Assessment Of Erythrocyte Morphology In Patients With Type 2 Diabetes Mellitus: A Pilotstudy Of Electron Microscopy-Based Analysis In Relation To Healthy Controls, Tülay Mortaş, Şenay Arikan Durmaz, Şaban Cem Sezen, Yasemi̇n Savranlar

Background/aim: The present study aimed to assess erythrocyte morphology in newly diagnosed type 2 diabetes mellitus patients using scanning electron microscopy. Materials and methods: In total, 30 patients admitted to endocrine outpatient clinics were included in the study. The patients were divided into two groups according to their fasting blood glucose levels: type 2 diabetes mellitus (n = 15, fasting blood glucose levels ≥ 126 mg/dL) and control (n = 15, fasting blood glucose levels < 99 mg/dL). The patient's demographic characteristics, haemoglobin A1c levels, and scanning electron microscopy findings regarding erythrocyte morphology were recorded. Results: There was no significant difference between the control and type 2 diabetes mellitus group in terms of the participants' age (51.13 ± 8.53 vs. 50.33 ± 8.72 years, p = 0.8) and the male/female ratio (9/6 vs. 9/6). In the control group, discocytes were abundant, echinocytes were rare, and spherocytes were absent. On the other hand, discocytes were less common and echinocyte-shaped erythrocytes were more common in the type 2 diabetes mellitus group than in the control group. In addition, spherocytes were detected in the type 2 diabetes mellitus group. Moreover, the diameter of discocytes was significantly lower (p = 0.014), and blood glucose and haemoglobin A1c levels were significantly higher (p < 0.05 for both) in the type 2 diabetes mellitus group than in the control group. Conclusion: Our findings indicate that high glucose levels in type 2 diabetes mellitus patients lead to significant alterations in erythrocyte morphology, including decreased erythrocyte deformability and the formation of echinocytes and spherocytes due to eryptosis. The possibility of decreased erythrocyte deformability due to excessive eryptosis may disturb microcirculation in newly diagnosed, treatment-naïve type 2 diabetes mellitus patients who do not have any complications.

The effects of prestorage leukoreduction and storage duration on the in vitro quality parameters of canine packed red blood cells (pRBCs) stored in saline-adenine-glucose-mannitol (SAGM) were investigated. Hematological, biochemical, and morphological changes in filtered and unfiltered pRBCs were determined each week during 42 days of storage. All of the parameters changed significantly during storage. The effects of leukoreduction on leukocyte and erythrocyte count, packed cell volume, hemoglobin content, and erythrocyte morphology were significant. While supernatant K increased during storage, glucose, Cl, and 2,3-diphosphoglycerate (2,3-DPG) concentrations decreased. The effects of melatonin hormone on rainbow trout (Oncorhynchus mykiss) erythrocyte carbonic anhydrase enzyme purified by using Sepharose 4B-L tyrosine-sulfanylamide affinity gel chromatography were investigated in vitro and in vivo. Carbonic anhydrase from Oncorhynchus mykiss erythrocytes was purified with a specific activity of ~400 EU/mg protein and ~200-fold in a yield of ~20. Melatonin did not inhibit carbonic anhydrase enzyme in vitro. Carbonic anhydrase activity of the control group was determined as 1597.7 ± 429.0 EU/g hemoglobin in vivo. In this paper, a simple and rapid method for the purification of 6-phosphogluconate dehydrogenase from rat erythrocytes together with an analysis of the kinetic behavior and some properties of the enzyme are considered. The purification steps comprised high-speed centrifugation, 20-50% ammonium sulfate precipitation and 2', 5'-ADP Sepharose 4B affinity gel chromatography. The yield was 78.4% and the specific enzyme activity was 5.15 EU/mg proteins. Cyclodextrins are used extensively in food, cosmetic, drug and chemical industries to increase aqueous solubility and stability, and reduce or eliminate the unpleasant taste and smell of many products. Experiments were conducted to examine the effect of a pre-haemolytic dose of methyl b-cyclodextrin (MBCD) on the osmotic fragility of erythrocytes collected from sheep, cattle and humans. Glucose 6-phosphate dehydrogenase (G6PD) was purified from goose erythrocytes and some characteristics of the enzyme were investigated. The purification procedure was composed of 3 steps: hemolysate preparation, ammonium sulfate precipitation, and 2', 5'-ADP Sepharose 4B affinity gel chromatography. Thanks to the 3 consecutive procedures, the enzyme, having a specific activity of 36.2 EU/mg protein, was purified for a yield of 68.79% and 3892 folds; to ascertain enzyme purity, SDS-PAGE was performed. Osmotic fragility and fatty acid composition in erythrocytes of mirror carp (Cyprinus carpio) kept at 8 °C, 16 °C and 28 °C were determined. Osmotic fragility in erythrocytes (EOF) increased significantly (0.19-0.24%) with the low water temperature. Significant declines in the quantity of saturated fatty acid (44.49-38.40%) and monounsaturated fatty acids (26.30-13.99%), and significant increases in that of polyunsaturated fatty acid (29.21-47.59%) were determined (P < 0.05). It was concluded that low water temperature caused a tolerable stress in mirror carps.

The aim of the present study was to define the reference values for erythrocyte and liver tissue glucose-6-phosphate dehydrogenase (G6PDH), glutathione peroxidase (GPX), glutathione reductase (GR) and superoxide dismutase (SOD) activities and also reduced glutathione (GSH) levels in order to obtain basal data for subsequent toxicological investigations. Contradictory results have been reported from various studies regarding the effect of age on susceptibility to oxidative damage. Using different parameters, we carried out a comparative study on the erythrocytes and erythrocyte membranes of healthy elderly and young adults to determine how resistance to oxidative damage is affected by aging. Thin layer chromatography and polyacrylamide gel electrophoresis under denaturing conditions were used to estimate changes in membrane phospholipids and cytoskeletal proteins, respectively. Arginase (L-arginine amidino hydrolase E.C. 3.5.3.1) is an enzyme catalyzing the last stage of the Krebs-Henseleit cycle and, thus, converting arginine into urea and ornithine. Arginase also plays a part in protein and polyamine synthesis and is affected by hormones in terms of metabolic status. In the present study, erythrocyte, saliva, and serum arginase activity together with serum urea and estrogen levels were examined in Akkaraman and Ivesi sheep in pregnancy and postpartum.
