Background/aim: Intravesical BCG treatment fails inexplicably in 30%-45% of patients for high-grade nonmuscle-invasive bladder cancer (NMIBC). We aimed to investigate the role of PD-1/PD-L1 interaction on BCG failure of high-grade NMIBC and to identify biomarkers for predicting BCG responsive cases. Materials and methods: Thirty BCG responsive and 29 nonresponsive NMIBCs were included in the study. Expressions of PDL1(SP-263), MSH2, MSH6, PMS2, and MLH1 were evaluated on pre- and post-BCG transurethral resection (TUR-B) specimens by immunohistochemistry. PD-L1(SP-263) expression was categorised as negative/low, high. DNA mismatch repair protein (MMR) expressions were classified as 'reduced' if ≤30% of nuclei stained, 'preserved' if >30% …

The Importance Of Histological Patterns On Pd-L1 Staining Heterogeneity: Should Weuse Pattern-Based Approach For Selecting Tumor Samples For Pd-L1 Testing In Lungadenocarcinomas?, Pinar Bulutay, Pinar Firat, Emi̇ne Handan Zeren, Suat Erus, Serhan Tanju, Mustafa Şükrü Di̇lege Jan 2021

Background/aim: Programmed death ligand-1 (PD-L1) is a predictive marker for immunotherapeutic agents. However, heterogeneous staining of PD-L1 can cause false-negative results. The aim of this study is to evaluate the importance of histological patterns on PD-L1 staining heterogeneity in lung adenocarcinomas (LAC). Materials and methods: PD-L1 immunohistochemistry (IHC) stain was performed to two different tissue cores of 128 LAC cases, and cut-off values are given for grouping the cases according to the percentage of staining (1%-10%, 11%-49%, 50%-100%). Staining rates between cores were compared and analyzed by their histological patterns. Also, the relation of the PD-L1 expression with the clinicopathological …

Immune Gene Therapy Of Cancer, Hakan Akbulut Jan 2020

Immune Gene Therapy Of Cancer, Hakan Akbulut

Turkish Journal of Medical Sciences

Cancer gene therapy emerged as a promising treatment modality 3 decades ago. However, the failure of the first gene therapy trials in cancer treatment has decreased its popularity. Likewise, immunotherapy has followed a similar course. While it was a popular and promising treatment with IL-2 and interferon and cancer vaccines in the 1980s, it later lost its popularity. Immunotherapy became one of the main options for cancer treatment with the successful use of immune checkpoint inhibitors in clinics approximately 10 years ago. The success of immunotherapy has increased even more with the introduction of cancer gene therapy methods in this …

Evaluation Of Biological Response Modification And Immunotherapeutic Activities Of Barley-Derived Arabinoxylans Against Coccidiosis In Commercial Broilers, Masood Akhtar, Muteen Humayun, Zafar Iqbal, Mian Muhammad Awais, Muhammad Irfan Anwar Jan 2018

Evaluation Of Biological Response Modification And Immunotherapeutic Activities Of Barley-Derived Arabinoxylans Against Coccidiosis In Commercial Broilers, Masood Akhtar, Muteen Humayun, Zafar Iqbal, Mian Muhammad Awais, Muhammad Irfan Anwar

Turkish Journal of Veterinary & Animal Sciences

Plant-derived polysaccharides have been reported for diverse biological response modification (BRM) and immunotherapeutic (IT) activities in animals by activating their defense systems to perform better immunogenically against invading pathogens. Keeping this in view, this study was conducted to assess the BRM and IT activities of barley (Hordeum vulgare L.)-derived arabinoxylans (AXs) against coccidiosis in broilers. Significantly enhanced (P < 0.05) in vivo and in vitro lymphoproliferative responses to T-cell mitogens were revealed in broilers administered barley AXs when compared to the control group. Humoral immune response in terms of antibody titers against sheep erythrocytes was also found higher (P < 0.05) in AX-administered chickens. Furthermore, AX-administered chickens also showed higher weekly weight gains and improved feed conversion ratios as compared to controls. Results of the challenge experiment showed that the percentage of protection and daily weight gains were statistically higher (P < 0.05) whereas mean oocysts per gram of droppings and lesion scores were significantly lower (P < 0.05) in the chickens administered barley AXs as compared to controls. In conclusion, barley-derived AXs led to better growth performance and elicited humoral and cellular immune responses in commercial broilers that persisted against coccidial infection in broiler chickens.

To investigate the necessity and efficacy of immunotherapy in children with allergic eye disease. Materials and methods: The study included 57 patients with allergic eye disease who were followed by the Department of Ophthalmology and Department of Pediatrics at Adnan Menderes University Hospital. Blood and skin prick tests were carried out on 43 patients. Results: There were 38 male and 19 female patients with a mean age of 11 ± 4 years. The mean follow-up was 31 ± 3 months and 28 ± 4 months in 38 vernal keratoconjunctivitis (VKC) and 19 perennial allergic conjunctivitis (PAC) patients, respectively. Immunotherapy was …
