Human cells that live in biological harmony can become cancerous. As they do so, they engage in anaerobic respiration, undergo rapid cell growth and division according to nonmitotic rules, and important changes take place in their metabolism. We conducted several investigations in an effort to block or change the metabolic characteristics of cancer cells using a typical cancerous cell line. Specifically,we investigated the effects of sodium selenite (Na_2 SeO_ 3 ) on antioxidant enzyme activities and reduced glutathione levels in Hepatoma G_ 2 cells, cells that show rapid glutamine uptake and metabolism. A Hepatoma G_ 2 cell line was cultured …

Turkish Journal of Medical Sciences

The aim of the present study was to investigate the role of the L-type voltage dependent Ca^2^+ channel blocker, nimodipine, in kainate induced toxicity in retinal ganglion cells of mice. Kainate in 10mg/kg was administered intraperitoneally following the vehicle or nimodipine. Nimodipine, 45 minutes prior to kainate in 10, 15, 25, 50, 100 and 250 mg/kg doses was administered intraperitoneally. Surprisingly, nimodipine potentiated the ganglional cell death induced by kainate, with respect to the control and kainate+vehicle treated group. In accordance with our results, blockage of L-type calcium channels by nimodipine may worsen the prognosis and survival of retinal neurons …

The Use Of A Nonradioactive Digoxigenin Labeled Probe For Detection Of Apo(A) Gene Size, Yeşi̇m İlçöl, Bülent İlçöl, Yavuz Taga

Turkish Journal of Medical Sciences

Elevated lipoprotein(a) [Lp(a)] concentrations are associated with premature coronary heart disease and early myocardial infarction. In the general human population the sizes of apolipoprotein [apo(a)] isoforms are inversely associated with lipoprotein(a) levels. Large isoforms are associated with low Lp (a) and small isoforms with high Lp(a) in the plasma. The size of the polymophism is directly correlated with the number of kringle-4 domains in apo(a). It has been shown by quantitative Southern blotting, and more recently by pulsed-field gel electrophoresis, that polymophism is the result of differences in the number of tandem kringle-4 repeats in the apo(a) gene. In our …

Establishment Of A Nonradioactive Molecular Diagnosis Of Fragile-X Syndrome, Zehra Oya Uyguner, Bernd Wollnik, Hülya Kayseri̇li̇, Turgut Tükel, Seher Başaran, Memnune Yüksel Apak

Turkish Journal of Medical Sciences

Fragile-X syndrome is a hereditary dynamic mutation disorder, predominantly caused by a large expansion of CGG trinucleotide repeats in the FMR 1 gene leading to methylation and down regulation of transcription of the gene. For the molecular diagnosis of the disease, the repeat locus in FMR 1 gene is primarily detected by Southern blotting with radioactively labeled probes. Unusually GC rich composition of the expanded region caused technical difficulties during PCR based testing and therefore is not performed by most diagnostic laboratories. We established a Southern blot method and a novel PCR protocol which enables the amplification of normal, premutated …

Telomeric Repeats Of Immortal Hamster Cells, Nedi̇me Serakinci, Jorn Koch

Turkish Journal of Medical Sciences

We have analyzed telomeres in four hamster cell lines to investigate how telomeres look in cells grown past the Hayflick limit. In all four cell lines telomeric signals were seen after detection with two different sensitive in situ methods (FISH with PNA probes and PRINS). The telomeric signals were not very strong, suggesting that the repeats are very short, but were nevertheless clearly visible. With the PRINS technique, it was further possible to probe also for the presence of variant telomeric repeats at the chromosome ends. With some of the variant repeat probes, a difference was seen between probes for …
