Sperm cryopreservation intensifies the use of assisted reproductive technologies in food animals. However, variations in freezability confront frozen-thawed semen applications. Hence, this groundwork aimed to identify the fundamental differences between high (HF) and low freezability (LF) ejaculated spermatozoa in swamp buffaloes, which were limitedly described for this agricultural species. Twelve (12) different samples of bubaline semen were collected, cryopreserved and categorized into freezability phenotypes using post-thaw sperm motility as the basis. The aim of this study was to investigate the effects of encapsulation with different divalent cations and temperatures on the quality of ram spermatozoa during cold storage. Experiment 1: diluted semen was allocated randomly into three groups: control, encapsulation with calcium (Ca2+) alginate and encapsulation with barium (Ba2+) alginate. The samples were stored at two different temperatures (4 °C or 16 °C) for 24 h and subsequently examined for spermatozoa quality. Comparison Of Different Methods For Sperm Vitality Assessment In Frozen-Thawed Holstein Bull Semen, Kakanang Buranaamnuay

The present study evaluated the accuracy of four methods, i.e. eosin-nigrosin, trypan blue, hypo-osmotic swelling test (HOST) and Hoechst 33342 (H342)/propidium iodide (PI) for assessing bovine sperm vitality. Frozen-thawed semen (n = 30) of Holstein bulls was layeredon 40%/80% percoll solutions to isolate viable spermatozoa. An aliquot of viable spermatozoa was kept at 37°C (live sample); the rest was submitted to cold shock (dead sample). The two aliquots were mixed in three proportions, corresponding to 0%, 50% and100% of viable cells; the sperm vitality was analyzed. Various protocols of feline spermatozoa cryopreservation have been reported and used in clinical practice. To compare the quality of post-thawed epididymal spermatozoa subjected to two different sperm freezing protocols, sperm samples were collected from domestic cats (n=7) after castration and divided into 2 aliquots as for Protocol 1 (P1) or Protocol 2 (P2). Semen cryopreservation alters sperm structure and biochemical properties, and subsequently impairs fertilizing ability. A combination of two different sugar molecules revealed an enhanced beneficial effect on frozen-thawed spermatozoa of various species. However, the report in ram has been scarce. The present study investigated the effect of different combinations of sugar supplementation on post-thawed ram semen qualities in a cryopreservative extender in the presence of a single sugar or a combination of sugars. Data showed that a combination of sucrose and trehalose significantly improved cryopreserved sperm motility, viability, longevity and acrosome integrity. The present study aims to investigate a suitable L-cysteine level to be added the lactose-egg yolk (LEY) extender to improve the quality of boar semen after freezing-thawing using Annexin-V and propidium iodide (PI) assay detected with flow cytometer. Ejaculates from twelve boars were processed in the LEY extender and added with 0, 5, 10 and 15 mM of L-cysteine. Computer-assisted sperm analysis (CASA) was initiated to reduce subjective bias on the motility assessment and to discriminate a series of motility patterns of boar semen. The objectives of this present study were to evaluate the motility pattern of frozen-thawed boar semen at 0 and 60 min after thawing using CASA. The study was aimed at finding out whether dog sperm could be labelled using 99mTc-HMPAO and finding the optimum incubation temperature and amount of 99mTc-HMPAO to label spermatozoa from fresh ejaculates and from frozen-thawed semen. A radiopharmaceutical was prepared by reconstituting 0.5 mg of HMPAO with 5 ml pertechnetate solution containing 100 kBq 99mTc. Semen samples were obtained from three dogs, pooled and divided into aliquots containing 100x106 spermatozoa each. Fresh and frozen-thawed semen samples were used. During sperm transport in the female pig, a proportion of the spermatozoa is arrested at the uterotubal junction and the isthmus, where an oviductal sperm reservoir is built up. The sperm reservoir maintains sperm viability and controls the release of fertilizing spermatozoa so that only a small subpopulation reaches the site of fertilization, thus diminishing the risk of polyspermy. In the pig, sperm transport is regulated, to facilitate normal fertilization of the ovulated oocytes, by the oviduct which contains a particular segment, the sperm reservoir. The main function of the sperm reservoir is to ensure that stored spermatozoa have maintained viability and fertilizing capacity. The process of sperm capacitation is delayed in the sperm reservoir, during the lengthy pre-ovulatory estrus by interactions between the spermatozoa, the oviduct epithelium and secretion products.
