Objectives: This study aimed to i) determine the kinetic changes of immune cell phenotypes, including an exhaustion marker PD-1, in spleens of murine sepsis ii) investigate the susceptibility against secondary fungal infection after sepsis, and iii) investigate the efficacy of an anti-PD-1 treatment in post-sepsis fungal infection. Methods: Cecal ligation and puncture (CLP) was used as a sepsis model and splenocytes post-CLP were assessed by flow cytometry. In addition, secondary post sepsis systemic fungal infections by Candida albicans or Aspergillus fumigatus administration at 5-day post-CLP were performed. Toll like receptor 3 (TLR3) is a member of TLR family functions to recognizes double-stranded RNA (dsRNA) produced by positive-strand RNA viruses and DNA virusesand generate signaling that stimulate body immune system against virus infection. However, recent evidence suggests that TLR3 signaling may also play a role in initiating the immunosuppressive property of the cells. Periodontium is a specialized tissue laying between tooth and surrounding bone. It functions as force cushion. In particular stimulation including mechanical force, periodontal ligament (PDL) cells which are mechanosensitive cells have the ability to secrete specific cytokines and proteins. PDL cells also have the potential to differentiate into osteoblast-like cells or fibroblast-like cells. PDL cells perceive combination of force in oral cavity in which major component is cyclic tensile force (CTF).
