Lef-1 mRNA levels were statistically elevated in cases of ovarian, fallopian tube or peritoneal cancer when compared to non-cancerous controls. Among cancer cases, levels of Lef-1 were statistically different between stage and histology. Lef-1 overexpression may be predictive of poor overall survival. These findings suggest that Lef-1 overexpression may contribute to ovarian, fallopian tube and peritoneal carcinogenesis, and that further investigation is warranted.

Mutations of the p53 tumor suppressor gene are associated with large differences (>7 vessels/HPF) in Microvessel density (MVD) counts between primary and metastatic tumor sites in patients with epithelial ovarian cancer. These data are consistent with models demonstrating p53 mutation functions directly to influence angiogenesis. This information supports continued therapy and research involving angiogenesis inhibitors in patients with ovarian cancer, especially in the setting of increased differences in MVD between primary and metastatic sites.

A simple meta-analysis of eight microRNA (miRNA) expression surveys of endometrial cancers reveals a panel of sixteen miRNAs that are significantly over-expressed (n = 15) or under-expressed (n = 1) in at least three surveys. Examination of these miRNAs indicates that they target mRNAs involved in a number of basic cellular processes including the crucial epithelial to mesenchymal transition (EMT) and hypoxia response. The central role played by these miRNAs is reinforced by the demonstration that they are all among the most ancient of all animal miRNAs. Wnt/b-catenin signaling appears to be important in a multitude of cancers. This review article details current knowledge of the Wnt/b-catenin/Lef1 pathway in generalized gland regulation and formation in addition to endometrial cancer. Wnt signaling is critical for the development of the female urogenital system, in particular mullerian duct formation. Dysregulation of this signaling pathway at multiple nodes has been observed in numerous tumor types, including endometrial cancer. In particular, nuclear accumulation of b-catenin correlates with tumor severity. As such, therapeutic modulation of Wnt signaling represents an emerging avenue for the treatment of cancers that rely on this pathway.

Optimal surgical cytoreduction continues to be an important prognostic variable in patients with ovarian cancer independent of p53 mutational status. Among patients with suboptimal cytoreduction a p53 mutation may offer a survival advantage.

Zmiz1 staining is elevated in patients with epithelial ovarian cancer. Tumors that are Zmiz1 positive are associated with mutations of the p53 gene. Zmiz1 overexpression may be associated with decreased survival in patients with epithelial ovarian cancer. Additional studies are needed to more clearly determine the role of Zmiz1 in patients with epithelial ovarian cancer.

H19 is a maternally expressed non-coding RNA located at chromosome 11p15.5 near the reciprocally imprinted insulin-like growth factor 2 (IGF2) gene. Though the function of H19 is unknown, it is transcribed during embryonic development after which transcription is absent in all but a few tissues including cardiac muscle, breast, ovary, uterus, and placenta. Linking H19, miR-675 and RB1 expression with serous tumors of the endometrium suggests that RB1 suppression may be a differentiating event in serous tumorigenesis.
