Leukotrienes (LT) are lipid mediators of the inflammatory response that play important roles in diseases such as asthma and atherosclerosis. Leukotriene A4 (LTA4) is synthesized from arachidonic acid (AA) by 5-lipoxygenase (5-LOX) with the help of 5-lipoxygenase activating protein (FLAP), a trimeric nuclear transmembrane protein. Exactly how 5-LOX and FLAP interact is not well understood, however FLAP is essential for the production of leukotrienes in vivo. I used nanodiscs (NDs), engineered membrane systems, as a tool to study the 5-LOX-FLAP interaction. FLAP was incorporated into NDs with a phospholipid composition to mimic of the nuclear membrane. Early life heart rate (HR) and heart rate variability (HRV) reflect autonomic maturation. Intervention with n-3 long chain polyunsaturated fatty acids (LCPUFAs) during pregnancy favorably affects fetal HR and HRV; similar observations have been reported with infant n-3 LCPUFA intake. Infant HR and HRV have not been assessed in relation to maternal fatty acid status during pregnancy. Further, exposure to intrauterine inflammation may underlie these observations, although this hypothesis has not been tested. Cystic fibrosis (CF) is a genetic disorder with a median survival age of 40.7 years. Chronic airway inflammation and dehydration are critical features of CF. The size and structure-dependent hydration and anti-inflammatory properties of glycosaminoglycans (GAGs) such as hyaluronic acid (HA) may help ameliorate these symptoms. The GAGs contained in farmed Alligator mississippiensis by-products offer a potential to improve waste management practices and increase revenue in the alligator industry, through their development for use in biomedical applications. Cardiovascular disease is the leading cause of death in the US, and circulating free fatty acids (FFAs) are known risk factors associated with cardiovascular inflammation. The influence of 14 dietary FFAs (including saturated, mono-, poly-unsaturated and trans), on the expression of inflammatory markers in human coronary arterial smooth muscle (HACSM) and endothelial (HCAEC) cells using a cell culture model was investigated. Diabetes is an inflammatory disease associated with hyperglycemia. Chronic exposure of pancreatic â-cells to glucolipotoxicity stimulates a low-grade inflammation associated with the release of pro-inflammatory cytokines such as interleukin-1â (IL-1â). Increased levels of IL-1â can initially lead to decreased insulin secretion and finally â-cell death. Ellagic acid and quercetin have been reported to be anti-inflammatory in several studies. Vitis rotundifolia (muscadine) or Emblica officinalis (amla) are good sources of ellagic acid and quercetin. Inflammatory cytokines play a significant role in the pathogenesis of obesity-related diseases and have been implicated as integral factors in both early and late phases of atherosclerosis. Lifestyle modifications such as increasing physical activity and making dietary changes to induce weight loss are part of the primary prescription for the treatment of metabolic syndrome. Additionally, physical activity has been implicated as a potentially effective regimen for the control of inflammation, yet little is known about the anti-inflammatory mechanistic alterations induced by physical activity.
