Comparison Of Standard Agglutination Tests, Enzyme Immunoassay, And Coombs Gel Test Used In Laboratory Diagnosis Of Human Brucellosis, Emi̇ne Emel Koçman, Memnune Selda Erensoy, Meltem Taşbakan, Meltem Çi̇çekli̇oğlu

Background/aim: It was aimed to evaluate the results of Rose Bengal (RB), ELISA total tests (IgM and IgG), and the Brucella Coombs gel test (BCGT), which are used as screening tests, with the combined results of a tube agglutination test (standard Wright test: SWT) and a tube agglutination test with antihuman globulin (AHG TAT). Materials and methods: Samples from 97 patients prediagnosed with brucellosis (age ≥18 years) were screened with RB, ELISA, and BCGT. SWT < 160 samples and RB-negative but ELISA- or BCGT-positive samples were tested by AHG TAT. SWT ≥ 160 or AHG TAT ≥ 160 was taken as reference. Results: Thirty-two of 56 RB-positive samples and one RB-negative but ELISA- and BCGT-positive sample were found to be ≥160 with SWT or AHG TAT. Sensitivity, specificity, accuracy, and agreement (kappa) values according to SWT ≥ 160 or AHG TAT ≥ 160 positivity were as follows, respectively: RB 96.9%, 62.5%, 74.2%, and 0.509; ELISA total 100%, 60.9%, 74.2%, and 0.515; BCGT test 97%, 70.3%, 79.4%, and 0.594. Conclusion: Sensitivities of the screening tests are similar, but positivities should be confirmed by more specific tests. Background/aim: Parvovirus risk in blood transfusion has become a popular research topic since there are limited data on parvovirus seroprevalence in blood donors in Turkey. The aim of this study was to investigate parvovirus seroprevalence in blood donors in Turkey. Materials and methods: Blood samples of 988 blood donors admitted to a university blood bank were obtained for parvovirus B19 IgM and IgG detection. The samples were analyzed using the ELISA method. The present study was undertaken to ascertain the role of NO in central nervous system (CNS) infection by Mycobacterium tuberculosis. Background/aim: Strongyloides stercoralis causes life-threatening hyperinfection or disseminated strongyloidiasis in immunocompromised patients such as HIV-positive, organ transplantation, and cancer patients. This study investigated the presence of strongyloidiasis in immunocompromised patients for the first time in Turkey. Background/aim: Midkine (MK), a heparin-binding growth factor, has an important role in cancer progression. The aim of this study was to determine MK expression in breast tissue and the preoperative and postoperative serum levels of patients with breast cancer. Materials and methods: Sixty-one patients with breast cancer participated in our study. The MK serum levels were measured pre- and postoperatively for these patients. To determine the possible relationship between toxoplasmosis and patients with first-episode schizophrenia (FES). Materials and methods: Seventy-three subjects with FES (15-54 years old; mean: 23.4 years) and 40 healthy individuals (20-54 years old; mean: 30.3 years) were enrolled in the study. Most of the FES patients (90.4%) and the control individuals (95.0%) were male. The aim of this study was to develop a human tumor necrosis factor alpha (hTNF-\alpha) ELISA system because specific and sensitive measurement of low levels of circulating TNF-a is very important for enlightening the immunopathologial mechanisms associated with TNF-\alpha. Monoclonal antibodies 6A4c and 8A6 were produced against hTNF-\alpha and were used as the capture antibody and the tracer antibody respectively for the first hTNF-\alpha ELISA system (6A4c/biotin-8A6). We performed a serological evaluation of herpes simplex virus (HSV) infections at the time of labor. In order to determine the seroprevalence of asymptomatic HSV infections in pregnant women, blood samples were collected for antibody measurement from 130 randomly selected pregnant women. Presence of HSV IgG and IgM antibodies in the serum samples was tested by ELISA. The prevalence of HSV-1 gG1 antibodies was 90% (117/130), that of HSV-2 gG2 antibodies was 63.1% (82/130) and that of HSV-2 IgM antibodies was 13.8% (18/130). Microalbuminuria is considered to be an early indicator of diabetic nephropathy. We have previously reported the establishment of A-HAlb/98 hybridoma secreting monoclonal antibody that specifically recognizes human serum albumin (HSA). In this report, an in-house enzyme-linked immunosorbent assay (ELISA) was developed using this antibody for the measurement of urinary albumin (UA) and UA was examined in 24 different samples. A-HAlb/98 was used as the capture antibody by immobilizing onto microtitre plates in ELISA format. The lack of a rapid and reliable method for diagnosis of active tuberculosis necessitated the development of new laboratory techniques. Diagnostic value of a re-cently developed ELISA for the detection of lgA antibodies to Kp 90 antigen of Mycobacterium tuberculosis was evaluated in this study and the results were compared with the conventional methods and polymerase chain reaction (PCR). Fifty six sera and body fluid (pleural, peritoneal, pericardial) samples from 23 patients with ac-tive tuberculosis and 26 control subjects were included in the study.
