2,4-disulfonyl α-phenyl-tert-butyl nitrone and its pharmaceutically acceptable salts are disclosed. These materials are useful as pharmaceutical agents for oral or parenteral, e.g. intravenous administration to patients suffering from acute central nervous system oxidation as occurs in a stroke or from gradual central nervous system oxidation which can exhibit itself as progressive central nervous system function loss. The materials are also used to ameliorate the side effects of oxidative-damage causing antineoplastic disease treatments.

Compositions for treating tissue damage from ischemia contain PBN, or active derivatives thereof, which are active during ischemia in preventing ATP depletion of the cells which predisposes them to subsequent injury during reperfusion, and which are active during reperfusion as oxygen radical scavengers, in a suitable pharmaceutical carrier for systemic or local administration, especially to the CNS, spinal column and eyes. Based on animal studies, the dosage for treating damage due to stroke is in the range of 10 to 300 mg/kg. Similar dosages are useful in treating damage resulting from free radical generation during inflammation, either as a product …

He was using the family-owned truck to haul a load of logs to the mill. A 21-year-old farm worker was killed in a tractor rollover incident. The victim was driving a tractor on a secondary public road, pulling an unloaded 18-foot flatbed trailer, when he lost control. After the tractor wheels went off the right side of the road, the victim overcorrected, which caused the vehicle to become unbalanced; he was thrown from the tractor as it overturned into a ditch. It was not equipped with an appropriate rollover protective structure (ROPS) or a seatbelt. Novel classes of inhibitors which selectively inhibit the cellular transport of normally transported substances, specifically polyamines are taught which comprise (i) polymers of the transported substance or (ii) protein or polypeptide conjugates of the transported substance. These inhibitors may be used in vitro to assess the effect of the transported substance on cellular functions and in vivo for treating disease conditions involving transport of the particular substance, e.g., a polyamine.

A 22-year-old logger was killed after being struck on the head by a falling snag (dead standing tree). The victim was working with one co-worker, who was selectively cutting trees which were approximately 22 inches in diameter. Standing approximately three feet from the tree being cut, the victim was not wearing a hard hat. The cut tree broke a snag about 35 feet up, causing the snag to fall and strike the victim on the head. Previous studies implicate the nonreceptor protein tyrosine kinase (PTK) p59fyn in the propagation of signals from the B cell antigen receptor. To elucidate the functions of this kinase, we examined B cell responsiveness in mice engineered to lack the hematopoietic isoform of p59fyn. Remarkably, antigen receptor signaling was only modestly defective in fynTnull B cells. In contrast, signaling from the interleukin (IL)-5 receptor which ordinarily provides a comitogenic stimulus with antiimmunoglobulin, was completely blocked. Our results document the importance of p59fynT in IL-5 responses in B cells, and they support a general model …

An apparatus for analyzing the chemical composition of tissue includes a near infrared light source, a fiber-optic probe, a mechanism for directing the light from the light source onto the tissue and detectors for detecting light reflected by the tissue. The light source transmits an incident beam having a wavelength ranging from 1000 to 2500 nm. A compound parabolic concentrator (CPC) is connected to a distal end of the fiber optic probe to focus or concentrate the wavelengths of light simultaneously and in parallel on a particular spot of tissue. Antibodies against HIV-1 proteins in HIV-1-infected individuals share a cross-reactive idiotype defined by the monoclonal antiidiotypic antibody 1F7 (5). Using a computer algorithm based on the molecular recognition theory, regions of inverse hydropathy between the variable sequence of 1F7 and human monoclonal anti-HIV-1 antibodies were identified, which are assumed to be involved in idiotype-antiidiotype contacts. A peptide was designed from the proposed contact in the variable heavy chain framework 3-complementarity determining region 3 (FR3-CDR3) of human antibodies and was synthesized. This peptide is recognized by the antiidiotype 1F7 and inhibits the binding of 1F7 to human anti-HIV-1 antibodies which express …

Fatalities associated with farm tractors are the most common cause of work-related death in the U.S. agricultural industry (1). To characterize farm-tractor-related fatalities in Kentucky, the Kentucky Fatality Assessment and Control Evaluation (KY FACE) Project studied all fatal farm injuries occurring among persons in that state during 1994, the initial year of operation for FACE in Kentucky. This report summarizes the results of that study.

We have generated an IgG1 murine monoclonal anti-idiotype antibody (Ab2) designated 3H1, which mimics a specific epitope on the carcinoembryonic antigen (CEA). Patients with CEA positive tumors are immunologically "tolerant" to CEA. We used 3H1 as a surrogate for CEA for vaccine therapy of 12 patients with advanced colorectal cancer. Each of the patients received a minimum of four intracutaneous injections of aluminum hydroxide precipitated 3H1 at either 1, 2, or 4 mg dosage per injection. 9 of 12 patients demonstrated anti-anti-idiotypic (Ab3) response to 3H1. All nine patients generated specific anti-CEA antibody demonstrated by reactivity with radiolabeled purified CEA; …

A 63-year-old retired bus driver was pinned underneath a rear tractor tire after it overturned while he was bush hogging. The 37-year-old tricycle tractor, not equipped with a ROPS or a seatbelt, had a bush hog attached. On the morning of the incident, the operator and his brother were mowing a portion of a pasture. At about 9:30 am, the victim started mowing at the bottom of a steep slope, and then progressed toward the top of the ridge. A kit and method of non-destructively radiolabelling a biomolecule substrate by halogenation, including the steps of: a) reacting a secondary amine with a chlorinating agent to produce an N-chloro secondary amine, (b) reacting the N-chloro secondary amine with a salt of a radiolabelled halogen to produce a radiolabelled halogenating agent, (c) reacting a solution of the biomolecule substrate with the radiolabelled halogenating agent, and (d) recovering a radiohalogenated biomolecule from the reaction of step (c). The kit and method allow for more complete iodination with a lessening of side reactions such as decomposition of the biomolecule substrate to be labelled.

Compositions containing as the active ingredient a spin-trapping reagent, preferably α-phenyl butyl nitrone (PBN) or spin-trapping derivatives thereof, in a suitable pharmaceutical carrier for administration to a patient are disclosed for treating or preventin symptoms associated with aging or other conditions associated with oxidative tissue damage. A method and apparatus are provided for magnetohydrodynamic acoustic-resonance, near-IR spectroscopy. The method includes a step of applying to a subject under study a magnetic field having a strength between 2,00-10,000 gauss, near-IR radiation having a wavelength between 800-3,000 nm and an acoustic wave having a frequency between 10 khz-1 Mhz. The method also includes the steps of inducing vibration of ions in the magnetic field and detecting an electric wave generated magnetohydrodynamically by the acoustic wave induced vibration of the ions. Lipoprotein lipase (LPL) is a central enzyme in lipoprotein metabolism and is expressed predominantly in adipose tissue and muscle. In these tissues, the regulation of LPL is complex and often opposite in response to the same physiologic stimulus. In addition, much regulation of LPL occurs post-transcriptionally. The human LPL cDNA is characterized by a long 3′-untranslated region, which has two polyadenylation signals. In this report, human adipose tissue expressed two LPL mRNA species (3.2 and 3.6 kb) due to an apparent random choice of sites for mRNA polyadenylation, whereas human skeletal and heart muscle expressed predominantly the longer 3.6-kb mRNA …
