Three Dimensional Localization Of Lesions From Digitized Mammograms, Amy L. Magnus

This thesis describes new algorithms to localize regions-of-interests (ROIs) three dimensionally from a pair of digitized mammograms. This work is intended to add a layer of sophistication to computer aided diagnosis by putting to use the fixed and measurable parameters of mammographic imaging. The fixed parameters are the source to film orientation and the podium-image distance. Measurable parameters are the rotation angle of the x-ray tube from the horizontal and the compression depth (the distance from compression paddle to contact podium distance) at the time of imaging. Prostate cancer is the most common malignancy and the second leading cause of cancer death in males in the United States. Additionally, the number of deaths attributed to prostate cancer is increasing at a rate of approximately 8% a year. Development of new diagnostic and therapy strategies are needed in order to improve the life expectancy of patients with this disease. One tool which may allow for improvements in prostate cancer diagnosis and therapy is the monoclonal antibody (MAb) 7E11-C5.3 which was first described in 1987. Antibodies against HIV-1 proteins in HIV-1-infected individuals share a cross-reactive idiotype defined by the monoclonal antiidiotypic antibody 1F7 (5). Using a computer algorithm based on the molecular recognition theory, regions of inverse hydropathy between the variable sequence of 1F7 and human monoclonal anti-HIV-1 antibodies were identified, which are assumed to be involved in idiotype-antiidiotype contacts. A peptide was designed from the proposed contact in the variable heavy chain framework 3-complementarity determining region 3 (FR3-CDR3) of human antibodies and was synthesized. We have generated an IgG1 murine monoclonal anti-idiotype antibody (Ab2) designated 3H1, which mimics a specific epitope on the carcinoembryonic antigen (CEA). Patients with CEA positive tumors are immunologically "tolerant" to CEA. We used 3H1 as a surrogate for CEA for vaccine therapy of 12 patients with advanced colorectal cancer. Each of the patients received a minimum of four intracutaneous injections of aluminum hydroxide precipitated 3H1 at either 1, 2, or 4 mg dosage per injection. 9 of 12 patients demonstrated anti-anti-idiotypic (Ab3) response to 3H1. Several mathematical models are developed to describe the effects of chemotherapy on both cancerous and normal tissue. Each model is defined by either a single homogeneous equation or a system of heterogeneous equations which describe the states of the normal and/or cancer cells. Periodic terms are added to model the effects of the chemotherapy. What we obtain are regions, in parameter space (dose and period), of acceptable drug regimens.

The models take into account various aspects of chemotherapy. These include, interactions between the cancer and normal tissue, cell specific chemotherapeutic drug, the use of non-constant parameters to aid in modeling …

More importantly, this study has shown that prophylactic mastectomy surgery is a viable and worthwhile treatment to prevent breast cancer recurrence. It is hoped that this information will increase the knowledge of breast cancer. For the same reason, this paper has attempted to explain what cancer is while describing how the genetics of a person cause or contribute to its development. Most of all, it is hoped that this paper will emphasize the severity of this disease in the United States today and tomorrow .

A logistic differential equation with a time-varying periodic parameter is used to model the growth of cells, in particular cancer cells, in the presences of chemotherapeutic drugs. The chemotherapeutic effects are modeled by a periodic parameter that modifies the growth rate of the cell tissue. A negative growth rate represents the detrimental effects of the drugs. A simple criterion is obtained for the behavior of the chemotherapy.

A 66 year old gentleman was diagnosed to have metastatic adenocarcinoma of the prostate in 1990, following a CT scan of the abdomen and trans-uretheral resection of prostate (TLJRP). He was started on estrogen therapy (Fos­festerol sodium). Two years later he presented to the emer­gency room with breathlessness and edema of the left ann of a week’s duration. He admitted to persistence of symptoms of pmstatism and was found to have bilateral axillary lymphade­nopathy and pitting edema of the left arm.
