Since the launch of the first orally available angiotensin II (AngII) type 1 receptor (AT1R) blocker (ARB) losartan (Cozaar) in the late 1990s, the class of ARBs (or ‘sartans’, short for Angiotensin-RecepTor-ANtagonistS) quickly expanded to include candesartan, eprosartan, irbesartan, valsartan, telmisartan, and olmesartan. All ARBs have high affinity for the AT1 receptor, expressed in various tissues, including smooth muscle cells, heart, kidney, and brain. Since activation of AT1R, the target of these drugs, leads, among other effects, to vascular smooth muscle cell growth, proliferation and contraction, activation of fibroblasts, cardiac hypertrophy, aldosterone secretion from the …

Post-traumatic stress disorder (PTSD) is more prevalent in women than men, yet much remains to be determined regarding the mechanism underlying this sex difference. Clinical and preclinical studies have shown that low estradiol levels during extinction of fear conditioning in rodents (i.e., cue exposure therapy in humans) leads to poor extinction consolidation and increased fear during extinction recall. The renin-angiotensin system (RAS) is also associated with stress-related pathologies, and RAS antagonists can enhance extinction consolidation in males. However, less is known about how estradiol and the RAS converge to alter fear extinction consolidation in females. Since estradiol downregulates the RAS, …

Developing A Biased Unmatched Bivalent Ligand (Bumbl) Design Strategy To Target The Gpcr Homodimer Allosteric Signaling (Camp Over Β-Arrestin 2 Recruitment) Within The Melanocortin Receptors., Cody J Lensing, Katie T Freeman, Sathya M Schnell, Robert Charles Speth, Adam T Zarth, Carrie Haskell-Luevano Jan 2019

Understanding the functional relevance of G protein-coupled receptor (GPCR) homodimerization has been limited by the insufficient tools to assess asymmetric signaling occurring within dimers comprised of the same receptor type. We present unmatched bivalent ligands (UmBLs) to study the asymmetric function of melanocortin homodimers. UmBLs contain one agonist and one antagonist pharmacophore designed to target a melanocortin homodimer such that one receptor is occupied by an agonist and the other receptor by an antagonist pharmacophore. First-in-class biased UmBLs (BUmBLs) targeting the human melanocortin-4 receptor (hMC4R) were discovered. The BUmBLs displayed biased agonism by potently stimulating cAMP signaling (EC

Protective Actions Of Oxytocin On Aortic Wistar And Shr Vsmcs, Varun Kumar Soti Jan 2019

Protective Actions Of Oxytocin On Aortic Wistar And Shr Vsmcs, Varun Kumar Soti

Student Theses, Dissertations and Capstones

Background. Oxytocin, known as a female reproductive hormone, has emerged as a cardiovascular hormone showing cardioprotective effects, for instance, negative chronotropic and inotropic effects and ANP release in cardiomyocytes and endothelium. However, oxytocin’s effect in VSMCs remains to be explored. Objectives. This study had three goals: characterizing oxytocin’s effect on proliferation, inflammation, oxidative stress, and vasocontraction pathway in Wistar VSMCs; determining oxytocin’s effect on Ang II; describing oxytocin’s effect and its interaction with Ang II in SHR VSMCs. Methods: Aortic VSMCs were isolated from Wistar rats and SHRs. Hemocytometer, MTT and 3H-Thymidine incorporation assays, and flow cytometry were employed for …

Enhancing The Sublingual Permeability Of Atropine Sulfate: Effect Of Ph And Penetration Enhancers, Rawan Saeed Bafail Jan 2019

Enhancing The Sublingual Permeability Of Atropine Sulfate: Effect Of Ph And Penetration Enhancers, Rawan Saeed Bafail

Student Theses, Dissertations and Capstones

Atropine Sulfate (AS) auto-injector (AtroPen®) is being used as an effective and safe antidote for the treatment of organophosphate (OP) pesticides or nerve gas poisoning. The use of AtroPen® is associated with several drawbacks including: bulky size, availability, affordability, invasiveness, and administration errors. Previously, AS fast disintegrating sublingual tablets (FDSTs) were developed and the feasibility of AS sublingual permeability were demonstrated. However, AS permeability was delayed due to the negative impact of higher doses of AS on FDST’s physical characteristics. Therefore, the aim in this research project was to optimize the previously developed AS FDSTs. It was hypothesized that optimizing …
