As the prevalence of cardiovascular diseases continues to exponentially grow in populations across the globe, the necessity of determining underlying factors, effective methods of diagnoses, and universally available preventive measures also grows. Early detection of endothelial dysfunction, a proven precursor of cardiovascular diseases, can be extremely impactful in encouraging preventative measures and early intervention before medical conditions become chronic. In recent years, ice plunging, a form of cryotherapy involving full body immersion in cold water, has gained popularity within circles of fitness and health practitioners, gaining the interest of people of all backgrounds. Certain parallels observed between the human physiological …
Investigating The Viral Nature Of Neuronal Capsid Genes, Michael Manuel Long
Investigating The Viral Nature Of Neuronal Capsid Genes, Michael Manuel Long
Master's Theses
Memory formation is an essential part of everyday life. Neuronal communication is thought to be a key driver behind memory formation. There are two heavily studied methods of neuronal communication: chemical (neurotransmitter-mediated) and electrical (physically attached). When issues arise environmentally or genetically, people can experience one of many devastating memory disorders i.e. Alzheimer’s disease and other Dementias. Due to their prevalence, the treatments for disorders that hinder memory formation are a high priority for pharmaceutical companies. Currently, many of the treatments available focus on neurotransmitters or neuronal activity, and scientists are far from a cure. However, there may be an …
Identifying And Minimizing Underspecification In Breast Cancer Subtyping, Jonathan Cheuk-Kiu Tang
Identifying And Minimizing Underspecification In Breast Cancer Subtyping, Jonathan Cheuk-Kiu Tang
Master's Theses
In the realm of biomedical technology, both accuracy and consistency are crucial to the development and deployment of these tools. While accuracy is easy to measure, consistency metrics are not so simple to measure, especially in the scope of biomedicine where prediction consistency can be difficult to achieve. Typically, biomedical datasets contain a significantly larger amount of features compared to the amount of samples, which goes against ordinary data mining practices. As a result, predictive models may fail to find valid pathways for prediction during training on such datasets. This concept is known as underspecification.
Underspecification has been more accepted …
Delayed Testosterone Treatment In A Rodent Model Of Repeat Mild Traumatic Brain Injury Attenuates Mechanisms Of Cellular Death In The Vestibular Nucleus, Michael Volyanyuk
Delayed Testosterone Treatment In A Rodent Model Of Repeat Mild Traumatic Brain Injury Attenuates Mechanisms Of Cellular Death In The Vestibular Nucleus, Michael Volyanyuk
Master's Theses
A common complaint after repetitive mild traumatic brain injury (rmTBI) is vestibular dysfunction, which can become persistent and decrease quality of life. Delayed testosterone treatment has been shown to reduce vestibular impairment and improve neuronal survival in the vestibular nucleus following rmTBI; however, the long-term molecular pathways associated with rmTBI and testosterone treatment remain widely unexplored. We utilized a 5-hit closed-head rodent rmTBI model and administered testosterone 35 days after injury (DPI). The vestibular nuclei were removed and prepared for RT-qPCR and western blotting. Gene expression and protein levels were elevated for markers of oxidative stress (NADPH oxidase 4) and …
Healing Sanctuary, Kammy Downs
Healing Sanctuary, Kammy Downs
Master's Theses
The natural world has hidden wisdom and resources that are disregarded as we misuse our responsibility to care for them. The benefits of being in tune with nature have been traded for a culture that manipulates nature on the altar of expediency, waste, and unquenchable desire. Healing Sanctuary presents a duality. My work speaks about the relationship between the seen and unseen, represented by ten larger-than-life drawings of medicinal herbal plants that have had a profound physical healing effect on me. At the same time, plants, roots, and seeds create visceral metaphors for mental health. Our accomplishments, outlook on life, …
Glial Endothelial Brain Cell Co-Cultures For Testing Signaling Response And Delivery Of Novel Materials Across Blood Brain Barrier, Neela Prajapati
Glial Endothelial Brain Cell Co-Cultures For Testing Signaling Response And Delivery Of Novel Materials Across Blood Brain Barrier, Neela Prajapati
Master's Theses
The brain accounts for 20% of overall energy metabolism in the body though it just comprises 2% of the total body mass but has a limited capacity of storing energy unlike other critical organs in the body such as the heart and liver. This energy along with oxygen and nutrients is supplied by cerebral blood flow (CBF), any interruption of which can cease the brain function within seconds with a potential irreversible neuronal injury, within minutes. Vascular cells along with astrocytes and neurons are a part of a recently developed concept known as the Neurovascular Unit responsible for Neurovascular coupling …

Master's Theses
Irritable bowel syndrome (IBS) is a common gastrointestinal (GI) disorder that affects an estimated 1 in 10 people globally (Black & Ford, 2020). Since IBS is such a common GI disorder worldwide it is important to understand that impact it has on quality of life. This study assessed the differences in the effects of IBS on the quality of life in patients with each of the three types of IBS: IBS-D, IBS-C, and IBS-M. Potential respondents were recruited via social media using an online survey, which collected demographics and assessed the effects of IBS on quality of life. The survey …
Development Of A Robust Methodology To Obtain And Assess Myogenic Precursor Cells For Their Use In Regenerative Therapies, Ricardo Lasa
Development Of A Robust Methodology To Obtain And Assess Myogenic Precursor Cells For Their Use In Regenerative Therapies, Ricardo Lasa
Master's Theses
Peripheral arterial occlusive disease (PAOD) is characterized by buildup of atherosclerotic plaque in peripheral arteries that leads to an occlusion that can interrupt the supply of blood to the peripheral tissue, causing downstream tissue ischemia/hypoxia. PAOD is estimated to affect over 200 million patients worldwide. Current surgical revascularization treatments can be effective in about half of the patient population, leading to a significant number of patients with no treatment options beyond pharmacological intervention and lifestyle modification. The decrease in blood flow downstream of the occlusion leads to increased blood pressure gradient in the microvasculature, specifically in vessels that connect arterial …

A Comparison Of Power Variables For Two Loading Methods In Weighted Vertical Jumps 2020, Jonathan Tanguay
Master's Theses
Training exercises to develop muscular strength and power, such as the weighted jump squat, commonly utilize barbells and dumbbells to increase resistance. Strength coaches often measure improvements in peak force, peak power, and rate of force development to determine effectiveness of a training program. The purpose of this study was to determine if equipment selection (barbells vs. dumbbells) affects peak force, peak power, and/or rate of force development in weighted jump squats. Thirteen physically active, college-aged males (age: 21.6 ± 2.0 years, height: 182.8 ± 9.7 cm, body mass: 87.2 ± 9.0 kg, lean mass: 72.3 ± 8.1 kg) performed …

Evaluation Of Endothelial Cell Responses To Elevated Glucose, Gabriella Sugerman
Master's Theses
Developing a tissue-engineered Blood Vessel Mimic (BVM) to represent diabetic macrovascular disease could expedite design of new vascular devices specifically tailored to diabetic patients. In contribution toward this model, this thesis assessed Human Umbilical Vein Endothelial Cell (HUVEC) responses to high glucose conditions. Interleukin 6 (IL-6) and Cluster of Differentiation 36 (CD36) were selected to signify oxidative stress activity, a hallmark of diabetic macrovascular disease. Next, activity of potential reference genes B2M, HPRT1, and ACTB was assessed. All genes were found to exceed acceptable variability, so the E-ΔC T method of data analysis was selected. Next, cellular responses to high …

The Acquisition And Analysis Of Electroencephalogram Data For The Classification Of Benign Partial Epilepsy Of Childhood With Centrotemporal Spikes, Jessica A. Scarborough
Master's Theses
In this thesis, I will expand upon each step in the process of acquiring and analyzing electroencephalogram (EEG) for the classification of benign childhood epilepsy with centrotemporal spikes. Despite huge advancements in the field of health informatics—natural language processing, machine learning, predictive modeling—there are significant barriers to the access of clinical data. These barriers include information blocking, privacy policy concerns, and a lack of stakeholder support. We will see that these roadblocks are all responsible for stunting biomedical research in some way, including my own experiences in acquiring the data for the second chapter of this thesis.
This second chapter …
The Unconventional Secretion Of Alpha-Synuclein By Autophagic Mechanisms And The Novel Characterization Of Pathological Alpha-Synuclein Associated Extracellular Vesicles, Kevin Burbidge
Master's Theses
Synucleinopathies are a group of neurodegenerative diseases associated with the intracellular accumulation of aggregated α-synuclein, a neurotoxic process resulting in cell death. Research indicates that pathology results from α-synuclein misfolding and self-propagation, as well as, cell-to-cell transmission. Furthermore, pathological α-synuclein transmission may occur through unconventional secretion.
In this thesis, we investigated how autophagy influences unconventional secretion of our novel α-synuclein dual-split protein model that we first verified by reproducing documented results from treatment of known autophagic regulators. Afterwards, we investigated how knocking out autophagic proteins ATG7 and Beclin-1 influenced α-synuclein secretion.
Additionally, we developed a methodology for analyzing extracellular vesicles …
Effects Of Prebiotics On Gut Bacterial Communities And Healing Of Induced Colitis In Mice, Krystyn Elizabeth Davis
Effects Of Prebiotics On Gut Bacterial Communities And Healing Of Induced Colitis In Mice, Krystyn Elizabeth Davis
Master's Theses
Inflammatory Bowel Diseases (IBD) cause chronic inflammation of the gastrointestinal tract and debilitating symptoms in those suffering from the diseases. After inducing colitis in a mouse model using Dextran Sulfate Sodium (DSS), prebiotics inulin and oligofructose enriched inulin (OEI) were used as treatments to determine their effects on the gut microbial community, physiological healing process, and immune response in the mice after initial inflammation and before subsequent inflammation, or relapse. The treatment with inulin led to an increase in regulatory T cell number, but this increase was not as significant as the increase induced by the OEI. Inulin increased the …
Power Output And Loads On The Lumbar Vertebrae During A Power Clean 2016, Joseph Keleher
Power Output And Loads On The Lumbar Vertebrae During A Power Clean 2016, Joseph Keleher
Master's Theses
The purpose of the present study was to compare the peak resultant torque, shear, and compressive forces acting at the L3/L4 junction of the lumbar vertebrae and the peak power output during 40% one repetition maximum (1RM) and 80% 1RM attempts of the power clean. It was hypothesized that performance of a power clean at 80% 1RM will result in greater peak resultant compressive force, torque and shear force at the L3/L4 junction of the lumbar spine than that of a 40% 1RM attempt. Power cleans at 80% 1RM will also result in higher maximum instantaneous power outputs than that …
Cellular Consequences Of Lysosomal Rupture Induced By Alpha Synuclein, Zachary Carpenter Green
Cellular Consequences Of Lysosomal Rupture Induced By Alpha Synuclein, Zachary Carpenter Green
Master's Theses
Misfolded, toxic forms of amyloid alpha synuclein (α-syn) have been implicated in Parkinson’s disease (PD) pathology and have been shown to spread throughout the brain in a prion-like manner, explaining the progressive loss of neurons in PD. The mechanisms of spread and cell death are unknown, however, evidence from previous studies suggests that neurons may undergo apoptosis due to the uptake of exogenous α-syn aggregate through the endocytic pathway and subsequent lysosomal rupture.
This study aimed to corroborate previous evidence of α-syn induced lysosomal rupture and investigate the cellular repercussions of rupture with respect to cell death and α-syn transfer. …
Estrogen Regulation Of Mir-181a Stability In Neurons, Chun Kim
Estrogen Regulation Of Mir-181a Stability In Neurons, Chun Kim
Master's Theses
Estrogens are an important class of steroid hormones involved in many cellular signaling processes. Levels of estrogens fluctuate across the lifespan suggesting that they target intracellular mediators, such as microRNAs, to fine-tune homeostatic cellular processes. Previous work in our lab demonstrated that 17β-estradiol (E2), the predominant circulating estrogen, stabilized miR-181a in a neuronal cell line derived from rat hypothalamus. These results lead to the hypothesis that E2 regulated miR-181a stability by altering the pool of endogenous RNA and by recruiting PNRC2 away from the miRNA degradation machinery. By utilizing miRNA degradation assays and immunostaining techniques, it was determined that the …

Through layer-by-layer method, the authors have constructed three Au nanoplexes: AuPEI/RNA/PEI, AuPEI/RNA/PEI-mPEG, and AuPEI/NA/PEI-PEG-FA. All the nanoplexes are characterized by UV-vis spectrometry, DLS, and zeta potential. The surface density of the first layer PEI and second layer RNA were also determined. Moreover, the Au nanoplexes can protect siRNA from RNase degradation and are stable in cell culture medium.
siGLuc-ppp, ssRNA80, and siF17 were delivered by the assembled Au nanoplexes, and the results were analyzed by GLuc assay, TB assay, and Luciferase assay. In terms of RNA delivery, the Au nanoplexes AuPEI/RNA/PEI exhibited excellent gene silencing efficiency (or causing …
Propagation Of Oligomeric Α-Synuclein And Amyloid-Β: Implications For Parkinson's And Alzheimer's Diseases, Matthew Stephen Planchard
Propagation Of Oligomeric Α-Synuclein And Amyloid-Β: Implications For Parkinson's And Alzheimer's Diseases, Matthew Stephen Planchard
Master's Theses
The aggregation of amyloidogenic proteins is a critical event in the pathology of a variety of neurodegenerative diseases, including Alzheimer’s disease (AD) and Parkinson’s disease (PD). The proteins α-synuclein (αS) and amyloid-β (Aβ) are involved in the formation of amyloid lesions observed in PD and AD, respectively. Both PD and AD exhibit a significant amount of co-pathology in clinical settings, and the αS and Aβ proteins have been shown to interact in vitro. Recent experimental consensus has shown oligomeric species to be significant, if not primary, sources of toxicity in these diseases. …
A Study Of Functional Recovery And Axonal Plasticity Following Tbi And Anti-Nogo-A Immunotherapy, Jennifer Crystal Biesiadecki
A Study Of Functional Recovery And Axonal Plasticity Following Tbi And Anti-Nogo-A Immunotherapy, Jennifer Crystal Biesiadecki
Master's Theses
Traumatic brain injury (TBI) is a leading cause of morbidity and mortality globally and often results in long term disability. Unfortunately, the success of rehabilitation techniques as therapy for TBI is limited, which may be due to the growth restrictive environment of the adult central nervous system (CNS). This environment is thought to result from glial scars, lack of neurotrophic factors, and the presence of an array of growth-inhibitory molecules. One such growth-inhibitory molecule is the protein Nogo-A. Our laboratory has shown that neutralization of Nogo-A with anti-Nogo-A immunotherapy after ischemic stroke results in improvement in functional recovery in the …
Biochemical Assay Optimization And Computational Screening Efforts To Identify Potential Luxs Inhibitors, Keeshia Q. Wang
Biochemical Assay Optimization And Computational Screening Efforts To Identify Potential Luxs Inhibitors, Keeshia Q. Wang
Master's Theses
Quorum sensing (QS) is a process of coordination of bacterial gene expression in response to cell population. System two QS is regulated by the small signaling molecule autoinducer-2 (AI-2) and is implicated in the infectious behaviors of various bacterial species. AI-2 is biosynthesized from S-ribosylhomocysteine (SRH) by the enzyme LuxS and induces interspecies cell-to-cell communication. Inhibition of LuxS would therefore inhibit interspecies QS. Herein, a search for novel molecular species that will competitively bind with SRH in the LuxS binding site is performed in silico. Computational screening results are then validated in vitro using an optimized LuxS inhibition …
Associations Between Alcohol Consumption And Fasting Blood Glucose In Young Adults, Julie Ann Lucca
Associations Between Alcohol Consumption And Fasting Blood Glucose In Young Adults, Julie Ann Lucca
Master's Theses
Current research shows moderate alcohol consumption is associated with decreased risk of diabetes and excessive consumption or binge drinking can cause insulin resistance and diabetes. In 2010, diabetes was the seventh leading cause of death in the United Statesand was responsible for significant health complications: blindness, kidney failure, and limb amputations, and is a large national economic burden. Fasting blood glucose (FBG) is a tool used to help diagnose diabetes. Abnormally high FBG, ≥100 mg/dl, is indicative of diabetes and pre-diabetes. Few studies have observed diabetic prevalence among young adults or college students. Studying young adults can help provide added …
The Role Of Dax-1 In Regulating Pluripotency In Mouse Embryonic Stem Cells, Anthony Torres
The Role Of Dax-1 In Regulating Pluripotency In Mouse Embryonic Stem Cells, Anthony Torres
Master's Theses
The orphan receptor Dax-1 is highly expressed in pluripotent embryonic stem (ES) cells and shows a correlative reduction in expression as these cells differentiate. While it is known that Dax-1 is expressed in pluripotent mouse ES cells, the precise function of Dax-1 in these cells is not as well understood. Recent studies employing RNA interference (RNAi) to specifically reduce the expression of the Dax-1 gene in mouse ES cells found that upon the knock down of Dax-1, ES cells differentiated. These findings indicate that Dax-1 functions in a novel role in the maintenance of a relatively undifferentiated state in ES …
Alpha-Synuclein Aggregates Activate The Nlrp3 Inflammasome Following Vesicle Rupture, Rudy Orlando Cedillos
Alpha-Synuclein Aggregates Activate The Nlrp3 Inflammasome Following Vesicle Rupture, Rudy Orlando Cedillos
Master's Theses
Parkinson's disease (PD) and related synucleinopathies are progressive neurodegenerative disorders that feature the accumulation of intracellular inclusions known as Lewy bodies (LBs) in the brain. The presynaptic protein α-synuclein is the primary constituent of LBs and has been documented to play a major role in the pathogenesis of synucleinopathies. Recently, aggregated α-synuclein has been implicated in prompting microglia-mediated inflammation, a process associated with the progression of neuronal death in neurodegenerative disorders. Although the mechanisms surrounding the induction of neuroinflammation are not well understood, the recently discovered inflammasome-forming NLR proteins have emerged as regulators of inflammation. In this study, we sought …

Resveratrol + Ethanol Preconditioning And Neuroprotection, Nastaran Khodaie
Master's Theses
Our lab established that moderate ethanol preconditioning (MEP) for 6 days blocked the Aâ25-35 toxicity in cerebellar mixed cell cultures and It has been shown that MEP cause the up regulation of the pre-survival enzymes such as proxiredoxin2 (PRX2), which suggests MEP can activate the cell survival pathways against upcoming toxins.
This study was performed to verify the possible combinatory effect of ethanol and resveratrol against Aâ25-35 toxicity. Both ethanol and resveratrol sub-protective concentrations were detected against Aâ25-35 in cerebellar mixed cell cultures. The combination of sub-protective levels of ethanol and resveratrol showed the significantly reduction in number of Aâ25-35 …

Correlations with algal community NMDS axis scores and the relative …
Determination Of The Molecular Mechanisms Of Hyperglycemia-Induced Changes In Cav3.2 Calcium Channel Properties, Ginny Marie Keeling
Determination Of The Molecular Mechanisms Of Hyperglycemia-Induced Changes In Cav3.2 Calcium Channel Properties, Ginny Marie Keeling
Master's Theses
Hyperglycemia can cause altered excitability due to increased CaV3.2 T-type calcium channel function, bestowing diabetics an increased neuropathy risk. The objective of this study was to understand the molecular mechanisms of increased CaV3.2 function during hyperglycemia in vitro, which leads to a 58% increase in current density and increased channel open probability (PO), among other changes. Two major findings are reported in this study:
1) The enzymatic elimination of extracellular N-acetylneuraminic acid (NANA) moieties reversed the increase in current density and the shift in PO, showing that hyper-glycosylation alters channel function in disease.
2) Treatment with GM1 (a ganglioside highly …
Ischemia Impairs Vasodilation In Skeletal Muscle Resistance Artery, Kyle Remington Struthers
Ischemia Impairs Vasodilation In Skeletal Muscle Resistance Artery, Kyle Remington Struthers
Master's Theses
Functional vasodilation in arterioles is impaired with chronic ischemia. We sought to examine the impact of chronic ischemia and age on skeletal muscle resistance artery function. To examine the impact of chronic ischemia, the femoral artery was resected from young (2-3mo) and adult (6-7mo) mice and the profunda femoris artery diameter was measured at rest and following gracilis muscle contraction 14 days later using intravital microscopy. Functional vasodilation was significantly impaired in ischemic mice (14.4±4.6% vs. 137.8±14.3%, p<0.0001 n=8) and non-ischemic adult mice (103.0±9.4% vs. 137.8±14.3%, p=0.05 n=10). In order to analyze the cellular mechanisms of the impairment, a protocol was developed to apply pharmacological agents to the experimental preparation while maintaining tissue homeostasis. Endothelial and smooth muscle dependent vasodilation were impaired with ischemia, 39.6 ± 13.6% vs. 80.5 ± 11.4% and 43.0 ± 11.7% vs. 85.1 ± 10.5%, respectively. From this data, it can be supported that smooth muscle dysfunction is the reason for the observed impairment in arterial vasodilation.
Comparison Of An Ultrasonic And A Non-Ultrasonic Endodontic Irrigation Protocol: A Clinical Study, Christopher M. Beus
Comparison Of An Ultrasonic And A Non-Ultrasonic Endodontic Irrigation Protocol: A Clinical Study, Christopher M. Beus
Master's Theses
The use of irrigation as a method of rendering canals bacteria free continues to be a source of investigation. Recent data suggests that an activated irrigation protocol such as passive ultrasonic irrigation can result in decreased intracanal bacteria. In addition, studies have shown that the activation of various irrigants such as NaOCl and Chlorhexidine can increase the sum antimicrobial effect. The purpose of this randomized, double blinded, prospective in-vivo clinical study was to compare the results of a non-activated irrigation protocol (NAI) using only NaOCl 1.5% to a passive ultrasonic irrigation protocol (PUI) using NaOCl 1.5%, EDTA 17% and Chlorhexidine …
Protein Kinase C Δ Is Critical For Nucleotide Excision Repair Of Cyclobutane Pyrimidine Dimers, Christopher Michael Negro
Protein Kinase C Δ Is Critical For Nucleotide Excision Repair Of Cyclobutane Pyrimidine Dimers, Christopher Michael Negro
Master's Theses
Nucleotide excision repair (NER) is the process by which cells identify and repair bulky, helix-distorting DNA lesions such as ultraviolet (UV) radiation-induced cyclobutane pyrimidine dimers (CPD) and 6-4 photoproducts (6-4PP). NER in human cells is a complex biochemical process requiring several complementation groups. Defects in complementation group genes result in rare, autosomal recessive cancer syndromes such as xeroderma pigmentosum (XP), cockayne syndrome (CS), and trichothiodystrophy, supporting the tumor suppressive effects of NER. Protein Kinase C delta (PKCδ) expression is lost in squamous cell carcinomas where it functions as a tumor suppressor gene. Additionally, in response to UV radiation, PKCδ is …

Morphological Development Of The Axial Skeletons Of Esox Lucius And Esox Masquinongy (Euteleostei: Esociforms), With Comparisons In Developmental And Mineralization Rates., Amanda Maria Burdi
Master's Theses
Developmental morphology of the axial skeleton of Esox lucius (i.e., northern pike and type species) and E. masquinongy (i.e., muskellunge) was investigated. More than 1,000 specimens were examined ranging in size from approximately 10 mm notochordal length (NL) post-hatching juveniles to over 80 mm standard length (SL) foraging sub-adults. Results show that regardless of individual variation, the relative sequence of bone formation and mineralization is consistent between the two species. This consistent developmental pattern enabled construction of an ontogenetic staging scheme of eight developmental stages, each characterized by one defining criterion. The first appearance in cartilage and/or first sign of …
