Startle disease is an inherited neurological disorder that causes affected individuals to suffer noise- or touch-induced non-epileptic seizures, excessive muscle stiffness and neonatal apnea episodes. Mutations known to cause startle disease have been identified in glycine receptor subunit (GLRA1 and GLRB) and glycine transporter (SLC6A5) genes, which serve essential functions at glycinergic synapses. Despite the significant successes in identifying startle disease mutations, many idiopathic cases remain unresolved. Exome sequencing in these individuals will identify new candidate genes. To validate these candidate disease genes, zebrafish is an ideal choice due to rapid knockdown strategies, accessible embryonic stages, and stereotyped behaviors. The …

INTRODUCTION: Current studies indicate that triple negative breast cancer (TNBC), an aggressive breast cancer subtype, is associated with poor prognosis and an early pattern of metastasis. Emerging evidence suggests that MUC4 mucin is associated with metastasis of various cancers, including breast cancer. However, the functional role of MUC4 remains unclear in breast cancers, especially in TNBCs.

Transcriptional activation by estrogen receptor (ER) is a key step to breast oncogenesis. Given previous findings that ADA3 is a critical component of HAT complexes that regulate ER function and evidence that overexpression of other ER coactivators such as SRC-3 is associated with clinical outcomes in breast cancer, the current study was designed to assess the potential significance of ADA3 expression/localization in human breast cancer patients. The long-term consequences of traumatic brain injury (TBI), specifically the detrimental effects of inflammation on the neurogenic niches, are not very well understood. In the present in vivo study, we examined the prolonged pathological outcomes of experimental TBI in different parts of the rat brain with special emphasis on inflammation and neurogenesis. Sixty days after moderate controlled cortical impact injury, adult Sprague-Dawley male rats were euthanized and brain tissues harvested. To evaluate the utility of immunohistochemical staining of cytokeratin 19 (CK19), HBME-1, and galectin-3 in cell block preparations of fine-needle aspiration cytology specimens as ancillary techniques for the differential diagnosis of papillary thyroid carcinoma. Materials and methods: Immunohistochemical detection of CK19, HBME-1, and galectin-3 was completed in 45 thyroid aspiration cytology specimens fixed in paraffin-embedded blocks consisting of the following cytologic diagnoses: 1) 16 cases of papillary carcinoma, 2) 16 cases suspicious for papillary carcinoma, and 3) 13 cases involving nonneoplastic lesions. To investigate the role of cell adhesion molecules CD44v6 and CEA in the occurrence, development, and metastasis of colorectal cancer (CRC). Materials and methods: The expression of CD44v6 and CEA in 76 CRC tissues and 43 normal colon tissues was detected using an immunohistological technique. The relationship between the 2 molecules’ expression and their pathologic characteristics, especially for metastasis, was investigated. Furthermore, the relevance between the expression of CD44v6 and of CEA was also assessed. Results: Expression of CD44v6 and CEA protein in CRC tissues was higher than in paracarcinoma tissue and normal colorectal mucosa (both P < 0.001). Positive expression of CD44v6 in CRC was associated with lymph node metastasis, Dukes’ stage, and histological differentiation. Higher expression intensity of CEA in CRC was related to lymph node involvement, distant metastasis, and invasion depth. Combining analysis of CD44v6 and CEA can improve the sensitivity and specificity in evaluating the metastatic potential of CRC (P < 0.05). In this study, the effects of capsaicin on the immunohistochemical distribution and gene expression of transforming growth factor alpha (TGF-\alpha) in ovarian tissue of rats at the pubertal age were examined. Materials and methods: Rats (50 days old) were divided into experimental, alimentation, and sham groups. The experimental group was injected with 1 mg/kg doses of capsaicin (10% ethanol and 1% Tween in an 80% distilled water mixture) subcutaneously every day for a period of 1 week. Macroautophagy is a cellular pathway that degrades intracellular pathogens and contributes to antigen presentation. Herpes simplex virus 1 (HSV-1) infection triggers both macroautophagy and an additional form of autophagy that uses the nuclear envelope as a source of membrane. The present study constitutes the first in-depth analysis of nuclear envelope-derived autophagy (NEDA). We established LC3a as a marker that allowed us to distinguish between NEDA and macroautophagy in both immunofluorescence and flow cytometry. Pathological hallmarks of AD include neurofibrillary tau tangles, amyloid beta (Abeta) accumulation and cholinergic degeneration. Cholinergic degeneration can be mimicked in rats by lesioning cholinergic neurons in medial septum. Hippocampal cholinergic denervation disrupts retrograde transport of nerve growth factor (NGF), leading to its accumulation, which subsequently triggers sprouting of noradrenergic sympathetic fibers from the superior cervical ganglia into hippocampus. Dr. McMahon's lab previously reported that coincident with this sprouting, there is an increase in cholinergic innervation that correlates with a recovery of M1 muscarinic receptor dependent plasticity at CA3-CA1 synapses and visual cortex.
