Chronic inflammation increases the risk of developing multiple chronic diseases, including cancer. The risk of developing colorectal cancer (CRC) specifically is increased in individuals who suffer from colitis, a characteristic of Inflammatory Bowel Disease (IBD). Diet is another risk factor for developing CRC, particularly an inflammation-promoting Western-type diet. Thus, supplementing a Western diet with functional foods containing anti-inflammatory polyphenols is a potential approach to decreasing CRC risk by reducing gut inflammation and altering gut microbiome profile. Atrial Fibrillation (AF) is the most common cardiac arrythmia in adults with likely sex-specific risk factors. Female sex hormones may be important in modulating risk for atrial fibrillation. We hypothesize that pregnancy and progesterone (P4), a hormone found in high levels during pregnancy with abrupt withdrawal immediately after parturition, modulates AF susceptibility in female goats. Cardiac specific TGF-?1 transgenic female goats and age-matched wild-type (WT) female goats were utilized. Pacemakers were implanted in all animals for continuous arrhythmia monitoring and AF inducibility. AF inducibility was evaluated using 5 separate 10 s bursts of atrial pacing (160 - 200 ms). In …

Morbidity in feedlot cattle due to disease is a common economical loss for feedlot producers. Utah is not typically considered a ‘feedlot state’, but there are several producers in the southern part of the state that specialize in receiving at-risk cattle. These cattle are at-risk because they are coming from areas known to be mineral deficient. Areas such as the western US, are known to be deficient in several trace minerals important in immune response. Declining body-wide levels of nicotinamide adenine dinucleotide (NAD) that are present in normal aging men are linked to decreased sperm quality and DNA integrity. When NAD is low, the amount of oxidative stress on the DNA appears to be increased, due to the limited regeneration of the antioxidant glutathione, which is NAD-dependent. We therefore hypothesize that the increased sperm DNA damage in the aging male is caused by an elevated formation of 8-oxoguanine in the DNA, caused by increased oxidative stress when NAD levels are low. Mitochondrial heteroplasmy, the presence of more than one population of mitochondrial DNA (mtDNA) in one cell, can cause metabolic issues and reduced ATP synthesis, even at relatively low levels. The presence of this condition in somatic cell nuclear transfer (SCNT) embryos likely contributes to reduced prenatal and postnatal viability: the majority of the oocyte’s mitochondria is still present at the time of nuclear transfer, as well as through any further embryonic development post-fusion and activation. Since 1980, the average age of first-time fathers has been increasing. With increased age, sperm chromatin quality and nicotinamide adenine dinucleotide (NAD) levels are reduced. In order to condense around protamines, sperm DNA undergoes breaks, at which time epigenetic reprogramming occurs. In humans, about 2% of DNA stays wound to histones in spermatozoa. Histones carry post-translational modifications that control gene activity and have the potential to transmit epigenetic information to the offspring. Mastitis, an infection of the mammary gland, is the most common and expensive animal health problem for the dairy industry and affects every dairy farm to some degree. Zika virus (ZIKV) causes no-to-mild symptoms or severe neurological disorders. To investigate the importance of viral and host genetic variations in determining ZIKV infection outcomes, we created three full-length infectious cDNA clones as bacterial artificial chromosomes for each of three spatiotemporally distinct and genetically divergent ZIKVs: MR-766 (Uganda, 1947), P6-740 (Malaysia, 1966), and PRVABC-59 (Puerto Rico, 2015). The composition of the gut microbiome can be affected by environmental factors, such as diet. The Western dietary pattern is associated with microbiome dysbiosis and adverse health outcomes, including obesity and metabolic disorders. Bovine mastitis, or inflammation of the mammary gland, has substantial economic and animal welfare implications. A genetic basis for mastitis resistance traits is recognized and can be used to guide selective breeding programs. The discovery of regions of the genome associated with mastitis resistance, and knowledge of the underlying molecular mechanisms responsible, can facilitate development of efficient mastitis control and therapeutic strategies. Zika virus (ZIKV) infection of pregnant women is associated with pathologic complications of fetal development. Here, we infect pregnant rhesus macaques (Macaca mulatta) with a minimally passaged ZIKV isolate from Rio de Janeiro, where a high rate of fetal development complications was observed. The infection of pregnant macaques with this virus results in maternal viremia, virus crossing into the amniotic fluid (AF), and in utero fetal deaths. We also treated three additional ZIKV-infected pregnant macaques with a cocktail of ZIKV-neutralizing human monoclonal antibodies (nmAbs) at peak viremia. Enterovirus D68 (EV-D68) virus has become more prevalent over the last 15 to 20 years. EV-D68 attacks the respiratory system and can cause severe disease in individuals who have underlying respiratory problems. There have also been reports of individuals with EV-D68 showing signs of neurological system problems and acute flaccid paralysis. Because of the increase in patients with EV-D68 and also the potential for neurological disease, an animal model is needed to study the disease and to evaluate experimental therapies for EV-D68 infection.

Trophoblast cells from bovine somatic cell nuclear transfer (SCNT) conceptuses express major histocompatibility complex class I (MHC-I) proteins early in gestation, and this may be one cause of the significant first-trimester embryonic mortality observed in these pregnancies. MHC-I homozygous-compatible (n = 9), homozygous-incompatible (n = 8), and heterozygous-incompatible (n = 5) SCNT pregnancies were established. The control group consisted of eight pregnancies produced by artificial insemination. Uterine and placental samples were collected on Day 35 ± 1 of pregnancy, and expression of MHC-I, leukocyte markers, and cytokines were examined by immunohistochemistry. A growing number of arenaviruses can cause a devastating viral hemorrhagic fever (VHF) syndrome. They pose a public health threat as emerging viruses and because of their potential use as bioterror agents. All of the highly pathogenic New World arenaviruses (NWA) phylogenetically segregate into clade B and require maximum biosafety containment facilities for their study. Tacaribe virus (TCRV) is a nonpathogenic member of clade B that is closely related to the VHF arenaviruses at the amino acid level. The data for this cross-sectional retrospective study are from surveys of 65 dairy-cattle herds in central New York, USA sampled between February, 1993 and March, 1995. The objective was to identify probability distributions of logarithmically transformed somatic-cell counts (linear score) for use in a simulation model of mastitis and milk quality. Probability density functions were estimated using maximum-likelihood estimators for the linear score of individual-cow composite milk samples culture negative and culture positive for the pathogens Streptococcus agalactiae, Streptococcus non-agalactiae, Staphylococcus aureus, and coagulase-negative staphylococci for the complete dataset and by bulk-tank somatic-cell count group (< 500 000, ≥ 500 000 SCC/ml). A series of acyclic phosphonomethylether nucleosides were synthesized and then evaluated for inhibitory activity against respiratory viruses of clinical significance using CPE inhibition, neutral red uptake and virus yield reduction assays. Of the 20 compounds synthesized, none significantly inhibited influenza A or B viruses or respiratory syncytial virus strains A2, Long or 18537; the selective indices (SI) were less than 10. A new compound, GS-2128 (2R, 5R-9-[2,5-dihydro-5-(phosphonomethoxy)-2-furanyl]adenine; D4API), selectively inhibited adenovirus 5 (SI>10) as did GS-0577 (9-(3-hydroxy-2-phosphonylmethoxypropyl)-adenine; HPMPA) and GS-0504 [(S)-1-[3-hydroxy-2-(phosphonylmethoxypropyl)]-cytosine; HPMPC]. Milk samples were collected from 108,312 dairy cows during 1601 farm visits made between January 1991 and June 1995. The herd visits were made by personnel from the Central Laboratory of the Quality Milk Promotion Services at Cornell University (Ithaca, NY) to farms located in central New York and northern Pennsylvania. Dairy Herd Improvement Association records were available for 32,978 cows in 327 herds. Intramammary infections, as defined by positive milk cultures, were present in 48.5% of all cows and in 36.3% of cows in herds enrolled in the Dairy Herd Improvement Association. A cell synchronization technique was used that consistently yielded numerous elongated chromosomes, which are necessary for producing high-resolution G-banding. The prometaphase stage of contraction was determined to have the greatest utility for detailed comparative analysis and, therefore, was used in the preparation of the proposed standard high-resolution karyotypes and idiogram.

A proposed standard high-resolution G-band karyotype of the domestic sheep and goat is presented, along with a high-resolution G-band idiogram of the goat. The karyotypes presented contain considerably more bands than contemporary karyotypes and provide much greater resolution for linear differentiation. A reliable technique for obtaining late prophase, prometaphase and greater numbers of metaphase chromosomes is presented. The increased number of mitoses and high resolution chromosomes were obtained using a modification of the typical three-day blood culture technique in which the cell cycle was temporarily blocked at the G1-s border with methotrexate. A high resolution G-band idiogram is presented using a system similar to the standardization of human chromosomes. The use of this technique and system of nomenclature will facilitate routine analyses of horse chromosomes and provide a basis for the identification and recording of chromosome aberrations in this …
