In vitro embryo production (IVP) can help alleviating reproductive difficulties in cattle and buffalo. This study compared qualitative and quantitative features of native cattle and buffalo ovaries and oocytes to understand them comprehensively. In addition, it was conducted to pick the ideal ovaries for oocyte collection and to assess either maturation affects oocyte diameter or not. The ovaries were collected from the local slaughterhouses of Dhaka city, Bangladesh. Next, the quantitative parameters of ovaries were measured, the follicular fluid was aspired, the oocytes were classified, and the diameters of oocytes were determined in both pre and postmaturation. Recent progress in genomic analysis and other biochemical methods has led to the discovery of a large population of microRNAs (miRNAs), which have been demonstrated to play important roles in diseases and a wide range of developmental processes. Characterization of miRNA expression profiles in different stages of ovarian follicular development and early embryogenesis has suggested the potential roles of miRNAs in follicular development, maturation of oocytes, and preimplantation embryonic development. This review focuses on the current studies of miRNAs involved in ovarian and early embryonic development in cattle.

This review focuses on recent research related to the avian ovary and, in particular, cellular mechanisms mediating embryonic growth of the ovary, postembryonic follicular development, and the process of follicular selection. Attention is drawn to a variety of fundamental, yet unresolved, aspects of ovarian function, several of which are uniquely related to avian reproductive strategies. Some notable differences in ovarian function between birds and the most closely related extant group, the crocodilians, are illustrated. Recovery rate and quality of oocytes were investigated from abattoir collected ovaries with and without corpora lutea (CL). The oocytes were categorized into four grades: grade-A, grade-B, grade-C, and grade-D based on the presence of the cumulus cells complex around the oocytes. The number of oocytes per ovary were 1.7 ±0.1 and 1.5 ±0.1, obtained from ovaries without CL and with CL, respectively. Recovery rate of different grades of oocytes from ovaries with CL and without CL were almost identical.

Stages of oocyte maturation in sea bass (Dicentrarchus labrax) gonads after LHRHa hormonal treatment were investigated. LHRHa was injected into sea bass broodstock with an oocyte diameter of 527-580 µm. A second injection was given 12 h after the first injection. For each injection a dose of 10 µg/kg was used. The objective of the present study was to investigate the ultrastructural changes seen in frozen porcine oocytes at the germinal vesicle and during in vitro maturation when using an ultrarapid freezing technique. The morphological changes were assessed by transmission electron microscopy (TEM). Immature oocytes were collected from pig ovaries at a local slaughter house in Nakorn Prathom province. The Use Of Fresh, Salt-Stored And Frozen, Immature And Mature Oocytes, To Assess The Penetration Ability Of Porcine Spermatozoa, Adisorn Adirekthaworn, Mongkol Techakumphu, Sudson Sirivaidyapong, Saroch Gnamkum

The objective of this study was to compare the penetration rate of sperm, collected from two Landrace Boars A and B, into fresh, salt-stored and frozen, immature and matured oocytes. The first experiment, performed on immature oocytes, revealed that the penetration rates for the three groups of oocytes was from Boar A 59.6%(59/99), 78.1%(75/96) and 77.8%(77/99) (p<0.05) and the number of sperm penetration each oocyte was 2.79±0.42, 2.97±0.29 and 2.29±0.26 respectively (p>0.05). While for Boar B, the percentages were 65.3%(62/95), 76.8%(73/95) and 77.8%(77/99) with the number of sperm per oocyte being 2.25±0.28, 3.63±0.42 and 2.57±0.36. The highest values were found in the salt-stored group (p<0.05). The second experiment involved sperm penetration into in vitro mature oocytes. No differences were found for the sperm penetration rate and the number of sperm per oocyte using semen from Boar A which were 85.1%(74/87), 86.1%(56/65) and 89.1%(79/88) and 13.87±1.45, 17.69±2.61 and 14.45±1.75. While for Boar B, the lowest penetration rate was found when using fresh oocyte, lower than the two other groups; 52.6%(50/95), 67.3%(64/95) and 69.1%(67/ 97)(p<0.05). The number of sperm entering each oocyte was also inferior in the fresh oocytes than in salt-stored and frozen ones; 1.55±0.31, 2.80±0.35 and 2.87±0.40 (p<0.05) respectively. The conclusion that can be drawn from the experiment is that a) there are significant differences in the sperm penetration ability of different the boars used and b) the type of oocytes; immature vs. mature and fresh vs. salt-stored vs. frozen influenced the results that were obtained. It maybe possible to apply such an in vitro penetration assay to determine boar semen fertility under field conditions.

Embryo And Oocyte Collection In Thai Swamp Buffalo (Bubalus Bubalis) After Superovulation, Mongkol Techakumphu, Yant Sukwong, Malee Apimeteetumrong, Siriwat Intaramongkol, Jintana Intaramongkol

Six-day embryos were recovered from 25 swamp buffalo cows treated with pregnant mare serum gonadotropin (2,500 iu PMSG, n=9) and follicle stimulating hormone (280 mg, n=12 and 400 mg FSH, n=4). Two techniques, non surgical collection from live animals and direct flushing after slaughter were compared. The oocytes were collected from ovaries immediately after slaughter by aspiration with or without ovarian chopping. The studies showed that 0.44+0.59 embryos were collected from PMSG group, while 1.38+1.31 embryos were collected in FSH group. Fifty percent to 60% of recovered embryos were transferable. The objective of this study was to determine the optimum condition for porcine oocyte preparation for in vitro fertilization. A total of 1,130 immature oocytes collected from superficial follicles, 3-6 mm in diameter were allocated to 3 experiments. In Experiment 1, the length of time for oocyte maturation were examined (n=469). Immature oocytes were cultivated in TCM199 NaHCO3 supplemented with 10 μg/ml of FSH/LH and 1 μg/ml of Estradiol-17ẞ for 24, 40 or 48 hrs.
