Methicillin-susceptible Staphylococcus aureus (MSSA) can arise from methicillin-resistant S. aureus (MRSA) following partial or complete excision of staphylococcal cassette chromosome mec (SCCmec). This study investigated whether multiresistant MSSA isolates from Irish hospitals, where MRSA has been endemic for decades, harbor SCCmec DNA. Twenty-five multiresistant MSSA isolates recovered between 2002 and 2006 were tested for SCCmec DNA by PCR and were genotyped by multilocus sequence typing and spa typing. All isolates lacked mecA. Three isolates (12%) harbored SCCmec DNA; two of these (genotype ST8 t190) harbored a 26-kb SCCmec IID (II.3.1.2) remnant that lacked part of mecI and all of mecR1, …

Momitochondrial Dysfunction And Dna Damage Post Exposure To Low Dose Radiation And "Bystander" Factor (S), Sharon Nugent Dec 2008

Momitochondrial Dysfunction And Dna Damage Post Exposure To Low Dose Radiation And "Bystander" Factor (S), Sharon Nugent

Doctoral

The 'bystander effect' describes radiation-like damage in un irradiated cells, either in the vicinity of irradiated cells or exposed to medium from irradiated cells. This study aimed to further characterize the, as yet poorly understood, mitochondrial response to both direct irradiation and bystander factor(s). Human keratinocyte epithelial and Chinese hamster ovarian cells were exposed to either y radiation or bystander factor(s) at doses of 5mOy, O.50y and 50y and examined 4-96 hours later. DNA damage was determined using deletion analysis, single strand conformation polymorphism analysis, long range PCR, semi-quantitative PCR and quantitative real-time PCR. Mitochondrial mass was determined using MitoTrackelM …

Articles

Epidemiological and clinical studies provide compelling support for a causal relationship between Helicobacter pylori infection and endothelial dysfunction, leading to vascular diseases. However, clear biochemical evidence for this association is limited. In the present study, we have conducted a comprehensive investigation of endothelial injury in bovine aortic endothelial cells (BAECs) induced by H. pylori-conditioned medium (HPCM) prepared from H. pylori 60190 [vacuolating cytotoxin A (Vac(+))]. BAECs were treated with either unconditioned media, HPCM (0-25% vol/vol), or Escherichia coli-conditioned media for 24 h, and cell functions were monitored. Vac(+) HPCM significantly decreased BAEC proliferation, tube formation, and migration (by up to …

Nanotechnology is regarded as one of the key technologies of the future and associated with high expectations by politics, science and economy. Artificially produced nanosized particles and nanoscale system components have new properties which are of importance for the development of new products and applications. Such new properties of materials and substances result from the special properties of surfaces and interfaces and in part, from the geometric shape of the material. In theory nanoparticles (NPs) can be produced from nearly any chemical; however, most NPs that are currently in use today have been made from transition metals, silicon, carbon (single-walled …

Conference Papers

The role of cyclodextrin’s (CD) in drug delivery has advanced in recent years and this may be attributed to its biocompatibility and well established synthesis. Chemical modification of CDs has shown to extend the physicochemical properties and the host capacity for a variety of drugs. β-CD has been widely used in the early stages of pharmaceutical applications because of its ready availability and its cavity size suitability for a wide range of drugs. Chemical modification of β-CD has proven to enhance aqueous solubilisation, microbiological stability and reduced toxicity in previous studies.1 Folate Receptors are over-expressed in several human cancers including …

Synthesis Of Novel Targeted Drug Delivery Systems For Cancer Therapy, Antonio Clementi

Masters

One of the most important goals of pharmaceutical science is local izing the pharmacological activity of the drug at the site of action. Drug delivery systems - are. molecular tools which without undesired interaction at other sites target a specific drug receptor. In this work a· novel folate-cyclodextrin conjugate was synthesised for potential use as a drug delivery system in cancer therapy. Folate was directly covalently bound to an amino derivative of p-Cyclodextrin with a view to decreasing the polydispersity of the carrier system. Ultimately the cyclodextrin cavity will be used to transport drugs to the site of action. This …

Ocular Aberrations Wavefront Sensor In Clinical Application, Matthew Sheehan

Masters

A versatile Hartmann-Shack (HS) wavefront sensor (WFS) was designed and constructed. Its advantages include simultaneous pupil centre determination, off-axis capability, realtime data displays, and efficient lenslet sampling orientation. The instrument is compact and transportable, making it convenient for use in a clinical setting. Subject alignment is achieved by the use of a parallel channel that is recombined with the sensing channel to simultaneously image the external eye and the HS spots onto a single charge coupled device (CCD). The pupil centre is determined using this image of the eye, rather than the HS spots. Off-axis measurements are possible throughout an …

Treg Depletion Inhibits Efficacy Of Cancer Immunotherapy: Implications For Clinical Trials., James Curtin, Marianela Candolfi, Tamer Fakhouri, Chunyan Liu, Anderson Alden, Matthew Edwards, Pedro Lowenstein, Maria Castro

Articles

BACKGROUND: Regulatory T lymphocytes (Treg) infiltrate human glioblastoma (GBM); are involved in tumor progression and correlate with tumor grade. Transient elimination of Tregs using CD25 depleting antibodies (PC61) has been found to mediate GBM regression in preclinical models of brain tumors. Clinical trials that combine Treg depletion with tumor vaccination are underway to determine whether transient Treg depletion can enhance anti-tumor immune responses and improve long term survival in cancer patients. FINDINGS: Using a syngeneic intracrabial glioblastoma (GBM) mouse model we show that systemic depletion of Tregs 15 days after tumor implantation using PC61 resulted in a decrease in Tregs …

An Investigation Of The Diets Of Infants Born In Ireland During The First Six Months Of Life, Roslyn Tarrant Apr 2008

An Investigation Of The Diets Of Infants Born In Ireland During The First Six Months Of Life, Roslyn Tarrant

Doctoral

Appropriate infant feeding practices play a crucial part in achieving optimal health outcomes. It is well established that the protection, promotion and support of exclusive breastfeeding during the first 6-months of life would decrease the health inequalities experienced by mothers and infants (WHO/UNICEF, 2003c). Optimal weaning practices also have significant implications for infant health, notably in relation to normal development, mineral balance and the development of obesity (Department of Health, 1994). Historically, Ireland has one of the lowest breastfeeding rates in Europe. Furthermore, prior Irish-based research indicates significant deficiencies relating to weaning practices among mothers during the first year of …

Resource Pack On Curriculum Design And Assessment To Promote Effective Learning., Christine O'Connor

Other resources

This resource pack is an overview of current considerations for academics designing programmes for third level education. The changing demographic of third level students along with employers’ demands has resulted in programme development with a focus on skills basis (Hyslop-Margison, 2001) to support a knowledge based society. The rationale behind the changes in curriculum design is introduced and further focus is emphasised in the areas of curriculum design models, assessment models and evaluation models. Examples of innovative curricula and assessment models in third level chemistry education will be incorporated during the pack which may be applied across other disciplines.

During the course of a day human skin is exposed to solar UV radiation that fluctuates in fluence rate within the UVA (290-315 nm) and UVB (315-400 nm) spectrum. Variables affecting the fluence rate reaching skin cells include differences in UVA and UVB penetrating ability, presence or absence of sunscreens, atmospheric conditions, and season and geographical location where the exposure occurs. Our study determined the effect of UVA fluence rate in solar-simulated (SSR) and tanning-bed radiation (TBR) on four indicators of oxidative stress---protein oxidation, glutathione, heme oxygenase-1, and reactive oxygen species--in human dermal fibroblasts after receiving equivalent UVA and UVB …

Articles

Weather-related health effects have attracted renewed interest because of the observed and predicted climate change. The authors studied the short-term effects of cold weather on mortality in 15 European cities. The effects of minimum apparent temperature on cause- and age-specific daily mortality were assessed for the cold season (October-March) by using data from 1990-2000. For city-specific analysis, the authors used Poisson regression and distributed lag models, controlling for potential confounders. Meta-regression models summarized the results and explored heterogeneity. A 1 degrees C decrease in temperature was associated with a 1.35% (95% confidence interval (CI): 1.16, 1.53) increase in the daily …

Ovarian Signalling:Insights Into The P13k/Pkb/Foxo And The Ras/Raf/Mek/Erk Pathways, Nicola Cahill Jan 2008

Ovarian Signalling:Insights Into The P13k/Pkb/Foxo And The Ras/Raf/Mek/Erk Pathways, Nicola Cahill

Masters

The ovaries are a pair of female reproductive organs that are involved in the production of gametes “oogenesis” and the production of hormones “steroidogenesis” to mediate the oestrous cycle, preparation of the uterus for pregnancy and the preparation of the breast for lactation www.lab.anhb.uwa.edu.au/mb140/corepages/femaleRepro). The oestrous cycle is controlled by the release of the pituitary gonadotrophins, follicle stimulating hormone (FSH) and lutenising hormone (LH). The physiological responses of ovarian steroidogenesis, oogenesis, folliculargenesis, ovulation and lutenisation to the gonadotrophins are accomplished by the activation of greater than one hundred ovarian target genes (Hunzicker-Dunn et al 2006). These processes are dependant on …

Making Sense Of Men's Workout Practices: The Body, Age And Identity, Andrew Dunne Jan 2008

Making Sense Of Men's Workout Practices: The Body, Age And Identity, Andrew Dunne

Masters

Background: Within sociology it is generally accepted that the body has become an object or “project” that is worked on and transformed as a central part of self-identity (Baudrillard 1998; Corrigan 1997; Featherstone 1991; Giddens 1991; Turner 1995, 1992; Shilling 2003). An alternative to such arguments, Leder (1990) conceptualizes the body as an “absent presence”. He argues that, while the body plays a central role in shaping our experience of the world, we are frequently oblivious to our own bodies. For Leder, bodywork is sporadic. He contends that specific social and/or physiological experiences cause the body to “dys-appear”, or enter …

Maternal Awareness Of The Association Between Paediatric Diet And Adult Disease Risk: A Follow-Up To The Dit-Coombe Hospital Study., Qianling Zhou, Katherine Younger, John Kearney

Conference Papers

Chronic disease has been traditionally attributed to risk factors in adulthood; however, current orthodoxy suggests that the true origins of such diseases are initiated much earlier in life from inappropriate nutritional circumstances arising during the formative years(1). The present study was undertaken to examine current maternal attitudes towards the importance of paediatric diet in relation to adult diseases and to determine the extent to which attitudinal differences between subgroups in the population exist. A non-randomised postal follow-up survey to the DIT–Coombe Hospital Infant Feeding Study was carried out, involving the recruitment of 209 mothers from a pool sample of 520 …

Towards Measuring Continuous Acoustic Feature Convergence In Unconstrained Spoken Dialogues, Spyros Kousidis, David Dorran, Yi Wang, Brian Vaughan, Charlie Cullen, Dermot Campbell, Ciaran Mcdonnell, Eugene Coyle

Conference papers

Acoustic/prosodic feature (a/p) convergence has been known to occur both in dialogues between humans, as well as in human-computer interactions. Understanding the form and function of convergence is desirable for developing next generation conversational agents, as this will help increase speech recognition performance and naturalness of synthesized speech. Currently, the underlying mechanisms by which continuous and bi-directional convergence occurs are not well understood. In this study, a direct comparison between time-aligned frames shows significant similarity in acoustic feature variation between the two speakers. The method described (TAMA) constitutes a first step towards a quantitative analysis of a/p convergence.

Session 1: Public Health Nutrition Breast-Feeding Practices In Ireland, Roslyn Tarrant, John Kearney Jan 2008

Session 1: Public Health Nutrition Breast-Feeding Practices In Ireland, Roslyn Tarrant, John Kearney

Articles

Breast-feeding is the superior infant feeding method from birth, with research consistently demonstrating its numerous short- and long-term health benefits for both mother and infant. As a global recommendation the WHO advises that mothers should exclusively breast-feed for the first 6-months of life, thus delaying the introduction of solids during this time. Historically, Irish breast-feeding initiation rates have remained strikingly low in comparison with international data and there has been little improvement in breast-feeding duration rates. There is wide geographical variation in terms of breast-feeding initiation both internationally and in Ireland. Some of these differences in breast-feeding rates may be …

Novel Functions Of Acyl-Coa Thioesterases And Acyltransferases As Auxiliary Enzymes In Peroxisomal Lipid Metabolism., Mary Hunt, Stefan Alexson

Articles

Peroxisomes are single membrane bound organelles present in almost all eukaryotic cells, and to date have been shown to contain approximately 60 identified enzymes involved in various metabolic pathways, including the oxidation of a variety of lipids. These lipids include very long-chain fatty acids, methyl branched fatty acids, prostaglandins, bile acid precursors, and xenobiotics that are either β-oxidized or α-oxidized in peroxisomes. The recent identification of several acyl-CoA thioesterases and acyltransferases in peroxisomes has revealed their various functions in acting as auxiliary enzymes in α- and β-oxidation in this organelle. To date, 9 functional acyl-CoA thioesterases and acyltransferases have been …

The Use Of Cellomics To Study Enterocyte Cytoskeletal Proteins In Coeliac Disease Patients, Bashir M. Mohamed, Conleth Feighery, Yvonne Williams, Anthony Davies, Dermot Kelleher, Yuri Volkov, Jacinta Kelly, Mohamed Abuzakouk

Articles

Coeliac disease is characterised by inflammation of small intestinal mucosa accompanied by abnormal villous architecture. It is now accepted that some patients with positive coeliac serology tests may have minor mucosal lesions that may not be apparent on routine histopathological analysis. The aim of the study was to perform detailed examination of enterocyte morphology and cytoskeletal structures using a high content analysis technology. Duodenal biopsies from 14 untreated and 10 treated coeliac patients and from 20 non-coeliac controls were examined. Tissue sections from six patients (study group subjects) before and after the development of gluten-sensitive enteropathy were also investigated. Immunohistochemical …

Object Detection And Classification With Applications To Skin Cancer Screening, Jonathan Blackledge, Dmitryi Dubovitskiy Jan 2008

Object Detection And Classification With Applications To Skin Cancer Screening, Jonathan Blackledge, Dmitryi Dubovitskiy

Articles

This paper discusses a new approach to the processes of object detection, recognition and classification in a digital image. The classification method is based on the application of a set of features which include fractal parameters such as the Lacunarity and Fractal Dimension. Thus, the approach used, incorporates the characterisation of an object in terms of its texture.

The principal issues associated with object recognition are presented which includes two novel fast segmentation algorithms for which C++ code is provided. The self-learning procedure for designing a decision making engine using fuzzy logic and membership function theory is also presented and …

We are all familiar with the dietary guideline exhorting us to eat so many portions of oily fish per week in order to boost our intakes of n-3 long-chain PUFA, but it is perhaps not so widely realised that the fish must themselves be provided with dietary n-3 long-chain PUFA or, possibly, their precursors (though, as in humans, the ability of carnivorous fish to elongate and desaturate n-3 PUFA appears to be limited).

Patient-Centred Laboratory Validation Using Software Agents, John Mcgrory, Jane Grimson, Frank Clarke, Peter Gaffney Jan 2008

Patient-Centred Laboratory Validation Using Software Agents, John Mcgrory, Jane Grimson, Frank Clarke, Peter Gaffney

Conference Papers

Guidelines are self-contained documents which healthcare professionals reference to obtain knowledge about a specific condition or process. They interface with these documents and apply known facts about specific patients to gain useful supportive information to aid in developing a diagnosis or manage a condition. To automate this process a series of Standard Operating Procedures (SOP) and workflow processes are constructed using the contents of these documents in order to manage the validation flow of a patient sample. These processes decompose the guidelines into workflow plans, which are then called using condition triggers controlled by a centralised management engine. The software …
