The yeasts Pichia pastoris and Kluyveromyces lactis were used to express several recombinant human proteins for further biochemical characterization. Two substitution variants of recombinant human enteropeptidase light chain (rhEPL) were engineered to modify the extended substrate specificity of this serine protease. Both were secreted as active enzymes in excess of 1.7 mg/L in P. pastoris fermentation broth. The substitution variant rhEPL R96Q showed significantly reduced specificities for the preferred substrate sequences DDDDK and DDDDR; however, the rhEPL Y174R variant displayed improved specificities for these substrate sequences relative to all other reported variants of this enzyme. Pseudomonas aeruginosa, a Gram-negative bacillus, commonly infects immunocompromised individuals and uses a variety of virulence factors to persist in these hosts. The posttranscriptional regulator, RsmA, plays a role in the expression of many virulence factors in P. aeruginosa. RsmA up regulates virulence factors used in colonizing hosts. However, regulation of rsmA is not well elucidated. Transposon mutagenesis was performed on P. aeruginosa containing a transcriptional rsmA-lacZ fusion to answer this question. Mutants were screened via β-galactosidase assay and transposon insertions identified via arbitrary PCR. A probable MFS transporter, we named mtpX, was one significant transposon mutant identified. …

Endometriosis is one of the most common gynecological diseases as it is thought to affect up to 15% of the female population. Characterized by the growth and proliferation of endometrial tissue outside of the uterine cavity, it is a complex condition with varying degrees of severity and can affect multiple regions of the body with symptoms ranging from a total lack of symptoms to debilitating pain and infertility. Mathematical models have been developed to study the transmission of resistant bacteria and the efficacy of preventive measures to slow its spread within a hospital setting. The majority of these models have assumed a constant total hospital population with the admission and discharge rates being equal throughout the duration. But a typical hospital population varies from day to day and season to season. The purpose of this pilot study was to define areas of opportunity to establish a foundation for the implementation of complimentary curriculum between the dietetics and dental hygiene programs at ETSU. While concussions are prevalent in soccer, outside the US there has been limited investigation into soccer players' knowledge and attitudes regarding concussions. The purpose of the study was to access English soccer player' concussion knowledge and attitudes. This study used a cross sectional mixed methodology design. Participants included 26 respondents (Age: 23.4 ± 4.5 years, playing experience 16.4 ± 4.4 years) playing for a Championship team in England (Response rate: 89.6%). Fifty percent of respondents had a diagnosed concussion history. In today's sport, NCAA Division I female athletes are under a constant pressure to reach an elite level of perfection in athletic performance, frequently adhering to the assumption that there is an inverse relationship between body size and performance level (Petrie & Sherman, 2007). Purpose: To examine the effect of three muscle potentiation protocols on changes in muscle architecture and the subsequent effect on jump power performance. Methods: Maximal (1RM) squat strength (Mean SD=178.3 ± 36.6kg), vertical jump power, and muscle architecture were obtained in 12 resistance trained men (25.2±3.6y; 90.67±12.7kg). Participants randomly completed three squatting protocols at 75% (3 x 10 reps), 90% (3 x 3 reps) or 100% (1 x 1) of their 1RM, or no workout (CON), with each protocol being separated by one week. Some 1.6 to 3.8 million sports-related concussions occur annually in the United States. Utilization of test batteries and exercise protocols are recommended to ensure athletes recover completely. Many batteries involve response time (RT) tests, which show response time increases post concussion. A major limitation of RT tests is that all are done in static position. Additionally, many studies show a lingering effect on RT. The addition of RT tests to check for lingering symptoms could be beneficial. The Center for Disease Control and Prevention estimated that there are 1.6-3.8 million sports related traumatic brain injuries in the United States annually which occur in both mainstream and extreme sports. Many of these concussions have been associated with collisional sports, like football, but high risk extreme sports, like motocross, also have many opportunities for collisions to occur.
