The Development Of Novel Apurinic/Aprymidinic Endonuclease/Redox-Factor 1 Inhibitors For The Treatment Of Human Melanoma, Bella Sharifi

Pharmaceutical Sciences (MS) Theses

Apurinic/apyrimidinic DNA repair endonuclease-1 (APE1), first recognized as an important DNA excision repair enzyme, is also known as Redox Factor-1 (Ref-1) involved in the activation of many nuclear transcription factors in both redox-dependent and independent manner. It has been well-documented that the overexpression of APE/Ref-1 contributes to the development of chemo-resistance and is associated with tumor progression in many human malignancies [1].

Our previous study in melanoma demonstrated that the development of novel inhibitors targeting the redox regulation domain of APE/Ref-1 is a promising strategy for melanoma treatment. To date, limited successes have been reported in developing novel …

Proteomics And Biological Evaluation Of Marshmallow (Althea Officinalis) Seeds, Mahshid Amini

Pharmaceutical Sciences (MS) Theses

Marshmallow (Althaea officinalis) belongs to family Malvaceae, a medicinal plant used in traditional and alternative medicine in various parts of the world. Marshmallow is one of the seven medicinal plants that are collectively known as Joshanda. It is a herbal preparation which has been used traditionally as a home remedy for the treatment of common cold, coughs, nasal congestion, and fever. Previous studies showed that crude extract and/or purified compounds from flowers, root, and leaves from Marshmallow have anti-microbial, anti-inflammatory, and other pharmacological effects. However, there is no data reported on the biological activity and the structure of proteins isolated …

Proteomic Evaluation And Cytotoxicity Of Red Maple (Acer Rubrum) Leaves, Saud Alshammari Aug 2019

Proteomic Evaluation And Cytotoxicity Of Red Maple (Acer Rubrum) Leaves, Saud Alshammari

Pharmaceutical Sciences (MS) Theses

Red maple (Acer rubrum), also known as swamp, water or soft maple, is endemic to eastern and central North America and was widely used as traditional medicine by the first peoples. Commercially, its well-known products include maple syrup and high-quality lumber. The potential medicinal benefits of phenolic compounds extracted from the red maple plant, such as glucitol-core containing gallotannins, include antioxidant, and antiglycation effects as well as their importance in cosmetic applications. Plant-derived proteins and peptides are important biomolecules; however to date, there is no published data on the identification of proteins/peptides from red maple leaves. Therefore, the …

Altering The Regiospecificity Of C6 Indole Prenyltransferase Enzymes Towards Drug Development, Ahmed R. Aoun Aug 2019

Altering The Regiospecificity Of C6 Indole Prenyltransferase Enzymes Towards Drug Development, Ahmed R. Aoun

Pharmaceutical Sciences (MS) Theses

Prenyltransferase (PT) biocatalysts are late-stage tailoring enzymes that modify natural products. PTs catalyze the attachment of prenyl moieties to natural product acceptors using pyrophosphate donors. This prenyl modification in small molecules leads to changes in structural and biological activities. Understanding the structural insights as well as the mechanisms by which PTs function allows us to utilize PTs as a unique approach towards drug development. PriB PT is an example of aromatic PTs that has been characterized previously as a L-tryptophan (L-Trp) C6 C-prenyltransferase (C6-C-PT). Analysis of PriB binding pocket, highlighted key residues that might play an important …

Identification Of Molecules By Spectral Imaging, Qamar Alshammari May 2019

Identification Of Molecules By Spectral Imaging, Qamar Alshammari

Pharmaceutical Sciences (MS) Theses

Spectral imaging is a powerful technique which uses the wavelength to identify/quantify the exact location and amount of the molecules. It facilitates the identification of materials and studying their properties through analyzing the way they interact with light. The study of light interaction with elements is called spectroscopy; spectroscopy examines how light behaves in the target and recognizes materials based on their spectral signatures. Spectral signatures can be compared to fingerprints which can be used to identify a person; spectral signatures can be used to identify materials. Therefore, we hypothesize that identifying the exact location and quantity of molecules present …

Targeting Primary Cilia Immune Receptor Proteins For The Treatment Of Polycystic Kidney Disease Mechanisms, Nedaa Alomari Apr 2019

Targeting Primary Cilia Immune Receptor Proteins For The Treatment Of Polycystic Kidney Disease Mechanisms, Nedaa Alomari

Pharmaceutical Sciences (MS) Theses

Background: Primary cilia are cellular organelles project from the cell surface of mammalian cell and play important roles in vertebrate development, organogenesis, health, and others genetic diseases. Primary cilium functions as a mechano-sensor and chemo-sensor. Defect in primary cilia causes the progression of polycystic kidney disease (PKD) which further leads to the inflammatory responses. We, therefore, investigated the role of Toll-like receptors 4 and 9 (TLR) in primary cilia towards PKD.

Purpose: The main purpose of the proposed study is to identify and target the immune reactive proteins i.e. TLRs in the primary cilia. By targeting those primary cilia immune …
