Fetal Whole-Body Fluid, Cardiovascular And Endocrine Dynamics : Computer Analysis Of Regulatory Mechanisms, Peter Stuart Gold

The dynamics of transcapillary fluid and protein distribution and endocrine regulation of cardiovascular function were studied in chronically instrumented fetal sheep during the third trimester. Average experimental changes in vascular pressures, blood volume, plasma hormone concentrations, hematocrit and plasma protein concentration were analyzed using mathematical modeling and parameter optimization techniques in order to determine values for the major parameters which regulate fluid, protein and endocrine dynamics and to determine endocrine dose-response relationships to changes in cardiovascular function. An investigation was undertaken to synthesize and evaluate the cardiovascular activity of two imidazole ring systems. Imidazoles with hypotensive activity would provide novel potential antihypertensive drugs. These novel structures may also find use as chemical probes in the study of the cardiovascular system and as prototypes for other potential cardiovascular agents.

The histamine derivative, 7,7-dipyridyl-4,5,6,7-tetrahydroimidazo-[4,5-c]pyridine (ligand l), was demonstrated to possess significant hypotensive activity in laboratory rats. A 15 mg/Kg dose of ligand 1 produces a statistically significant decrease in blood pressure when injected IP. This hypotensive effect is also associated with marked increase in heart rate. A high percentage of fat in the diet has been implicated through epidemiological studies in several human cancers. Studies in experimental animals have produced evidence that increasing the fat in the diet decreases the animal’s ability to restrict tumor development and growth. In previous work done in this laboratory with BALB/c mice injected with herpes simplex Type 2-transformed mouse cells (H238), the effects of a diet which greatly enhanced tumor growth was compared with one which restricted tumor growth. There is evidence that specific nutrients may modify hormonal balance and contribute to breast cancer etiology. To investigate the relationship between dietary nutrients and plasma reproductive hormones and to determine hormone-nutrient interrelationships, we measured plasma estrogens, androgens. progesterone and prolactin levels in 10 premenopausal Seventh-day Adventist (SDA) vegetarian and 10 premenopausal SDA nonvegetarian women. Over three days in each of three consecutive months. diet records and fasting midluteal blood samples were collected. The nonvegetarians consumed significantly more protein, total and saturated fats, oleic and linoleic acids, and cholesterol than did the vegetarians. Hormonal status of the two groups did not …

An animal tumor model which has been used by many investigators interested in colon cancer is the 1,2-dimethylhydrazine (DMH)-rat or DMH-mouse tumor model. The effect of nine gram positive and grain negative cocci and bacilli, which are indigenous to the oral cavity, on the in vitro growth of Bacteroides gingivalis was examined in this study. A pure culture of B. gingivalis is was obtained from the American Type Culture Collection. This thesis is based on a semiquantitative analysis of four nuclear relay centers [nucleus tractus solitarius (NTS), parabrachial nucleus (PBN), ventroposteromedial nucleus (VPM), gustatory neocortex (GNC)] in the chorda tympani nerve pathway. Metabolic changes were examined by the 2-deoxyglucose method in which one chorda tympani nerve was chemically stimulated and its contralateral side lesioned. Coronal sections of four taste centers were prepared for autoradiography and Nissl staining. Densities of the autoradiograms were determined by means of a video based image processor. Acrosin is an acrosomal enzyme which is reported to be involved in sperm penetration through cumulus, zona, and ooplasma. This enzyme has been reported to be significantly lower in sperm of infertile men compared to sperm of fertile men. The acrosin assay which has been developed can be used for studies in patients with suspected infertility and may be used on a fraction of most semen specimens being evaluated in a clinical andrology laboratory.

The purpose of this study was to investigate the effects of mandibular expansion by orthopaedic and surgical procedures in the growing rat mandible.

Sixty-four 35 day old Sprague Dawley Harlan rats were divided into four groups of 16 rats each and categorized as the control, orthopaedic expansion, surgery without graft and surgery with graft respectively. Prefabricated appliances were used for orthopaedic expansion of the mandibles and maintaining surgical expansion at the symphysis in the respective groups. Lectins are proteins of non-immune origin, which bind to specific carbohydrates (McCoy, 1986). Being that virtually all cell membranes contain carbohydrate components, lectins have been used to identify all types of cells. Alcoholic Hyaline (AH) and keratin intermediate filaments possess ultrastructural, biochemical and immunochemical similarities. This study compares human AH, human stratum corneum keratin (HSCK) and normal human liver cytokeratin (HCK) by quantitative immunochemical means, using polyclonal as well as monoclonal antibodies. Enzyme-linked immunosorbent assays (ELISAs) were employed, as well as "immunoblotting" and immunocytochemical techniques. HCK was isolated using Triton X-100 and "high-salt” buffers and exhibited seven major polypeptide bands on SDS-PAGE (41-58 kd). Antisera were made toward HCK and the two major HCK bands. Continuous administration of estrogens for 7-9 months, both steroidal and nonsteroidal, to male BALB/c mice, leads to the formation of testicular Leydig cell tumors. Three days following the subcutaneous implantation of a pellet of estrogen in cholesterol, there is a peak in the incorporation of ³H-tymidine into the DNA of the interstitial cells. These effects are hypothesized to be mediated by the estrogen receptor system in the Leydig cell. Luteinizing hormone (LH) increases intracellular concentrations of adenosine 3', 5 '-cyclic monophosphate and the phosphoinositides, phosphatidylinositol (PI), PI 4'-phosphate (PIP) and PI 4' .5 1 - bispbosphate (PIP₂). It is believed that cAMP and the phosphoinositides act concertedly to regulate mitochondrial conversion of cholesterol to pregnenolone. The present study investigated the effectiveness of health related feedback as a positive reinforcer of abstinence from smoking during the 11 week period following cessation. Fifty-three subjects were randomly assigned to one of three groups: (a) maintenance program plus health related feedback (MF), (b) maintenance program only (M) and, (c) discussion control (D). All groups received an aversive treatment for smoking after which the MP group received coping skills raining and several forms of health related feedback, the M group received coping skills training only, and the D group received discussion.
