Evaluation Of Diagnostic Methods For The Detection Of Pestiviruses In Clinical Samples, Peli̇n Tuncer Göktuna, Kadi̇r Yeşi̇lbağ

Turkish Journal of Veterinary & Animal Sciences

There are several commonly used diagnostic methods to detect pestiviruses for routine diagnostic purposes. In the present study, we aimed to compare virus isolation-indirect immunoperoxidase monolayer assay (VI-IIPMA), antigen capture ELISA (ACE), and RT-PCR for the detection of pestiviruses in clinical samples. Out of 246 samples tested (11 serum, 119 swab, 116 tissue), 28 samples (11.39%) were positive and 218 (88.61%) were negative using the VI-IIPMA method. Using ACE, 70 samples (28.46%) were positive and 176 (71.54%) were negative. Finally, using RT-PCR analysis, we detected 19 (7.72%) positive and 227 (92.28%) negative samples. Inconsistencies were observed among results of the …

Turkish Journal of Medical Sciences

Background/aim: Strongyloides stercoralis causes life-threatening hyperinfection or disseminated strongyloidiasis in immunocompromised patients such as HIV-positive, organ transplantation, and cancer patients. This study investigated the presence of strongyloidiasis in immunocompromised patients for the first time in Turkey. Materials and methods: Serum and stool samples were collected from 108 patients (25.9% of them were chronic renal failure and 74.1% were renal transplantation patients) who were admitted to Ege University Medical School in İzmir, located in western Turkey. Serum samples were analyzed by ELISA (DRG, Germany) and the presence of 18S rRNA gene of S. stercoralis was detected in stool samples by real-time …

A Study Of The Chikungunya Virus In Humans In Turkey, Tuğba Atalay, Sedat Kaygusuz, Ahmet Kürşat Azkur Jan 2017

A Study Of The Chikungunya Virus In Humans In Turkey, Tuğba Atalay, Sedat Kaygusuz, Ahmet Kürşat Azkur

Turkish Journal of Medical Sciences

Background/aim: The chikungunya virus (CHIKV) is a mosquito-borne disease and has recently been causing explosive outbreaks. The CHIKV has spread throughout all continents. Although the first chikungunya case imported from India to Turkey was reported in 2012, there is no detailed epidemiologic study in Turkey yet. The aim of this study was to investigate the seroprevalence of the CHIKV in Turkey. Materials and methods: ELISA was used to screen 500 random serum samples of healthy people collected from Kırıkkale, which is located in central Anatolia in Turkey. The results were verified by indirect immunofluorescence test (IIFT). Results: The results showed …

Comparison Of Microscopy, Elisa, And Real-Time Pcr For Detectionof Giardia Intestinalis In Human Stool Specimens, Yunus Emre Beyhan, Zeynep Taş Cengi̇z Jan 2017

Comparison Of Microscopy, Elisa, And Real-Time Pcr For Detectionof Giardia Intestinalis In Human Stool Specimens, Yunus Emre Beyhan, Zeynep Taş Cengi̇z

Turkish Journal of Medical Sciences

Background/aim: This study included patients who had digestive system complaints between August 2015 and October 2015. The research was designed to compare conventional microscopy with an antigen detection ELISA kit and the TaqMan-based real-time PCR (RT-PCR) technique for detection of Giardia intestinalis in human stool specimens. Materials and methods: Samples were concentrated by formalin-ether sedimentation technique and microscopic examinations were carried out on wet mount slides. A commercially available ELISA kit (Giardia CELISA, Cellabs, Brookvale, Australia) was used for immunoassay. DNA was extracted from fecal samples of about 200 mg using the QIAamp Fast DNA Stool Mini Kit (QIAGEN, Hilden, …

Nitric Oxide In Cerebrospinal Fluid Of Central Nervous System Tuberculosis: Correlations With Culture, Antibody Response, And Cell Count, Kavitha Kumar, Prashant Giribhattanavar, Shripad Patil Jan 2017

Nitric Oxide In Cerebrospinal Fluid Of Central Nervous System Tuberculosis: Correlations With Culture, Antibody Response, And Cell Count, Kavitha Kumar, Prashant Giribhattanavar, Shripad Patil

Turkish Journal of Medical Sciences

Background/aim: The role of nitric oxide (NO) has been established in infection over the years. NO functions by inhibiting the growth of intracellular pathogens. The present study was undertaken to ascertain the role of NO in central nervous system (CNS) infection by Mycobacterium tuberculosis. Materials and methods: A total of 781 chronic meningitis cerebrospinal fluid (CSF) samples suspected of CNS tuberculosis (TB) were categorized based on M. tuberculosis culture positivity, anti-TB antibody response, and CSF cell count and were analyzed for NO. Results: We found that NO levels were positive in 10.88% of the CSF samples. Positivity for NO was …

Evidence Of Foot-And-Mouth Disease Virus Excretion In The Milk Ofapparently Healthy Vaccinated Buffaloes In Islamabad, Pakistan, Hafiziftikhar Ahmed, Umer Farooq, Aamer Bin Zahur, Khalid Naeem, Asma Latif, Hamid Irshad Jan 2017

Evidence Of Foot-And-Mouth Disease Virus Excretion In The Milk Ofapparently Healthy Vaccinated Buffaloes In Islamabad, Pakistan, Hafiziftikhar Ahmed, Umer Farooq, Aamer Bin Zahur, Khalid Naeem, Asma Latif, Hamid Irshad

Turkish Journal of Veterinary & Animal Sciences

The aim of the study was to determine the seroprevalence of foot-and-mouth disease (FMD) and isolation of the FMD virus (FMDV) in the milk of apparently healthy Asian buffaloes. A cross-sectional study was conducted in periurban dairy farms (n = 20) on ≥20 animals per farm in the suburbs of Islamabad, Pakistan. Sera samples (n = 200) were collected to monitor FMDV seroconversions. The analysis of serum samples using ELISA indicated a high seroprevalence of FMD (78%; 156/200) in Asian buffaloes. Milk samples were collected from FMDV seropositive animals only, and virus-specific signals were detected in 12.2% (19/156) of the …
