Immunoblot Analysis Of Campylobacters Of Chickens, Mehmet Akan, Jale Erdeğer, Zi̇ya İlhan

The aim of this study was to determine immunogenic structures by immunoblotting in thermophilic Campylobacter strains isolated from chickens. The bands obtained from C. jejuni strains were found to be distributed between 10.8 and 116.5 kD by SDS-PAGE analysis of whole cell proteins. The bands 78, 63, 52, 48.5, 43.5, 29.8, 25.6, 22, 18.5, 17.2 and 16.5 kD were detected as the major common bands in all C. jejuni strains. Electrophoretic analysis of C. coli strains revealed that protein band profiles ranged between 10.3 and 118 kD, and 76, 63-62, 52, 48.5, 44 and 30.5 kD bands were all major …

In this study we examined factors affecting the colonization of campylobacters in chick intestines, colonization times and elicited immune responses. In chicks, infected orally by minimal infective dose _{100} (MID_{100}), colonization in the intestines was observed to reach 100% during the 3-week period following C. jejuni and C. coli inoculation. When the SDS-PAGE protein profiles of the inoculated and isolated strains were examined similarities of 98.3% among C. jejuni strains and 98.5% among C. coli strains were determined. The purpose of this study was to isolate Campylobacter species from the intestines and livers of chicken and to identify Campylobacter coli and Campylobacter jejuni by both conventional methods and Polymerase Chain Reaction (PCR). Four specific primers derived from the ceuE gene present in the genomes of C. coli and C. jejuni were used for PCR identification. In the examination of 150 intestine and liver samples by culture and PCR, 25 (16.6%) and 32 (21.3%) were identified as C. coli and C. jejuni, respectively. In this study, two subjects, 1) whether Visceral Larva Migrans (VLM) takes place when T. canis larvae-infected chick livers are eaten and 2) whether any differences are present between larval distributions, clinical signs and pathological disorders of the infections caused by eggs and infected liver, were investigated in the chick-mouse model. For this purpose, 15-day-old broiler chicks (n=42) and 3-month-old albino mice (n=62) were used. Thirty chicks were each infected orally with 5000 infective T.canis eggs. A total of 251 tracheal swabs and a total of 102 tracheae, lungs, airsacs, livers and spleens were collected from commercial layer, broiler and broiler parent stock flocks with respiratory infection. From turkey flocks, 96 tracheal swabs and 5 tissue samples from the trachea, lung, airsac, liver and spleen were also taken. Cultivations on Blood Agar supplemented with 5-10%sheep blood, on Chocolate Agar and on MacConkey Agar were incubated at 37ºC for 24-72 hours in the presence of 10%CO_2. A trivalent Escherichia coli vaccine (TECA) was prepared from serotypes O1, O2 and O78 and one series of TECA was combined with an inactivated Newcastle Disease vaccine. TECA and combined vaccine were experimentally investigated for immunogenic effects in pullets and hens. This study was carried out at two stages consisting of rearing and laying periods: (1) A number of fourty chicks aged 15 days were subcutaneously vaccinated with 0.2 ml TECA .
