The purpose of this study was to measure hormonal and metabolic markers in humans to see if simulating dehydration would emulate measures that are seen in prediabetic humans. 60 volunteers were divided into equal categories by sex and body mass index of normal or overweight. Participants completed two oral glucose tolerance tests (OGTT) after being intravenously infused with a isotonic or hypertonic saline solution for 120 minutes in a counter balanced order. Developing a tissue-engineered Blood Vessel Mimic (BVM) to represent diabetic macrovascular disease could expedite design of new vascular devices specifically tailored to diabetic patients. In contribution toward this model, this thesis assessed Human Umbilical Vein Endothelial Cell (HUVEC) responses to high glucose conditions. Interleukin 6 (IL-6) and Cluster of Differentiation 36 (CD36) were selected to signify oxidative stress activity, a hallmark of diabetic macrovascular disease. A chronically elevated vasopressin level has been associated with an impaired glucose regulation system in humans. A recent study in patients with diabetes suggested that low water intake is associated with impaired glucose regulation. The aim of this study was to examine the effect of hypertonic saline infusion on glucose regulation in healthy females. Thirty healthy, non-diabetic females performed a 2-h infusion followed by a 4-h oral glucose tolerance test (OGTT). The purpose of this study is to determine whether different global diets correlate with the increase in type 2 diabetes rates. Type 2 diabetes is the main diabetes type being looked at due to it being the one that can be maintained primarily by glycemic control, which is controlled by a good diet. Five different countries from different regions will be examined: Brazil, China, Micronesia, Nigeria, and the United States. The global prevalence of metabolic syndrome (MetS) and its associated components (high fasting glucose, waist circumference, blood pressure, triglycerides, and low HDL) have increased over the past few decades. Without proper diabetes self-management education, achieving good glycemic control is difficult. Arterial blood pressure (BP) undergoes a 24-hour oscillation that peaks in the active day and reaches a nadir at night during sleep in humans. Reduced nocturnal BP fall (also known as non-dipper) is the most common disruption of BP circadian rhythm and is associated with increased risk of untoward cardiovascular events and target organ injury. Up to 75% of diabetic patients are non-dippers. However, the mechanisms underlying diabetes associated non-dipping BP are largely unknown. Furthermore, the combination of diabetes and depression is associated with increased morbidity and mortality. Worldwide, the prevalence of cardiometabolic diseases, particularly type II diabetes mellitus (T2DM), and to a lesser extent, metabolic syndrome (MetS), has increased dramatically. Despite this increase, there is still a lack of robust biomarkers for cardiometabolic diseases to secure better clinical outcomes.
