Crippling Rapid Evolution Of Metastasis And Drug Resistance In A549 Non-Small Cell Lung Cancer Cells With The Clinically Relevant Hsp90 Inhibitor Auy922, Nickolas Anthony Bacon Jan 2021

Crippling Rapid Evolution Of Metastasis And Drug Resistance In A549 Non-Small Cell Lung Cancer Cells With The Clinically Relevant Hsp90 Inhibitor Auy922, Nickolas Anthony Bacon

Theses, Dissertations and Capstones

The ability for species to evolve new features in response to changing circumstances in order to survive and propagate is a ubiquitous observation on both the macroscopic and microscopic levels of living systems. It should be no surprise, then, that diseases such as cancer utilize their own forms of adaptation to perpetuate themselves when exposed to external threats. Indeed, concepts drawn from Darwinian evolution are now widely accepted to help explain certain aspects of carcinogenesis and malignant progression, the sum of which have come to be known as the theory of tumor evolution. Since metastasis and drug resistance are features …

Effects Of Aging And Gender On Regulators Of Muscle Adaptation In F344/Bn Rat Model, Satyanarayana Paturi Jan 2008

Effects Of Aging And Gender On Regulators Of Muscle Adaptation In F344/Bn Rat Model, Satyanarayana Paturi

Theses, Dissertations and Capstones

Sarcopenia is the loss of muscle mass and strength that occurs with aging. Here we examine the effects of aging and gender on the regulation of molecules believed to regulate muscle growth and adaptation in the F344/BN rat. In male animals, soleus and EDL muscle/body weight ratio declined continuously with aging while muscle atrophy in female animals plateaued at 26-months and remained constant thereafter. Aging increased the phosphorylation of protein kinase-B (Akt) and the mammalian target of rapamycin (mTOR) in the female but not male soleus muscle. This finding was associated with the attenuation of muscle atrophy observed in female …

Chronic Acetaminophen Treatment Influences Indices Of Reactive Oxygen Species Accumulation In The Aging Fisher 344 X Brown Norway Rat Aorta, Sarath Meduru Jan 2007

Chronic Acetaminophen Treatment Influences Indices Of Reactive Oxygen Species Accumulation In The Aging Fisher 344 X Brown Norway Rat Aorta, Sarath Meduru

Theses, Dissertations and Capstones

We have previously reported that aging in the Fisher 344 X Brown Norway (FBN) rat aorta is characterized by increased levels of ROS and alterations in cell signaling. Acetaminophen was found to scavenge free radicals in recent ischemia-reperfusion studies. Here we examined if chronic treatment with a therapeutic dose of acetaminophen attenuates age-associated increase in aortic ROS accumulation and signaling. FBN rats (27 month old; n=8) were subjected to 6 months of treatment with a therapeutic dose of acetaminophen (30mg/kg/day), with age-matched untreated FBN rats as controls. Protein oxidation levels were altered in control and treated aortae compared to aortae …

Whole Animal And Cellular Strategies In The Regulation Of Bone Mass In The Rat, Weidong Geng Jan 2000

Whole Animal And Cellular Strategies In The Regulation Of Bone Mass In The Rat, Weidong Geng

Theses, Dissertations and Capstones

In order to determine the strategies in regulation of bone mass in the rat and better understand the pathophysiological mechanism of the postmenopausal osteoporosis, a series of studies were conducted at the whole animal and cellular level. At the whole animal level, female Sprague-Dawley rats were ovariectomized (Ovx) or sham-operated (control) at 18 weeks and the entire skeleton obtained at 24 weeks (baseline) or after an additional 31 day (28 week) interval on a normal (1.0%) or deficient (0.02%) calcium diet. Ovx rats on the normal calcium diet showed a 42% increase in whole body bone resorption and a 70% …

Differences In Lean And Obese Skeletal Muscle Myoblasts Of Adult And Post-Natal La/N (Faf) Rats: A Confocal Description Of The Skeletal Muscle Myoblast, J. Adam Noah Jan 1998

Differences In Lean And Obese Skeletal Muscle Myoblasts Of Adult And Post-Natal La/N (Faf) Rats: A Confocal Description Of The Skeletal Muscle Myoblast, J. Adam Noah

Theses, Dissertations and Capstones

Mutant strains of obese rats are currently being studied to reveal the mechanisms of increased lipid synthesis and decreased lipid oxidation in these rats. It was originally thought that much of the fat production that leads to obesity is the result of metabolic processes in the liver and adipose tissue. Studies by Kahle et al.3 showed that the liver and adipose tissue account for only 22% and 7% respectively, of total fatty acid synthesis. It was also shown that 21 % of total de novo fatty acid was localized in the postural skeletal muscle.

The dry weight of the …

Relationship Of Resting And Submaximal Cardiovascular Measures To Vo[2]Max In Untrained Children, Thomas A. Cappaert Jan 1997

Relationship Of Resting And Submaximal Cardiovascular Measures To Vo[2]Max In Untrained Children, Thomas A. Cappaert

Theses, Dissertations and Capstones

Participation by children in organized sport continues to increase. A 1994 survey of 10,000 households by the National Sporting Goods Association (1994) found that approximately 40,000 children between the ages of 7-17 years participate in an organized sport or recreational activity annually. In the realm of organized sports, especially endurance activities, there continues to be interest in how cardiovascular fitness is attained and how it can be maximized. Measurement of cardiovascular fitness is an important piece to that puzzle.

The most common and most descriptive measure for children and adults, is the maximal oxygen uptake or max (Astrand & Rodahl, …

Use Of The (3)H-Tetracycline Rat Model In The Study Of Skeletal Compartmentalization And Metabolism Of Calcium In The Maturing Male And Female Rat And The Role Of The Skeleton In Calcium Homeostasis In The Adolescent Male Rat, Darrin Lee Demoss Jan 1996

Use Of The (3)H-Tetracycline Rat Model In The Study Of Skeletal Compartmentalization And Metabolism Of Calcium In The Maturing Male And Female Rat And The Role Of The Skeleton In Calcium Homeostasis In The Adolescent Male Rat, Darrin Lee Demoss

Theses, Dissertations and Capstones

The (3)H-tetracycline bone labeling procedure was employed to monitor bone resorption from urinary loss curves in male and female rats of various ages. In addition, whole body dry skeletal mass and the loss of (3)H-tetracycline from individual bones was determined. It was found that the dry skeletal mass/body mass ratio of 24-week-old females was 30-40% greater than that for males. The urinary loss of (3)H-tetracycline was described by a double exponential equation indicating the presence of two distinct and independent exchangeable bone compartments. Both compartments decrease in size with age, but their label loss activities were different. The label loss …
