X-Ray Microanalysis In Cryosections Of Natively Frozen Paramecium Caudatum With Regard To Ion Distribution In Ciliates, M. Schmitz, R. Meyer, K. Zierold

Scanning Electron Microscopy

Cells of Paramecium caudatum were shock-frozen without pretreatment for cryoultramicrotomy and freeze-dried for subsequent X-ray microanalysis. Na, Mg, P, S, Cl, K, and Ca were detected in different amounts in several subcellular compartments. In particular, calcium was localized below the cell surface (pellicle). Trichocysts were found to contain significant amounts of Na in their base but not in the tip. Na, Mg, P, S, Cl, K, Ca were found in electron dense deposits within the lumen of the contractile vacuole. A small K concentration was found in the cytoplasm and in the mitochondria. The literature on scanning electron microscopy (SEM) pertaining to the retina has been surveyed and described.

The first two papers on SEM and retina appeared in 1969. Most of the earlier studies concentrated on descriptions [by SEM alone, or with light microscopy (LM) and transmission electron microscopy (TEM)] of the appearance of various retinae and retinal cells [fish, newt, primates, rodents and rabbits, bullfrog]; or embryology of the chick retina. Earlier studies in the performance analysis of concurrency control algorithms in distributed database systems (DDBS) have assumed that the message transmission time between any two nodes of a network is constant. Hence they disregard the effect of communication subnet related parameters such as network traffic, network topology, the capacity of transmission channels and the size of messages transmitted. In this paper an analytical model is used to estimate the delays in transmission channels for a DDBS in a long haul network. A Rapid Technique For Counting Cracks In Rocks, W. B. Durham, J. M. Beiriger, H. C. Weed

Scanning Electron Microscopy

Using a scanning electron microscope (SEM) and an image analyzer, we have developed a technique for counting and measuring cracks in rocks which is more efficient than traditional techniques in which an operator performs all image analysis functions. The key aspect of the technique is that black-on-white tracings of fresh cracks, which can be made rather rapidly by an operator, are measured and digitized by an image analyzer. The most time-consuming step in the process has now become the generation of SEM micrographs and pertinent chemical (mineralogical) information, not the quantification of crack structure. The purpose of this study was to determine whether defoliation, a condition of stress caused by Choristoneura fumiferana herbivory, induces a disturbed pathologic condition in presumably normal tissue produced by the affected balsam fir trees.

Techniques used in this investigation included dry weight determination, light microscopy, scanning electron microscopy (SEM), transmission electron microscopy (TEM), and biochemical and physical analyses.

Terminal branch sections that contained shoots with buds and needles were cut from the upper crown on opposite sides of the tree.

The study and isolation of strains of thermophilic anaerobic bacteria, Clostridium thermocellum. has been widely committed. The isolation of six strains of Clostridium thermocellum from samples representing a variety of environments were conducted. They were characterized as to growth rate, production of reducing sugars, ethanol and acetaic acid production from cellulose, base composition of DNA and the presence of plasmid DNA.

Results show that considerable differences exist among strains with regard to growth rate and ability to solubilize cellulose. However, they were all remarkably similar in ethanol production which was similar to a standard laboratory strain of these species. The ...

On 6 July 1984, at 6 PM CDT, I saw a Common Moorhen at the Capitol Beach marsh, in Lincoln. It was about 10 yards to the north when I first saw it, and I could see the red facial shield and red bill with yellow tip, the yellow legs, a thin white horizontal line along the side, and prominent white patches along either side of dark undertail coverts. An Exceptional "Fall" Migration Of Shorebirds Along The Big Bend Reach Of The Platte River, Gary R. Lingle, Thomas E. Labedz

The Big Bend reach of the Platte River extends about 80 miles from Overton to Chapman, Nebraska. This area is characterized by braided channels with numerous riverine islands bordered by cottonwoods (Populus deltoides) and willows (Salix spp.) along the high banks. Habitat between the high banks consists of open water, barren sandbars and mudflats, herbaceous islands, shrub islands, and floodplain forest.

Prolonged summer flows during 1983 were among the highest recorded this century; in excess of 20,000 cfs (cubic feet per second). Spring 1984 also consisted of high flows resulting in much scouring and shifting of alluvial sediments. By ...

The 1984 Fall Field Day was held (officially) from noon to noon, 6/7 October, at the4-H Camp at Halsey National Forest. The weather was not the best—light rain early Saturday morning, only a couple of short showers in the afternoon, and no rain Sunday, both days dull and gray. But the attendance was good—87 were officially registered—but the added manpower could not make up for the lateness in the season and the poor weather—the tally on or near the Forest was only 81. We evaluated the effects of screening for multiple endocrine neoplasia type 2A (MEN-2A) in 12 families. Genealogical studies going back to 1730 show a common ancestry for seven Swedish families and one American family. The total number of patients included 105 individuals, 68 of whom were diagnosed by our screening program. Our screening methods for medullary carcinoma of the thyroid (MTC) had an optimal sensitivity and specificity. The frequency of gene carriers detected in MEN-2A families was 55%. This project concentrated on the development and subsequent results of an integrated deinking system. Although the chemistry and basic principles are quite different, a few European companies are successfully combining the two·systems by taking advantage of the differences.

Advantages of an integrated system over the conventional type included higher brightness, lower ash content and decreased sensitivity to particle size.

The experiment consisted of two different two-stage deinking systems. Each system was run twice; once with a sodium-precipitate chemistry and once with a calcium precipitate chemistry.

Paper made from TMP fibers offers good opacity and stiffness, but its use is limited because of its low strength. Chemical post-treatment of TMP offers possibilities of improving strength. Sulphonation of lignin appears to be the most economically feasible treatment. This thesis was oriented toward finding out if sulphonation of TMP fibers would improve the strength properties of TMP handsheets. Southern pine TMP fibers were treated with sodium sulfite (Na2SO3). Refiner peroxide bleaching trials were carried out at the Abitibi-Price Research pilot plant using a Sunds-Defibrator CD300 pilot-scale refiner. Results show that, at the pilot scale, blowline addition of bleach liquor gives better brightness gains than addition in the refining zone. Using this technique, brightness gains equivalent to tower bleaching can be obtained when the bleaching parameters are properly controlled. The most important factors were post-refiner retention time and temperature, and the alkali charge in the bleach liquor. These three factors were shown to be highly interdependent. A collaborative group for the study of multiple endocrine neoplasia type 2 (MEN-2) syndromes comprised of clinicians and laboratory scientists has been set up in the United Kingdom. Its aims are 1) to provide a basis for collaborative work on MEN-2; 2) to establish a register of patients; 3) and specifically to conduct studies aimed at defining the best policy for screening the families of apparently sporadic patients, to establish radioimmunoassays for family screening, and to identify large kindreds for genetic linkage studies using DNA polymorphisms.
