Melanopsin, expressed in a subset of retinal ganglion cells, mediates behavioral adaptation to ambient light and other non-image forming photic responses. This has raised the possibility that pharmacological manipulation of melanopsin can modulate several CNS responses including photophobia, sleep, circadian rhythms and neuroendocrine function. Here we describe the identification of a potent synthetic melanopsin antagonist with in vivo activity. Novel sulfonamide compounds inhibiting melanopsin (opsinamides) compete with retinal binding to melanopsin and inhibit its function without affecting rod/cone mediated responses. We report here identification of novel mimicry epitopes for interphotoreceptor retinoid-binding protein (IRBP) 201–216, a candidate ocular antigen that causes experimental autoimmune uveoretinitis (EAU) in A/J mice. One mimicry epitope from Ehrlichia canis (EHC), designated EHC 44–59, induced cross-reactive T cells for IRBP 201–216 capable of producing T helper (Th)1 and Th17 cytokines, but failed to induce EAU in A/J mice. In addition, animals first primed with suboptimal doses of IRBP 201–216 and subsequently immunized with EHC 44–59 did not develop EAU; rather, the mimicry epitope prevented the disease induced by IRBP 201–216. In sugar cataract formation in rats, aldose reductase (AR) actitvity is not only linked to lenticular sorbitol (diabetic) or galactitol (galactosemic) formation but also to signal transduction changes, cytotoxic signals and activation of apoptosis. Using both in vitro and in vivo techniques, the interrelationship between AR activity, polyol (sorbitol and galactitol) formation, osmotic stress, growth factor induction, and cell signaling changes have been investigated. The heterogeneous nuclear ribonucleoprotein K (hnRNP K) is a member of the family of hnRNPs and was recently shown in a genome-wide small interfering RNA (siRNA) screen to support vesicular stomatitis virus (VSV) growth. To decipher the role of hnRNP K in VSV infection, we conducted studies which suggest that the protein is required for VSV spreading. Virus binding to cells, entry, and nucleocapsid uncoating steps were not adversely affected in the absence of hnRNP K, whereas viral genome transcription and replication were reduced slightly. A critical question surrounding emergence of novel strains of avian influenza viruses (AIV) is the ability for wild migratory birds to translocate a complete (unreassorted whole genome) AIV intercontinentally. Virus translocation via migratory birds is suspected in outbreaks of highly pathogenic strain A(H5N1) in Asia, Africa, and Europe. As a result, the potential intercontinental translocation of newly emerging AIV (e.g. A(H7N9) from Eurasia to North America via migratory movements of birds) remains a concern. Obesity is an epidemic in many developed nations and maternal obesity can result in developmental alterations in offspring that have long-lasting effects. Two experiments were conducted to determine the impact of maternal obesity on placental development and early embryonic growth and muscle development. Experiment one utilized obese Lethal Yellow (LY) and normal weight C57BL/6 (B6) dams to assess how maternal obesity alters skeletal muscle development in mid-gestational embryos. Embryos from LY dams exhibited decreased embryo and placental weights as well as an increase in the myogenic marker desmin. Microtubule transport of herpesvirus capsids from the cell periphery to the nucleus is imperative for viral replication and, in the case of many alphaherpesviruses, transmission into the nervous system. Using the neuroinvasive herpesvirus, pseudorabies virus (PRV), we show that the viral protein 1/2 (VP1/2) tegument protein associates with the dynein/dynactin microtubule motor complex and promotes retrograde microtubule transport of PRV capsids. Functional activation of VP1/2 requires binding to the capsid protein pUL25 or removal of the capsid-binding domain. A 1.5-year-old second-parity Large Black X Tamworth cross sow from a well-managed 20-sow, unvaccinated, pasture-raised herd of pigs in upstate New York aborted a litter of 7 variably mummified near-term fetuses and stillborn piglets. This sow had no signs of ill health other than abortion; it was housed in a group with 4 other sows and a 2-year-old Gloucestershire Old Spot boar. One of the stillborn piglets from this litter was submitted to the New York State Animal Health Diagnostic Center for necropsy; no placental tissue was submitted with the piglet. Background: Bovine viral diarrhea virus is one of the most significant and costly viral pathogens of cattle worldwide. Alphavirus-derived replicon particles have been shown to be safe and highly effective vaccine vectors against a variety of human and veterinary pathogens. Replicon particles are non-propagating, DIVA compatible, and can induce both humoral and cell mediated immune responses. This is the first experiment to demonstrate that Alphavirus-based replicon particles can be utilized in a standard prime/boost vaccination strategy in calves against a commercially significant bovine pathogen.

A recent controversial hypothesis suggested that the bactericidal action of antibiotics is due to the generation of endogenous reactive oxygen species (ROS), a process requiring the citric acid cycle (tricarboxylic acid [TCA] cycle). To test this hypothesis, we assessed the ability of oxacillin to induce ROS production and cell death in Staphylococcus epidermidis strain 1457 and an isogenic citric acid cycle mutant. Our results confirm a contributory role for TCA-dependent ROS in enhancing susceptibility of S. epidermidis toward β-lactam antibiotics and also revealed a propensity for clinical isolates to accumulate TCA cycle dysfunctions presumably as a way to tolerate these …

An 8-month-old crossbred ewe, normal upon physical examination, was humanely euthanized for tissue collection. After approximately 3 weeks in tissue culture, fungi began budding out of cells obtained from the choroid plexus. After an additional 3 weeks, budding was observed in kidney cell cultures and eventually in monocyte cultures as well. Serum from the lamb was submitted to the Veterinary Diagnostic Laboratory at Colorado State University for fungal diagnosis and was found negative for Aspergillus, Blastomyces, Coccidioidomycosis and Histoplasmosis. Infectious myonecrosis virus (IMNV) is a significant and emerging pathogen that has a tremendous impact on the culture of the Pacific white shrimp Litopenaeus vannamei. IMNV first emerged in Brazil in 2002 and subsequently spread to Indonesia, causing large economic losses in both countries. No existing therapeutic treatments or effective interventions currently exist for IMNV. RNA interference (RNAi) is an effective technique for preventing viral disease in shrimp. Innate immune sensing of viral nucleic acids triggers type I interferon (IFN) production, which activates interferon-stimulated genes (ISGs) and directs a multifaceted antiviral response. ISGs can also be activated through IFN-independent pathways, although the precise mechanisms remain elusive. Here we found that the cytosolic exonuclease Trex1 regulates the activation of a subset of ISGs independently of IFN. Both Trex1−/− mouse and TREX1-mutant human cells express high levels of antiviral genes and are refractory to viral infections. The IFN-independent activation of antiviral genes in Trex1−/− cells requires STING, TBK1 and IRF3 and IRF7. We also found that Trex1-deficient cells display expanded …

Glutathionylation has emerged as a key modification required for controlling protein function in response to changes in cell redox status. Recently, we showed that the glutathionylation state of uncoupling protein-3 (UCP3) modulates the leak of protons back into the mitochondrial matrix, thus controlling reactive oxygen species production. However, whether or not UCP3 glutathionylation is mediated enzymatically has remained unknown because previous work relied on the use of pharmacological agents, such as diamide, to alter the UCP3 glutathionylation state. In the United States, the blaCMY-2 gene contained within incompatibility type A/C (IncA/C) plasmids is frequently identified in extended-spectrum-cephalosporin-resistant (ESCr) Escherichia coli strains from both human and cattle sources. Concerns have been raised that therapeutic use of ceftiofur in cattle may increase the prevalence of ESCr E. coli. Background: MicroRNAs (miRNAs) are small non-coding RNAs that play critical roles in regulating post transcriptional gene expression. Gall midges encompass a large group of insects that are of economic importance and also possess fascinating biological traits. The gall midge Mayetiola destructor, commonly known as the Hessian fly, is a destructive pest of wheat and model organism for studying gall midge biology and insect – host plant interactions.

α-Herpesvirinae subfamily members, including herpes simplex virus type 1 (HSV-1) and bovine herpes virus 1 (BHV-1), initiate infection in mucosal surfaces. BHV-1 and HSV-1 enter sensory neurons by cell-cell spread where a burst of viral gene expression occurs. When compared to non-neuronal cells, viral gene expression is quickly extinguished in sensory neurons resulting in neuronal survival and latency. The HSV-1 latency associated transcript (LAT), which is abundantly expressed in latently infected neurons, inhibits apoptosis, viral transcription, and productive infection, and directly or indirectly enhances reactivation from latency in small animal models. This study evaluated the effect of post-weaning development system on heifer ADG, reproductive performance, and subsequent feed efficiency as a pregnant heifer. Heifers were developed on dormant pasture and grazed corn residue or dormant pasture and placed in a drylot. The following winter, a subset of pregnant heifers were placed in a Calan Broadbent individual feeding system during late gestation. Drylot-developed heifers had greater BW from pre-breeding through pregnancy diagnosis and greater overall ADG during development. However, there was no difference in reproductive performance. Pre-calving BW, ADG, and G:F tended to be greater for drylot heifers. Heifers developed on corn …

Nebraska Beef Cattle Reports

Two identically designed trials were conducted in separate years at the University of Nebraska Haskell Agricultural Laboratory, Concord, Neb., using 192 crossbred heifers (96/trial). Within a trial, heifers were assigned randomly to 2 groups (3 pens/group): 1) treatment (TRT) animals were administered synthetic hormones via subcutaneous implants (Ralgro and Revalor-H) and fed Melengesterol Acetate (MGA), or 2) control (CON) animals with no synthetic hormone provided.

The effect of heifer development system on primiparous heifer performance grazing corn residue during late gestation was investigated. Weaned heifers grazing corn residue tended to have reduced final BW after corn residue grazing compared to heifers grazing winter range. However reproductive performance for the two treatments was similar. When grazing corn residue as pregnant heifers during late gestation, heifers developed on corn residue had improved ADG compared to drylot-developed heifers and tended to have increased ADG compared to winter range-developed heifers. Adaptation to corn residue grazing as a developing heifer improves primiparous heifer performance grazing corn residue during late gestation.

Two experiments evaluated the feeding value of grass hay bales previously treated with CCDS in growing cattle diets. In Experiment 1, heifers fed bales treated with 20% CCDS (DM) gained less than those fed an equal level of dried distillers grains plus solubles and nontreated hay. In Experiment 2, ADG and F:G linearly improved with increasing CCDS levels. Furthermore, supplementing cattle to meet metabolizable protein requirements when fed diets of CCDS and hay did not improve ADG at levels greater than 15% CCDS. A grazing study was conducted to determineif field peas are a good naturalbinder for dried distillers grains-based range cubes. Cattle supplemented dried distillers grains in the bunk or a 25% field pea/75% distillers grains cube fed on the ground gained similarly and outgained cattle supplemented dried distillers on the ground. A seven-year study from 2005-2011 was conducted to evaluate four grazing management strategies for backgrounding yearling steers on smooth bromegrass pastures. Economic budgets were used to calculate profit differences with current (April 2012) market prices. Overall, cattle receiving supplement had greater net returns, lower cost of gain, and lower breakeven prices. In recent years fertilizer prices have increased at a greater rate than land costs in Nebraska, making it more economical to use a lower stocking rate instead of fertilizing pastures. As land prices increase, the incentive to use either N fertilizer or DDGS supplementation increases.

Corn residue is used for forage and feed, but residue removal effects on soil properties and yield is a concern. Residue removal effects on corn yields and soil organic carbon is site specific. Removing 50% of the residue from rain fed sites reduced corn yield by 1.9 bu/ac, whereas removing 40% of the residue from irrigated sites increased corn yield by 15.4 bu/ac. However, removing 53% of the residue increased soil erosion by 30%. Agronomic practices such as reduced tillage, cover crops, or manure may offset residue removal impacts. Residue removal should be based on site-specific characteristics and management, but …

A grasshopper control study was conducted to evaluate the effectiveness of a new class of systemic chemical. The new pesticide Prevathon® (high and low levels) was compared to Coragen®, Dimilin®, and a nontreated check. Grasshoppers were numerically reduced the most by Coragen and Prevathon, though not significant. The highest level of Prevathon did not numerically impact beneficial insects in general. Biomass and forage quality were not significantly impacted by chemical treatment. However, forage biomass was numericallygreatest for the highest level of Prevathon. The interaction between corn processing method and condensed distillers solubles (CDS) was evaluated using either steam-flaked corn (SFC) or dry rolled corn (DRC). As CDS replaced corn at either 15 or 30% of the diet DM, DMI intake decreased quadratically for both SFC and DRC. Within DRC- based diets, ADG increased quadratically with 15% CDS being greatest and F:G improved quadratically with 30% inclusion being best. When SFC was used as the grain source, ADG increased linearly and F:G improved quadratically with increasing levels of CDS. Cattle were adapted to a finishing diet over 20 days while feeding 33 or 44 g/ton (DM) of Rumensin. Following grain adaptation, all cattle were fed a common finishing diet containing 33 g/ton Rumensin for the remainder of the feeding period. Feeding 44 g/ton of Rumensin during the adaptation period did not affect performance during the adaptation period or over the entire feeding period when compared to the 33 g/ton adaptation level.

Finishing cattle performance was evaluated using NEXT ENHANCE essential oils in finishing diets. Treatments consisted of 1) control (CON), 2) NEXT ENHANCE at 300 mg/day (NE), 3) Rumensin and Tylan at 360 and 90 mg/day, respectively (RT), or 4) NEXT ENHANCE plus Rumensin and Tylan (NERT). No NEXT ENHANCE by Rumensin/Tylan interaction was observed. Steers fed Rumensin/Tylan had decreased F:G and increased live final BW and marbling score. The prevalence of liver abscesses decreased 46% when steers were fed Rumensin/Tylan. Including NEXT ENHANCE in finishing diets did not impact performance or carcass characteristics.
