Native breeds are excellent sources of genetic variation. Anatolian native breeds are relatives of the first cattle domesticated and are ancestors of many European breeds. Therefore, this study aimed to evaluate the genetic variation of OLR1, ANXA9, MYF5, LTF, IGF1, LGB, CSN3, PIT1, MBL1, CACNA2D1, and ABCG2 markers in Turkish Grey Steppe, Anatolian Black, and East Anatolian Red cattle. The analysis included 367 cattle and the genotyping was performed by the PCR-RFLP. Population genetics indices including heterozygosity, the number of effective alleles, the polymorphism information content, fixation index, and the level of possible variability realization, and moreover, the genetic diversity …

The present study examined the variability of MHC class II DRB exon 2 gene using restriction fragment length polymorphism analysis of PCR-amplified fragments PCR-(RFLP), and its association with Haemonchus contortus infestation in Salem Black and Tellicherry goat population. Animals were naturally exposed to mixed infestation of endoparasites, predominantly Haemonchus contortus. Pooled fecal coproculture and larval identification showed predominant presence of haemonchus (L3) larva. Fecal egg count (FEC) and packed cell volume (PCV) were used as indicator traits. All the three studied loci, TaqI, BsaI, and BsaHI, were polymorphic having two alleles and three genotypes. The loci showed low to moderate …

Water buffalo breeding is practiced in various provinces of Turkey, and is also included in the National Breeding Project. It was aimed in this study to investigate polymorphisms on exon 4?intron 5 and exon 4 only of the kappa-casein (CSN3) gene for milk yields of water buffalo bred in Sivas Province. Blood samples were taken from 135 water buffaloes. The phenol/chloroform method for DNA extraction was conducted on collected blood. Polymerase chain reaction (PCR) was used to amplify CSN3-specific regions, and amplified products were run using 2% agarose gel electrophoresis (AGE). Various restriction endonucleases (RE) were utilized in respect to …

Turkish Journal of Veterinary & Animal Sciences

Litter size is one of the most important economic traits for pig production as it is directly related to production efficiency. To evaluate the effects of the adiponectin (ADIPOQ) gene on porcine litter size, we investigated ADIPOQ polymorphisms in three breeds, Wannan Black pigs, Berkshire pigs, and BW pigs (crossbred pigs produced from mating male Berkshire pigs and female Wannan Black pigs), using the PCR-SSCP method. Three SNPs (c. 178G > A, c. 1165A > G, and c. 1138G > A) were identified. In Wannan Black pigs, association analysis indicated that the c. 178G > A SNP genotype was significantly associated with an effect …

Three pairs of primers were designed to detect single nucleotide polymorphisms (SNPs) of exon 2 of DIO2 in nonseasonal and seasonal reproduction breeds. The polymorphisms were detected using SeqMan II and analyzed by PCR-RFLP. The sheep DIO2 gene was cloned and its mutations were detected in five sheep breeds whose reproductive seasonality and litter sizes were different. Association analyses were performed to reveal the relationships between SNPs and reproductive seasonality as well as litter size in sheep. The results showed that a 1041-bp DNA fragment of the sheep DIO2 …

Turkish Journal of Veterinary & Animal Sciences

Transmissible spongiform encephalopathies (TSEs) are a group of diseases that affect the nervous system and lead to death. The putative infectious agent is the host-encoded prion protein (PrP) and it appears that the development of scrapie in sheep is closely related to polymorphisms in the host PrP gene. The aim of this study was to investigate three well-known polymorphisms in exon 3 of the PrP gene (at codons 136, 154, and 171) that have a clear and significant effect on scrapie in sheep. Four other polymorphisms (at codons 145, 185, 231, and 237) that are yet undefined were also investigated. …
