Fibronectin binding protein F1 (Sfb1) of Streptococcus pyogenes (group A streptococcus [GAS]) is a well-characterized adhesin that has been shown to induce protection in mice against a lethal intranasal GAS challenge after intranasal immunization with cholera toxin B subunit (CTB) as adjuvant. With a murine skin infection model, we have shown that Sfb1/CTB vaccination neither elicits opsonizing antibodies nor prevents systemic bacterial growth and dissemination to internal organs after a subcutaneous GAS challenge. These results indicate that an Sfb1-based vaccine should be complemented with additional protective antigens in order to be used in areas such as the tropical north of …

We examined breast cancer risk in association with an overall glycemic index (GI), glycemic load (GL), and dietary carbohydrate and sugar intake in a prospective cohort of 49,613 Canadian women enrolled in the National Breast Screening Study who completed a self-administered food frequency questionnaire between 1980 and 1985. Linkages to national mortality and cancer databases yielded data on deaths and cancer incidence, with follow-up ending between 1998 and 2000. During a mean follow-up of 16.6 years, we observed 1,461 incident breast cancer cases. GI, GL, total carbohydrate, and total sugar intake were not associated with breast cancer risk in the …

The Bioconductor R package multtest implements widely applicable resampling-based single-step and stepwise multiple testing procedures (MTP) for controlling a broad class of Type I error rates, in testing problems involving general data generating distributions (with arbitrary dependence structures among variables), null hypotheses, and test statistics. The current version of multtest provides MTPs for tests concerning means, differences in means, and regression parameters in linear and Cox proportional hazards models. Procedures are provided to control Type I error rates defined as tail probabilities for arbitrary functions of the numbers of false positives and rejected hypotheses. Atrazine concentrations in the Big Blue River Basin (BRB) in Nebraska and Kansas periodically exceed the U.S. EPA Maximum Contaminant Level (MCL) of 3 µg L^-1. The present study is focused on watershed variables influencing surface runoff of atrazine. The assessment includes the impact of corn and sorghum planting progress (indicating atrazine application), rainfall, antecedent soil water content, and soil restrictive layer on stream-measured weekly atrazine load in independent BRB subwatersheds for 1997 - 2004. The publication of Rachel Carson’s Silent Spring in 1962 is widely regarded as one of the major events that launched the modern environmental movement. Silent Spring is a compelling blend of stories, natural history, human values, and biological facts. In this essay we consider Carson’s attitude toward animals in Silent Spring and in other texts. Despite the facts that she was raised to love Nature and animals, little direct attention has been given to Carson’s views about our moral responsibilities to, and the moral standing of animals. Significant coral reef community development along the eastern shelf of the United States continues northward of the Florida Keys through Miami-Dade, Broward, Palm Beach, and Martin Counties, Florida (to Latitude 27° N). These Southeast Florida high-latitude coral communities have approximately 30 species of stony corals, stony coral coverage of 2-3%, and a diverse assemblage of reef gorgonians, sponges, and fishes.

This system lays within 3 km of the coast offshore a highly urbanized area comprising a population of over 5 million people (the population of Broward County alone exceeds 1.7 million). These reefs are important economic assets: a 2001 economic …

Great attention and energy has been spent investigating reattachment techniques for dislodged and fragmented scleractinian corals; however there has been a lack of controlled experimentation on how to restore dislodged gorgonians following a disturbance event, such as a ship grounding. Unfortunately, reef damage events occur frequently off southeast Florida. As an example, since 1998 at least five freighters have grounded on the reefs near Ft. Lauderdale, Broward County. These freighters dislodged many scleractinian and gorgonian corals and often destroyed thousands of square feet of reef habitat. After these events, restoration efforts concentrated on stabilizing loose debris and rubble, and reattaching …

Advances have come from such international bodies as the Organisation for Economic Co-operation and Development (OECD), the World Health Organisation (WHO), the European and Mediterranean Plant Protection Organisation (EPPO), and the European Centre for Ecotoxicology and Toxicology of Chemicals (ECETOC) (1–8). Risk assessments have played a critical role in the development of various regulations within the European Commission (EC) as well as in other parts of the world, including the United States, Canada, and Japan (9–17). This study describes the most extensive effort to characterize the chemistry, toxicology and community of the sediments of the Mattaponi and Pamunkey Rivers. This was accomplished using a study design modified to expand the number of stations occupied by reducing the cost of analyses by compositing replicate samples collected from each study site rather than performing toxicity tests on these samples individually. In previous studies, the variability in field replicate samples was equivalent to the variability in laboratory replicates. This design has long been used to analyze samples for various chemical contaminants as a cost savings endeavor. More ....

Conserving amphibian populations requires knowledge of a species and its habitat relationships. The four-toed salamander (Hemidactylium scutatum) is listed as Special Concern in Maine and 11 additional states and provinces, Threatened in Illinois, and Endangered in Indiana (Appendix A). Little is known of H. scutatum ecology despite the species' extensive range. Infrequent sightings of H. scutatum throughout its range may indicate either low numbers or that the species' behavior make detection difficult. Records for H. scutatum in Maine existed from only 32 sites before my study, and the total number of occurrences of this species in Maine is …

The ever increasing need for accurate predictions of global environmental change under greenhouse conditions has sparked immense interest in an abrupt, century‐scale cooling around 8200 years ago, with a focal point in the North Atlantic and with hemispheric teleconnections. Despite considerable progress in the unraveling of this striking feature, including a conceivable driving mechanism (rapid drainage of proglacial Lake Agassiz/Ojibway and a resulting reduced strength of North Atlantic thermohaline circulation), several key questions remain unanswered. It is not well established what effect these materials may have on the immature root dentin. A concern would be the resistance to fracture of the remaining root dentin. This report presents the results of striped bass (Marone saxatilis) tagging and monitoring activities in Virginia during the period 1 August 2003 through 31 August 2004. It includes an assessment of the biological characteristics of striped bass taken from the 2004 spring spawning run, estimates of annual survival based on annual spring tagging, and the results of the fall 2003 directed mortality study that is a collaborative effort with the Maryland Department ofNatural Resources. Bicistronic And Fused Monocistronic Transcripts Are Derived From Adjacent Loci In The Arabidopsis Genome, Jyothi Thimmapuram, Hui Duan, Lei Liu, Mary A. Schuler

Comparisons of full-length cDNAs and genomic DNAs available for Arabidopsis thaliana described here indicate that some adjacent loci are transcribed into extremely long RNAs spanning two annotated genes. Once expressed, some of these transcripts are post-transcriptionally spliced within their coding and intergenic sequences to generate bicistronic transcripts containing two complete open reading frames. Others are spliced to generate monocistronic transcripts coding for fusion proteins with sequences derived from both loci. RT-PCR of several P450 transcripts in this collection indicates that these extended transcripts exist side by side with shorter monocistronic transcripts derived from the individual loci in each pair. An important step in building a multiple regression model is the selection of predictors. In genomic and epidemiologic studies, datasets with a small sample size and a large number of predictors are common. In such settings, most standard methods for identifying a good subset of predictors are unstable. Furthermore, there is an increasing emphasis towards identification of interactions, which has not been studied much in the statistical literature. We propose a method, called BSI (Bayesian Selection of Interactions), for selecting predictors in a regression setting when the number of predictors is considerably larger than the sample size with a focus …

In this study, we tested the hypothesis that a CB₁ TMH3-4-5-6 aromatic microdomain, which includes F3.25(190), F3.36(201), W5.43(280), and W6.48(357), is centrally involved in CB₁ receptor activation, with the F3.36(201)/W6.48(357) interaction key to the maintenance of the CB₁-inactive state. We have shown previously that when F3.36(201), W5.43(280), and W6.48(357) are individually mutated to alanine, a significant reduction in ligand binding affinity is observed in the presence of WIN 55,212-2 and SR141716A but not CP55,940 and anandamide. Eighty published studies were appraised to document the potential stress associated with three routine laboratory procedures commonly performed on animals: handling, blood collection, and orogastric gavage. We defined handling as any non-invasive manipulation occurring as part of routine husbandry, including lifting an animal and cleaning or moving an animal's cage. Significant changes in physiologic parameters correlated with stress (e.g., serum or plasma concentrations of corticosterone, glucose, growth hormone or prolactin, heart rate, blood pressure, and behavior) were associated with all three procedures in multiple species in the studies we examined. The group A Streptococcus (GAS) is an important pathogen responsible for a wide range of human diseases. Fibronectin binding proteins (FBPs) play important role in promoting GAS adherence and invasion of host cells. The gene prtF2 encodes a FBP and is contained in approximately 60% of GAS strains. In the present study we have examined 51 prtF2-positive GAS strains isolated from the Northern Territory of Australia and describe two genotypes of prtF2, which are mutually exclusive. Both genotypes have been previously identified in the literature as pfbp and fbaB. We show these genotypes map to the same chromosomal location within …
