the domains to bind and transfer copper are not yet fully understood. Each metal-binding domain is about 72 amino acid residues long and contains the conserved binding motif GMTCXXC, of which the cysteines are involved in the copper coordination. In this thesis we develop two types of structure preserving Jacobi algorithms for com puting the symplectic singular value decomposition of real symplectic matrices and complex symplectic matrices. Unlike general purpose algorithms, these algorithms produce symplectic structure in all factors of the singular value decomposition.

Our first algorithm uses the relation between the singular value decomposition and the polar decomposition to reduce the problem of finding the symplectic singular value decomposition to th a t of calculating the structured spectral decomposition of a doubly structured m atrix. We derive new first-order necessary and sufficient optimality conditions characterizing global minimizers in mathematical programming and optimal controlproblems. These conditions are based on level sets of an objective functional and they do not assume special structure of a problem (convexity, linearity, etc.). For a mathematical programming problem of minimization of a smooth functional on some compact convex set with equality nonlinear constraints, we derive first-order optimality conditions in the form of a generalized Lagrange multiplier rule. This rule should hold for any point from the level set of the objective functional corresponding to a global minimizer. We demonstrate that these …
