Measurement Of Urine Albumin Levels With A Monoclonal Antibody Based In-House Elisa, Cemaletti̇n Aybay, Resul Karakuş Jan 2003

Measurement Of Urine Albumin Levels With A Monoclonal Antibody Based In-House Elisa, Cemaletti̇n Aybay, Resul Karakuş

Turkish Journal of Medical Sciences

Microalbuminuria is considered to be an early indicator of diabetic nephropathy. We have previously reported the establishment of A-HAlb/98 hybridoma secreting monoclonal antibody that specifically recognizes human serum albumin (HSA). In this report, an in-house enzyme-linked immunosorbent assay (ELISA) was developed using this antibody for the measurement of urinary albumin (UA) and UA was examined in 24 different samples. A-HAlb/98 was used as the capture antibody by immobilizing onto microtitre plates in ELISA format. HSA-specific polyclonal rabbit immunoglobulin was labelled with biotin and then used as the tracer antibody. The dynamic assay range of ELISA for albumin was established to …

Development Of A Diagnostic And Screening Elisa System For Measuring Tetanus Antitoxoid Levels, Cemaletti̇n Aybay, Resul Karakuş, Ahmet Gökhan Gündoğdu Jan 2003

Development Of A Diagnostic And Screening Elisa System For Measuring Tetanus Antitoxoid Levels, Cemaletti̇n Aybay, Resul Karakuş, Ahmet Gökhan Gündoğdu

Turkish Journal of Medical Sciences

Tetanus is a vaccine-preventable disease of global importance. When acquired it has a high mortality rate. Screenings of special subpopulations such as women of childbearing age provide important clues about tetanus vaccination coverage, and tools for mass protective measures, e.g, protective measures for neonatal tetanus. In addition to its reliability and sensitivity, the main advantage of a mass-screening tool should be its cost-effectiveness. We aimed to develop a cost-effective and sensitive enzyme-linked immunosorbent assay (ELISA) that can be used for diagnostic research as well as mass screening purposes for antitetanus antibodies. Our in-house antitetanus ELISA was validated and tested for …

Serological And Microbiological Investigation On Turkey Rhinotracheitis Virus (Trtv) Infection Of Chickens And Turkeys, Yavuz Çokal, Ayşi̇n Şen Jan 2003

Serological And Microbiological Investigation On Turkey Rhinotracheitis Virus (Trtv) Infection Of Chickens And Turkeys, Yavuz Çokal, Ayşi̇n Şen

Turkish Journal of Veterinary & Animal Sciences

A total of 426 serum samples were examined from 9 broiler and 7 broiler breeder flocks showing clinical signs of swollen head syndrome (SHS). Turkey rhinotracheitis virus antibodies in 8 broiler flocks were between 31.8 and 75.9% and in broiler breeder flocks between 5.4 and 48.6%, with a commercial ELISA. Typical clinical symptoms were observed for SHS in broilers and broiler breeders. During the study, turkey flocks showing clinical signs were not detected. The antibodies to TRTV were between 5.5 and 80.0% in 7 out of 10 broiler flocks, and between 20.0 and 57.1% in 3 out of 4 fattening …

Determination Of Bacteriological Qualities And Aflatoxin M_1 Levels Of Commercially Available Fresh Kashar Cheeses, Uğur Günşen, İlhan Büyükyörük Jan 2003

Determination Of Bacteriological Qualities And Aflatoxin M_1 Levels Of Commercially Available Fresh Kashar Cheeses, Uğur Günşen, İlhan Büyükyörük

Turkish Journal of Veterinary & Animal Sciences

In the present study, 125 samples of commercially available vacuum packaged fresh kashar cheese randomly collected between October 1999 and December 2000 were analysed for coliform bacteria, Escherichia coli, Salmonella, Staphylococcus aureus, moulds and yeasts and levels of aflatoxin M_1 (AFM_1). AFM_1 levels were determined using ELISA. The mean levels of coliform bacteria, S. aureus, moulds and yeasts and AFM_1 were 3.9 x 10^3, 6.25 x 10^2, 2.4 x 10^4 cfu/g and 206.23 ± 15.88 ng/kg, respectively. E. coli was identified in five of the 18 samples (27.8%) positive for coliform bacteria. Salmonella was not isolated from any of these …

Comparison Of Ifat And Elisa Results In Mice Experimentally Infected With Toxocara Canis, Hasan Ayçi̇çek, Mehmet Tanyüksel Jan 2003

Comparison Of Ifat And Elisa Results In Mice Experimentally Infected With Toxocara Canis, Hasan Ayçi̇çek, Mehmet Tanyüksel

Turkish Journal of Veterinary & Animal Sciences

Toxocariasis is a disease which indicates a wide symptomatic spectrum and occurs usually by the ingestion of infective Toxocara canis (rarely Toxocara cati) eggs by humans as a paratenic host. In this study, the sera samples of experimentally infected mice were evaluated on different days by IFAT and ELISA. Regarding the sera samples obtained on the 15^{th} and following days of the study, the groups seropositivity in IFAT by adult and larval sectional antigens and in TES-ELISA were 70, 73.3 and 96.7%, respectively. In the control group, seronegativity in IFAT by adult and larval sectional antigens, and in T. canis …

Developing Elisa For Antibodies To Rinderpest Virus (Rpv) By Using Reference And Partially Purified Rpv Antigens, Aykut Özdarendeli̇, Hakan Bulut, Mehmet Zi̇ya Doymaz, Yusuf Bolat Jan 2003

Developing Elisa For Antibodies To Rinderpest Virus (Rpv) By Using Reference And Partially Purified Rpv Antigens, Aykut Özdarendeli̇, Hakan Bulut, Mehmet Zi̇ya Doymaz, Yusuf Bolat

Turkish Journal of Veterinary & Animal Sciences

The aim of this study was to develop ELISA to examine antibodies against rinderpest virus (RPV) for vaccination programmes in the Malatya and Elazığ regions. Serum samples from 180 clinically healthy cattle in Malatya were obtained in the spring of 1995 and 157 serum samples from cattle were taken in the spring of 1996 in Elazığ. The 337 serum samples were investigated with ELISA. To design the ELISA, reference purified RPV antigen obtained from Pirbright Animal Health Institute was used. RPV RBOK vaccine strain was provided by Central Veterinary Control and Research Institute, Ankara. It was used to infect the …

Level Of Aflatoxin In Some Fish Feeds From Fish Farming Processes, Feed Factories And Imported Feeds, Gülşen Altuğ, Gülsün Özyurt Jan 2003

Level Of Aflatoxin In Some Fish Feeds From Fish Farming Processes, Feed Factories And Imported Feeds, Gülşen Altuğ, Gülsün Özyurt

Turkish Journal of Veterinary & Animal Sciences

Aflatoxins that are toxic metabolites for human and animals were determined in some fish feed. Eighty-five unit samples taken from "fish farming processes", "feed factories" and "imported feeds" in 1998, 1999 and 2000 were analyzed. In the analysis, thin layer chromatography (TLC) and enzyme linked immunosorbent assay (ELISA) technique were used. Consequently, aflatoxin levels above 20 ppb were detected in 20 samples and from 21.2 to 42.4 ppb in 85 samples. In 22 samples aflatoxin levels from 5.0 to 20.0 ppb were detected. Aflatoxin B1 analysis was performed on all samples with aflatoxin levels of more than 20 ppb. Most …
