Ovarian cancer (OC) is one of the most common, yet deadly, cancers women face. There are multiple ways it can be treated, such as hysterectomy, cytoreductive surgery, and/or chemotherapy, depending on the grade of the cancer. Important risk factors to note are genetics, talc usage, endometriosis, obesity, gene mutations, and cigarette smoking. Non-oral, long-acting contraception (LARC), such as IUDs (intrauterine device) and implants, are the most effective methods of pregnancy prevention. It is uncertain that they can protect against OC like oral contraceptives can. If LARCs can be proven useful in preventing OC, the mortality rate of OC will decrease, …

The role of natural products in drug development is well established. In recent years, marine cyanobacteria have been regarded as a major source of biologically active metabolites with chemical and Figure 5 pharmacological diversity. These cyanobacterial natural products serve as a promising source of drug A. leads for the discovery of therapeutic agents used in the treatment of many diseases of interest, such as CNS disorders, pain, and cancer. Acute Respiratory distress Syndrome (ARDS) and Acute Lung Injury (ALI) are inflammatory lung pathologies consisting of non-hydrostatic pulmonary edema leading to hypoxia and impaired gas exchange in the lungs. ARDS/ALI is both difficult to study and treat as it is not in itself a specific pathology but rather a syndrome consisting of many pathologies that vary case by case. It is, however, consistently characterized by an explosive acute inflammatory response in the lung parenchyma leading to hypoxia. Insects have the ability to reflect the chemical environment that they have developed in. Thus, necrogenous insects can reflect the chemical environment of cadavers they have fed on. Forensic scientists can potentially utilize insect larvae as an alternate biological matrix to detect drugs in cadavers that are skeletonized, burned, or otherwise badly damaged. Analysis of insects using paper spray ionization mass spectrometry (PSI-MS), an analytical method requiring little to no sample preparation, could prove a rapid, cost-effective, and non-destructive alternative form of toxicological analysis. This thesis mainly focuses on the introduction of the background and work have been done in the areas of antifolates development, such as folate function, its three uptake mechanisms inside human cells, antifolates’ role in chemotherapy, et. al. In addition, the Structure-Activity-Relationship design rationale for the series of antifolates will also be discussed. Nevertheless, the details of synthesizing these pyrrolo[2,3-d]pyrimidines as potential antifolates have been described, including chemistry reviews on the pyrrolo[2,3-d]pyrimidine scaffold, and the challenges encountered and the solutions how to solve or improve in order to achieve better yield.

Obsessive compulsive disorder (OCD) is a psychiatric disorder characterized by recurrent intrusive thoughts (obsessions) that cause significant anxiety and distress, leading to repetitive behaviors (compulsions) that an individual feels driven to perform. The symptoms of OCD can range from mild to so severe that it can be incapacitating to an individual’s life. Treatment is often prescribed, including both pharmacological and behavioral therapy. Overall, 70% of patients starting treatment experience a significant improvement; however, there is still a portion of patients with severe OCD that do not respond to first and second-line treatment plans. This dissertation describes the design, synthesis and biological evaluation of monocyclic and bicyclic pyrimidine-base heterocycles as single agents with combination chemotherapy potential having both antiangiogenic effects and cytotoxic effects. This dissertation also describes selective tumor targeting with 5-substituted pyrrolo[2,3-d]pyrimidines analogs with heteroatom bridge substitution as GARFTase inhibitors.

The work in this dissertation is centered on identifying structural features that are necessary for inhibition of tubulin polymerization as well as for inhibition of one or more of the receptor tyrosine kinases (RTKs)- vascular endothelial growth factor receptor-2 (VEGFR2), platelet derived growth factor receptor-β (PDGFRβ) and epidermal growth factor receptor …
