The changing character of Maine’s coastal ecosystems has accelerated in full view of the users most reliant on nearshore habitats and the species they support. Adaptation and diversification facilitate continued harvest of Maine’s iconic ocean bounty, which supports the coastal economy despite compounding stresses from upland development, warmer and more acidic waters, and invasive species. Use intensification in the nearshore environment calls for coordinated discussions to nurture and innovate within and across habitats to preserve their longevity and sustain the people that depend on them. Cardiovascular disease (CVD) is a leading cause of mortality globally. Metabolic diseases, such as obesity, are major risk factors for CVD development. Obesity changes adipose tissue physiology, thereby contributing to the establishment and progression of CVD. The perivascular adipose tissue (PVAT) is a unique depot that surrounds the vasculature. During obesity, PVAT secretes factors that induce vascular inflammation and contraction. Recent work has indicated a role for adipose-derived exosomes in regulating disease pathology. However, very little is known about the importance of PVAT-derived exosomes in modulating vascular health. Primary Cilia Of The Cardiac Neural Crest & Hedgehog-Mediated Mechanisms Of Congenital Heart Disease, Lindsey A. Fitzsimons

Elimination of primary cilia in cardiac neural crest cell (CNCC) progenitors is hypothesized to cause a variety of congenital heart defects (CHDs), including atrioventricular septal defects, and malformations of the developing cardiac outflow tract. We present an in vivo model of CHD resulting from the conditional elimination of primary cilia from CNCC using multiple, Wnt1:Cre-loxP, neural crest-specific systems, targeting two distinctive, but critical, primary cilia structural genes: Intraflagellar transport protein 88 (Ift88) or kinesin family member 3A (Kif3a). CNCC loss of primary cilia leads to widespread CHD, where homozygous mutant embryos (MUT) display a variety of outflow tract malformations, septation …

Renal cell carcinoma (RCC) is the 8th most common cancer in the United States, with the clear cell variant (ccRCC) being the most prevalent. Over 14,000 people die every year to RCC, with rates continuing to increase with an aging general population. Patients suffering from metastatic RCC (mRCC) have extremely poor prognoses, with a 5-year survival of only 11.2%. Current treatment options include resection of primary lesions, tyrosine kinase inhibition (Sunitinib, Pazopanib), mTOR inhibition (Temsirolimus, Everolimus), and immune checkpoint inhibition (Nivolumab, Atezolizumab). With the growing attention on widespread plastic usage and its impact on the environment and human health, the need for sustainable alternatives to petroleum-based plastics is more important than ever. One of the most promising solutions is bioplastics; however, current bioplastics struggle to compete with the material properties of petroleum- based plastics. Agar is a sustainable algae-derived hydrocolloid polysaccharide that can be used for bioplastics and biofilms. Despite promising characteristics, bioplastics made from agar are brittle. Dry eye can result from multiple causes, including a reduction in the quantity or altered composition of tears. The lacrimal gland is the major supplier of the aqueous tear components, including water, electrolytes, and proteins. Reduced output from the lacrimal glands results in aqueous tear deficiency, a specific subclass of dry eye disease (DED) marked by inadequate tear volume. The prevalence of DED is 1.5-2 times greater in women than men, yet a comprehensive comparison of dry eye symptoms in male and female animals in an animal model of DED has not been performed. Dickeya dianthicola (Samson) causing blackleg and soft rot was first detected in potatoes grown in Maine in 2014. Previous work has suggested that insects, particularly aphids, may be able to vector bacteria in this genus between plants, but no conclusive work has been done to confirm this theory. In order to determine whether insect-mediated transmission is likely to occur in potato fields, two model potato pests common in Maine were used: the Colorado potato beetle (Leptinotarsa decimlineata Say) and the green peach aphids (Myzus persicae Sulzer). Commercial production of lowbush blueberry (Vaccinium angustifolium Aiton) in Maine relies primarily on managed honeybee hives; however, naturally occurring wild bees are more efficient pollinators of the crop. Wild bees have short foraging distances and must nest near crop fields to provide pollination services. After crop bloom, the surrounding landscape must provide sufficient forage to maintain wild bee populations for the remainder of the growing season. Lowbush blueberries in Maine are produced in a mixed-use landscape with two distinct landscape contexts. Current repositories for protein-protein interactions and high throughput screening methods focus on individual gene products and do not consider the significance of calcium induced conformational changes. These limitations suggest the need for alternative strategies to better define the calpain2 interactome. Affinity capture coupled with LC-MS/MS and proteomic analysis of the recovered proteins provides a powerful approach to identify protein-protein interactions for the heterodimeric calpain2. CAPN2 (rat) was modified to be catalytically incompetent (C105A) and fused with a C-terminal 15 residue peptide optimized for biotinylation by the biotin protein ligase, BirA. The study of contemporary evolution and eco-evolutionary dynamics is classically defined in terms of genetic evolution, but the actual suite of processes driving contemporary trait change is likely much more complex than often credited. This dissertation considers additional mechanisms of trait change that might be important to an emerging model system for study of contemporary evolution and eco-evolutionary dynamics. Specifically, the research focuses on phenotypically plastic and demographic trait variation in Eastern and Western Mosquitofish (Gambusia affinis and G. holbrooki) facing the major ecological gradient of predation risk. The acreage of pollinator-dependent crops continues to expand across the globe. Simultaneously, honey bee hives – an annually rented commodity that growers rely on – are more expensive every year and in some cases, scarce. In response, pollinator-dependent growers seek alternative pollinators. One approach is installing bee pasture on farms, a strategy that enables systems-based farmers to become in-situ farm-scale habitat managers.
