Cadmium (Cd) is frequently used in various industrial applications and is a ubiquitous environmental toxicant, also present in tobacco smoke. An important route of exposure is the circulatory system whereas blood vessels are considered to be main stream organs of Cd toxicity. Our previous results indicate that cadmium chloride (CdCl2) affects mean arterial blood pressure in hypertensive rats. We hypothesized that Cd alters the intracellular calcium transient mechanism, by cadmium-induced stimulation of MAPKs (ERK 1 & 2) which is mediated partially through calcium-dependent PKC mechanism. Microbial content of wastewater in two abattoirs and the impact on microbial population of receiving soil was studied in Agege and Ojo Local Government Areas in Lagos State, Nigeria. Wastewater samples were collected from each of the abattoirs over three months period and examined for microbial content. Soil samples contaminated with the wastewaters were also collected and analyzed for microbial content as compared to soil without wastewater contamination in the neighborhood (control). Some physico-chemical parameters of the samples such as total dissolved solid, chemical oxygen demand, etc., were examined. One such system, the nasal delivery system, shows good prospect for effective delivery of some drugs, however, it is yet to be well established for others. The mechanism(s) by which arsenic exposure contributes to human cancer risk is unknown; however, several indirect cocarcinogenesis mechanisms have been proposed. Many studies support the role of As in altering one or more DNA repair processes. In the present study we used individual-level exposure data and biologic samples to investigate the effects of As exposure on nucleotide excision repair in two study populations, focusing on the excision repair cross-complement 1 (ERCC1) component. Surveys of DSP (Diarrheic Shellfish Poisoning) toxin profiles in the mussel Mytilus galloprovincialis from coastal waters of the central Adriatic Sea, over the years 1995 to 2001, demonstrate that incidents of shellfish toxicity in this area are dominated by the occurrence of okadaic acid (OA) and a PTX derivative, 7-epi-pectenotoxin-2-seco acid (7-epi-PTX-2SA). Toxin composition and the relative ratio of toxic components in shellfish is in correlation with the occurrence of DSP producing organisms from the Dinophysis species along with Prorocentrum micans and Lingulodinium polyedrum. Previous Spatial Memory Training And Nicotine Administration Alleviates Cognitive Deficits Produced By Medial Frontal Cortex Lesions In Rats., Rachel L. Norris

Rats were administered nicotine (0.3 mg/kg) for 11 consecutive days before and after an electrolytic medial frontal cortex lesion. Behavioral testing was arranged so that the rats were tested on the RAM 1 day after drug administration followed by behavioral testing on the MWT 19 days after drug treatment, or tested on the MWT 1 day after drug administration followed by testing on the RAM4 days after drug treatment. Results of MWT testing showed that regardless of the drug/behavioral testing interval, lesioned rats given nicotine demonstrated enhancement relative to saline-treated animals. The major histocompatibility complex (MHC) is a gene family responsible for many critical functions of the immune system in most vertebrates. The MHC consists of three classes differentiated by their structure and function, and MHC class I encodes antigen binding proteins as well as chaperone and accessory proteins such as tapasin. The purpose of this project is to reconstitute several human MHC class I molecules in their peptide-filled and peptide-deficient forms, and to purify these proteins for biochemical study. The expressed proteins include wild type and mutant variants of the fusion protein human leukocyte antigen HLA-B*0801-fos, and human beta-2-microglobulin (β2m). …

Cigarette smoke contains sufficient carbon monoxide to induce maternal and fetal hypoxia. Hypoxia is a known teratogen, and consequently maternal smoking has been the focus of many studies on adverse birth outcomes, including cleft lip and palate. Current literature of epidemiological studies on smoking and clefts suggests a modest but statistically significant increase in risk of clefting associated with maternal smoking. A biological condition that may also contribute to hypoxia is anemia. This study examines airborne chemical exposure to Air Force small arms range instructors during M16 firing of lead and lead-free bullets. Historical range information collected from 63 active duty Air Force bases identified that two thirds of the Air Force military ranges within the Continental United States are currently firing lead-free ammunition. Average 8-hr TWA exposures for lead during M16 firing of leaded ammunition were 17% of the Occupational Exposure Limit (OEL). Task exposures, representing worst case conditions, were found to be 1.2 time the OEL. Two indoor and four outdoor firing ranges currently firing frangible lead-free ammunition were evaluated …

The persistence of polycyclic aromatic hydrocarbons (PAHs) in marine sediments may be influenced by benthic invertebrate bioturbation. Through processes such as deposit-feeding and enhancement of microbial metabolic activity PAHs may be remobilized from the sediment compartment, and either transferred to organisms at higher trophic levels or to the overlying water column, both processes inevitably changing the bioavailability of the PAH. Accumulation of contaminants from one level in the food chain to the next depends on feeding rate and assimilation efficiency, two factors that basically vary with food quality and contaminant type. The present experiments are part of a larger study designed to investigate the influence of husbandry parameters on the life history of the ramshorn snail, Marisa cornuarietis, in order to identify suitable husbandry conditions for maintaining multi-generation populations in the laboratory for use in ecotoxicological testing. The present study was conducted to determine the half-life, assess the toxicity, and passive protection efficacy of purified immunoglobulin G (IgG) from Anthrax Vaccine Adsorbed (AVA) vaccinated human donors. Half-life determinations were calculated from the reportable values obtained using the anti-PA ELISA assay and the Centers for Disease Control's (CDC) "ELISA for Windows" software. For toxicity evaluations animals were observed clinical for one hour post administration and for 14-days post-treatment. The protection efficacy was determined based upon the mortality results from a lethal Bacillus anthracis aerosol challenge. While no protection was achieved in this delayed exposure scenario, the study yielded …

Chemical form (i.e., species) can influence metal toxicokinetics and toxicodynamics and should be considered to improve human health risk assessment. Much evidence now exists regarding radiation-induced bystander effects, but the mechanisms involved in the transduction of the signal are still unclear. The mitogen-activated protein kinase (MAPK) pathways have been linked to growth factor-mediated regulation of cellular events such as proliferation, senescence, differentiation and apoptosis. Activation of multiple MAPK pathways such as the ERK, JNK and p38 pathways have been shown to occur after exposure of cells to radiation and a variety of other toxic stresses. Impaired vascular reactivity is a hallmark of several cardiovascular diseases that include hypertension and diabetes. This study compared the changes in vascular reactivity in age-matched experimental hypertension and diabetes, and, subsequently, tested whether these changes could be affected directly by ascorbic acid (10 microM). Endothelium-derived nitric oxide (NO) modulation of ascorbic acid effects was also investigated. All the experiments were performed in the presence of a cyclooxygenase inhibitor, indomethacin (10 microM). Results showed that the endothelium-dependent and -independent relaxations induced by acetylcholine (ACh) and sodium nitroprusside (SNP), respectively, were blunted to a similar extent in isolated aortic rings from age-matched …

Bioassay-guided fractionation was performed on a crude dichloromethane extract of Kaempferia galanga L. using chromatography techniques. Screening of the extract for biological activity started with the brine shrimp lethality bioassay, followed by the study of its antihypertensive activity on anaesthetized rats, which involved monitoring of the extract's effect on mean arterial blood pressure. The components of the fractions obtained from the separation procedures were analyzed using gas chromatography (GC). The yield of the CH2Cl2 extract was 0.29% of the crude plant extract. During 2002 and 2003 samples, collected every five miles along the Ohio River, were analyzed for total cultivable bacteria, ciprofloxacin resistant bacteria, erythromycin resistant bacteria, tetracycline resistant bacteria, and fecal coliforms. During 2004 samples were analyzed for total cultivable bacteria, ciprofloxacin resistant bacteria, sulfamethizole resistant bacteria, tetracycline resistant bacteria, Virginiamycin resistant bacteria, total coliforms, and Escherichia coli. The present study was conducted to assess the toxicity of nanosized silver particles (Ag-15nm, 30nm and 55nm) in rat alveolar macrophages. For toxicity evaluations, cellular morphology, mitochondrial function (MTT assay), membrane leakage (LDH assay) and reactive oxygen species generation (ROS) were assessed after a 24h under control and exposed conditions. The morphological appearance of control and exposed cells were observed by an inverted, phase contrast microscope and the uptake of nanoparticles was observed using the CytoViva Ultra Resolution Imaging (URI) system.
