Reduction Of Iron (Iii) And Humic Substances Plays A Major Role In Anaerobic Respiration In An Arctic Peat Soil, David A. Lipson, Mony Jha, Ted K. Raab, Walter C. Oechel
Ted K. Raab
Arctic peat soils contain vast reserves of organic C and are largely anaerobic. However, anaerobic respiration, particularly the role of Fe(III) and humic substances as electron acceptors, is not well understood in such ecosystems. We investigated these processes in a drained thaw lake basin on the Arctic coastal plain near Barrow, Alaska. We measured concentrations of soluble Fe and other potential electron acceptors, described the microbial community, and performed experiments in the laboratory and field to measure net rates of Fe(III) reduction and the relationship of this process to C cycling. In most areas within the basin, aerobic conditions existed …

Leaching Mechanism Of Semiconducting Minerals, Fathi Habashi
Fathi Habashi
The mechanism of leaching of semiconducting minerals such as PbS, ZnS, UO2, etc., has been the subject of intensive speculation by hydrometallurgists in the early 1950s. The electrochemical mechanism proposed in 1970 by the author in volume 2 of his Principles of Extractive Metallurgy avoids the assumption of forming intermediate complexes that cannot be isolated or identified.

Phenotypic Leaf Variation In Avicennia Marina In Tropical Australia: Can Discrete Subpopulations Be Recognised In The Field?, Peter Saenger, Lyndon O. Brooks
Dr Lyndon O Brooks
The geographic patterns of phenotypic variation in leaf morphology traits were studied in the mangrove Avicennia marina (Forsk.) Vierh. in tropical Queensland, Australia, to determine whether discrete subpopulations could be recognised in the field. Significant differences in the various leaf characters occurred among the sites, which were not explained by longitude or latitude, nor by inter-site distances. Hierarchical cluster analysis of the estimated site means showed no coherent geographical groupings of the sites, suggesting that site populations do not follow a differentiation by distance model. Principal component analysis showed that site groupings with consistent leaf morphological characteristics could be identified, …

A Novel Totivirus And Piscine Reovirus (Prv) In Atlantic Salmon (Salmo Salar) With Cardiomyopathy Syndrome (Cms), Torstein Tengs
Dr. Torstein Tengs
BACKGROUNDCardiomyopathy syndrome (CMS) is a severe disease affecting large farmed Atlantic salmon. Mortality often appears without prior clinical signs, typically shortly prior to slaughter. We recently reported the finding and the complete genomic sequence of a novel piscine reovirus (PRV), which is associated with another cardiac disease in Atlantic salmon; heart and skeletal muscle inflammation (HSMI). In the present work we have studied whether PRV or other infectious agents may be involved in the etiology of CMS.RESULTSUsing high throughput sequencing on heart samples from natural outbreaks of CMS and from fish experimentally challenged with material from fish diagnosed with CMS …

Attribution Of Spatial And Temporal Variations In Terrestrial Methane Flux Over North America, X. F. Xu, Hanqin Tian, C. Zhang, M. L. Liu, Wei Ren, G. S. Chen, Chaoqun (Crystal) Lu, L. Bruhwiler
Chaoqun (Crystal) Lu
The attribution of spatial and temporal variations in terrestrial methane (CH4) flux is essential for assessing and mitigating CH4 emission from terrestrial ecosystems. In this study, we used a process-based model, the Dynamic Land Ecosystem Model (DLEM), in conjunction with spatial data of six major environmental factors to attribute the spatial and temporal variations in the terrestrial methane (CH4) flux over North America from 1979 to 2008 to six individual driving factors and their interaction. Over the past three decades, our simulations indicate that global change factors accumulatively contributed 23.51 ± 9.61 T g CH4-C (1 Tg = 1012 g) …

Investigation Of The Gas Phase Reactivity Of The 1-Adamantyl Radical Using A Distonic Radical Anion Approach, D. G. Harman, Stephen J. Blanksby
Stephen Blanksby
The gas phase reactions of the bridgehead 3-carboxylato-1-adamantyl radical anion were observed with a series of neutral reagents using a modified electrospray ionisation linear ion trap mass spectrometer. This distonic radical anion was observed to undergo processes suggestive of radical reactivity including radical-radical combination reactions, substitution reactions and addition to carbon-carbon double bonds. The rate constants for reactions of the 3-carboxylato-1-adamantyl radical anion with the following reagents were measured (in units 10 12 cm3 molecule 1 s 1): 18O2 (85±4), NO (38.4±0.4), I2 (50±50), Br2 (8±2), CH3SSCH3 (12±2), styrene (1.20±0.03), CHCl3 (H abstraction 0.41±0.06, Cl abstraction 0.65±0.1), CDCl3 (D abstraction …

Photodetachment of the oxyallyl radical anion leads to formation of the oxyallyl diradical, an elusive transient molecule involved in many organic reactions. As described by W. C. Lineberger et al. in their Communication on page 8509 ff., the photoelectron spectrum reveals that the oxyallyl ground state is singlet and the lowest triplet state is only 55 meV higher in energy. The spectral profile indicates that the planar singlet state is the transition state for ringopening of cyclopropanone, whilst the CCC bending motion is activated upon photodetachment to the triplet state.

Online Ozonolysis Methods For The Determination Of Double Bond Position In Unsaturated Lipids, Michael Thomas, Todd W. Mitchell, Stephen J. Blanksby
Stephen Blanksby
Modern lipidomics relies heavily on mass spectrometry for the structural characterization and quantification of lipids of biological origins. Structural information is gained by tandem mass spectrometry (MS/MS) whereby lipid ions are fragmented to elucidate lipid class, fatty acid chain length, and degree of unsaturation. Unfortunately, however, in most cases double bond position cannot be assigned based on MS/MS data alone and thus significant structural diversity is hidden from such analyses. For this reason, we have developed two online methods for determining double bond position within unsaturated lipids; ozone electrospray ionization mass spectrometry (OzESI-MS) and ozone-induced dissociation (OzID). Both techniques utilize …

Identification Of Abundant Alkyl Ether Glycerophospholipids In The Human Lens By Tandem Mass Spectrometry Techniques, Roger J. Truscott, Jane Deeley, Todd W. Mitchell, Michael Thomas, Stephen J. Blanksby
Stephen Blanksby
Previous studies have shown that the human lens contains glycerophospholipids with ether linkages. These lipids differ from conventional glycerophospholipids in that the sn-1 substituent is attached to the glycerol backbone via an 1-O-alkyl or an 1-O-alk-1'-enyl ether rather than an ester bond. The present investigation employed a combination of collision-induced dissociation (CID) and ozone-induced dissociation (OzID) to unambiguously distinguish such 1-O-alkyl and 1-O-alk-1'-enyl ethers. Using these methodologies the human lens was found to contain several abundant 1-O-alkyl glycerophos-phoethanolamines, including GPEtn(16:0e/9Z-18:1), GPEtn(11Z-18:1e/9Z-18:1), and GPEtn(18:0e/9Z-18:1), as well as a related series of unusual 1-O-alkyl glycerophosphoserines, including GPSer(16:0e/9Z-18:1), GPSer(11Z-18:1e/9Z-18:1), GPSer(18:0e/9Z-18:1) that to our …

Fragmentation Pathways Of 2,3-Dimethyl-2,3-Dinitrobutane Cations In The Gas Phase, Martin Paine, Benjamin Kirk, Simon Ellis-Steinborner, Stephen J. Blanksby
Stephen Blanksby
2,3-Dimethyl-2,3-dinitrobutane (DMNB) is an explosive taggant added to plastic explosives during manufacture making them more susceptible to vapour-phase detection systems. In this study, the formation and detection of gas-phase [M+H](+), [M+Li](+), [M+NH4](+) and [M+Na](+) adducts of DMNB was achieved using electrospray ionisation on a triple quadrupole mass spectrometer. The [M+H](+) ion abundance was found to have a strong dependence on ion source temperature, decreasing markedly at source temperatures above 50 degrees C. In contrast, the [M+Na](+) ion demonstrated increasing ion abundance at source temperatures up to 105 degrees C. The relative susceptibility of DMNB adduct ions toward dissociation was investigated …

The alkaline perhydrolysis of the nerve agent O-ethyl S-[2-(diisopropylamino)ethyl] methylphosphonothioate (VX) was investigated by studying the ion-molecule reactions of HOO- with O,S-dimethyl methylphosphonothioate in a modified linear ion-trap mass spectrometer. In addition to simple proton transfer, two other abundant product ions are observed at m/z 125 and 109 corresponding to the S-methyl methylphosphonothioate and methyl methylphosphonate anions, respectively. The structure of these product ions is demonstrated by a combination of collision-induced dissociation and isotope-labeling experiments that also provide evidence for their formation by nucleophilic reaction pathways, namely, (i) S(N)2 at carbon to yield the S-methyl methylphosphonothioate anion and (ii) nucleophilic …

In order to assist with the development of more selective and sensitive methods for thyroid hormone analysis the [M-H]– anions of the iodothyronines; T4, T3, rT3, (3,5)-T2 and the non-iodinated thyronine (T0) have been generated by negative ion electrospray mass spectrometry. Tandem mass spectra of these ions were recorded on a triple quadrupole mass spectrometer and show a strong analogy with the fragmentation pathways of the parent compound, tyrosine. All iodothyronines also show significant abundances of the iodide anion in their tandem mass spectra, which represents an attractive target for MRM analysis, given that iodothyronines are the only iodine bearing …

A Comparison Of The Gas Phase Acidities Of Phospholipid Headgroups:Experimental And Computational Studies, M. C. Thomas, Todd Mitchell, Stephen J. Blanksby
Stephen Blanksby
Proton-bound dimers consisting of two glycerophospholipids with different headgroups were prepared using negative ion electrospray ionization and dissociated in a triple quadrupole mass spectrometer. Analysis of the tandem mass spectra of the dimers using the kinetic method provides, for the first time, an order of acidity for the phospholipid classes in the gas phase of PE < PA << PG < PS < PI. Hybrid density functional calculations on model phospholipids were used to predict the absolute deprotonation enthalpies of the phospholipid classes from isodesmic proton transfer reactions with phosphoric acid. The computational data largely support the experimental acidity trend, with the exception of …

The protection of plant resources in urban areas is a growing conservation concern. Inventory activities that document species presence and stewardship plans that protect and enhance these areas are needed. The results of a botanical inventory of the Crooked Creek Community Juan Solomon Park in Indianapolis, Indiana, are reported in this paper. The 46-acre park contains three distinct habitats, supporting a wide variety of plants. One hundred seventy-nine vascular plant species from 64 families were identified, including 53 (29.6%) non-native species that are naturalizing within the park. Despite its high percentage of alien species and urban setting, the park is …

The deluge of scientific research data has excited the general public, as well as the scientific community, with the possibilities for better understanding of scientific problems, from climate to culture. For data to be available, researchers must be willing and able to share them. The policies of governments, funding agencies, journals, and university tenure and promotion committees also influence how, when, and whether research data are shared. Data are complex objects. Their purposes and the methods by which they are produced vary widely across scientific fields, as do the criteria for sharing them. To address these challenges, it is necessary …

Background: Data generated using ‘omics’ technologies are characterized by high dimensionality, where the number of features measured per subject vastly exceeds the number of subjects in the study. In this paper, we consider issues relevant in the design of biomedical studies in which the goal is the discovery of a subset of features and an associated algorithm that can predict a binary outcome, such as disease status. We compare the performance of four commonly used classifiers (K-Nearest Neighbors, Prediction Analysis for Microarrays, Random Forests and Support Vector Machines) in high-dimensionality data settings. We evaluate the effects of varying levels of …
Genetic Analysis Of The Federally Endangered Winged Mapleleaf Mussel To Aid Proposed Re-Introduction Efforts, Kevin J. Roe
Genetic Analysis Of The Federally Endangered Winged Mapleleaf Mussel To Aid Proposed Re-Introduction Efforts, Kevin J. Roe
Kevin J. Roe
The winged mapleleaf, Quadrula fragosa, historically occurred in the Mississippi, Tennessee, Ohio, and Cumberland river drainages, but has suffered severe population and range reductions. At the time that the species was federally listed as endangered, its range was thought to have been reduced to a stretch of the St. Croix River between northwestern Wisconsin and east-central Minnesota. Recently, morphologically “Q. fragosa-like” specimens were discovered at sites in Arkansas (Ouachita River and Saline River), Missouri (Bourbeuse River), and Oklahoma (Little River). Subsequently, a plan was proposed to re-introduce Q. fragosa into portions of its historic range where its been extirpated from …

Continental-scale estimations of terrestrial methane (CH4) and nitrous oxide (N2O) fluxes over a long time period are crucial to accurately assess the global balance of greenhouse gases and enhance our understanding and prediction of global climate change and terrestrial ecosystem feedbacks. Using a process-based global biogeochemical model, the Dynamic Land Ecosystem Model (DLEM), we quantified simultaneously CH4 and N2O fluxes in North America's terrestrial ecosystems from 1979 to 2008. During the past 30 years, approximately 14.69 ± 1.64 T g C a−1 (1 T g = 1012 g) of CH4, and 1.94 ± 0.1 T g N a−1 of N2O …
