Purpose: This thesis attempted to quantitatively analyze the individual cell fate choice in resistant head and neck UM-SCC38 cells exposed to cisplatin using the most current techniques available.

Methods: UM-SCC-38 cells were grown in Dulbecco’s modified Eagle’s medium (DMEM) supplemented with 10% fetal bovine serum (FBS). They were treated with cisplatin and ATM/ATR inhibitors of known dosages. Using live cell imaging, one hundred cells were tracked in each experiment and their behaviors were analyzed and entered into Microsoft Excel Spreadsheet to generate cell profile graphs. Objective: The aim of this study was to assess outcomes related to general quality of life, daytime sleepiness and functional sleep outcomes, functional outcomes of orthognathic surgery, and facial aesthetics in patients undergoing maxillomandibular advancement (MMA) surgery for the treatment of obstructive sleep apnea (OSA).

Materials and Methods: This was a cross-sectional self-report study. A questionnaire was constructed using questions drawn from previously validated questionnaires. The survey was distributed to 25 patients who underwent MMA surgery for the treatment of OSA at LHSC in London, Ontario by a single surgeon between 2002 and 2013.

Bone development and repair involve complex processes that include interaction between cells and their surrounding matrix. In the body, bone sialoprotein (BSP) expression is up-regulated at the onset of mineralization. BSP is a multifunctional acidic phosphoprotein with collagen-binding, hydroxyapatite nucleating, and integrin recognition (RGD sequence, which is important for cell-attachment and signaling) regions. Mice lacking BSP expression (Bsp-/-), exhibit a bone phenotype with reductions in bone mineral density, bone length, osteoclast activation, and impaired bone healing. Objective: The objective of this study was to determine the effect of pulsating vibration on the sliding resistance between orthodontic brackets and stainless steel wires. D2 dental students (n=25) were recruited and instructed on the use of 3M LAVA and PVS techniques using three pedagogic methods: 1) video lecture (P1), 2) investigator-led demonstration (P2), and 3) independent clinical exercise (P3). Periodontal disease is an oral inflammatory disorder that afflicts roughly 46% of the adults in the U.S. Currently, treatment of periodontal disease involves the removal of plaque from the gingival pocket (with possible antibiotic treatment) and if necessary, gingival surgery. To our knowledge, no therapeutic approach exists that promotes host-biofilm homeostasis by limiting pathogen recolonization of the oral cavity after prophylaxis or treatment. Although we know that children covered by Medicaid often do not receive needed dental services, the extent of the problem in Maine is not fully understood. In particular, rural communities in Maine may face challenges for families to access dental services for their children given low reimbursement rates and fewer available providers. This project addresses these gaps in knowledge by examining MaineCare- funded utilization and dental care access for children with MaineCare in four rural Maine towns in the Lakes Region.

The goal of this study is to use the improved imaging capability of cone-beam computerized tomography (CBCT) to investigate the relationship between vertical facial patterns and mandibular tooth-alveolar morphology in the adolescent population. Pre-treatment orthodontic records were obtained from the UNLV School of Dental Medicine archival dental records. One hundred and seventy three patients (72 males, 101 females) between the ages of 12 and 18 years were included in this study. Orthodontic tooth movement involves activation of osteoblasts and osteoclasts in response to mechanical forces. Extracellular nucleotides, released in response to mechanical stimuli, signal through P2 receptors in osteoblasts. P2X7 receptors are ATP-gated cation channels implicated in mechanotransduction in bone. Corticosteroids such as dexamethasone exhibit anti-inflammatory effects and their chronic use has been linked to serious side-effects such as osteoporosis. We investigated gene expression following P2X7 receptor activation with benzoylbenzoyl-ATP (P2X7 receptor agonist) in osteoblast precursor cells in the presence or absence of dexamethasone. For proper orthodontic diagnosis and treatment planning, an accurate measurement of the relationship of the maxilla to the mandible in the sagittal plane is required. The ANB measurement has long been the gold standard for explaining this association. The purpose of this longitudinal study was to evaluate three linear measurements of the maxillomandibular anteroposterior relationship and determine which one best correlates with ANB. The constructed Frankfort horizontal-mandibular plane angle bisector (cFMAB-Wits), maxillomandibular bisector (MMB-Wits), and Frankfort-mandibular plane angle bisector (FMAB-Wits) were measured using a Wits’-type analysis and compared to ANB.

Objective: The purpose of this ex vivo study was to assess the changes in tooth color and existing orthodontic bracket bonding when exposed to a bleaching agent. Methods: 120 freshly extracted human premolars were equally divided into 4 groups: a negative-control group, a positive-control bleaching group, a bracket-bonded group, and a bracket bonded-bleached group. Purpose: The purpose of this study was to determine if visible tattoos affected the perceptions of dental consumers toward the professionalism and credibility of dental hygienists.

Methods: An 11 item, IRB approved investigator designed survey was administered online to 203 subjects invited to participate via a commercial web source. Surveys were randomized according to respondents' birth month. Current drug therapies have lost their effectiveness in controlling the fungal species known as C. albicans; therefore, the need for newer drugs becomes crucial. Histatin 5 (Hst 5), a naturally occurring peptide found in the oral cavity, has demonstrated strong effects on controlling candidiasis. Here, we have proposed that the adhesion of cells through surface proteins is an important process. To allow the analysis of surface proteins, the concept of on-whole cell tryptic digestions were developed. Aim: The purpose of this study was to determine the prevalence of orthodontic extraction cases and the rationale for the decision-making. Materials & Methods: The records of consecutive patients who had completed orthodontic treatment at Graduate Orthodontic Clinic, Loma Linda University School of Dentistry, between January 2010 and December 2014 were reviewed for prevalence of orthodontic extractions. Cone beam computed tomograms (CBCTs) of patients, that had extraction(s) prescribed as part of orthodontic treatment, were analyzed dentally and skeletally. Tooth Length Measurements On 3t Mr Images: A Retrospective Study, Kevin G. Murray

Introduction: The purpose of this study was to determine if MRI can be used to accurately measure tooth lengths. Methods: MRI tooth length measurements were compared with “actual tooth lengths” as measured on CBCT scans. Twenty three subjects received two scans (one CBCT and one MRI). Tooth length was measured and compared between the resultant images. Intraclass correlations were used for statistical analysis. Results: Tooth length measurements made on MRI scans showed moderate to almost perfect agreement with tooth length measurements made on CBCT scans. Higher levels of agreement were present in the maxillary arch compared to the mandibular arch. …
