Pigs challenged with hypoglycosylated variants of Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) major surface protein GP5 exhibited increased production of PRRSV-neutralizing antibodies relative to the levels of neutralizing antibodies produced by pigs immunized with wild type (wt) or glycosylated GP5. This invention provides for methods of obtaining improved immune responses in pigs to PRRSV, compositions useful for obtaining the improved immune responses as well as isolated polynucleotides that encode hypoglycosylated variants of PRRSV major surface protein GP5.

The nucleocapsid protein (N) of vesicular stomatitis virus and other rhabdoviruses plays a central role in the assembly and template functions of the viral N-RNA complex. The crystal structure of the viral N-RNA complex suggests that the central region of the N protein interacts with the viral RNA. Sequence alignment of rhabdovirus N proteins revealed several highly conserved regions, one of which spanned residues 282 to 291 (GLSSKSPYSS) in the central region of the molecule. Alanine-scanning mutagenesis of this region suggested that replacement of the tyrosine residue at position 289 (Y289) with alanine resulted in an N-RNA template that is …

A recombinant vesicular stomatitis virus (VSV-PeGFP-M-MmRFP) encoding enhanced green fluorescent protein fused in frame with P (PeGFP) in place of P and a fusion matrix protein (monomeric red fluorescent protein fused in frame at the carboxy terminus of M [MmRFP]) at the G-L gene junction, in addition to wild-type (wt) M protein in its normal location, was recovered, but the MmRFP was not incorporated into the virions. Subsequently, we generated recombinant viruses (VSV-PeGFP-∆M-Mtc and VSV-∆M-Mtc) encoding Mprotein with a carboxy-terminal tetracysteine tag (Mtc) in place of the M protein. A potentially painful experience may modify normal behavioural responses. To gauge the importance of pain relative to predation or social status, we presented competing stimuli, a predator cue or an unfamiliar social group, to two groups of noxiously treated rainbow trout, Oncorhynchus mykiss. In the predator cue experiment, fish were classified as bold or shy. Noxiously stimulated fish did not show antipredator responses, suggesting that pain is the imperative. In the social status experiment, noxiously stimulated fish held individually and undisturbed showed an increase in respiration rate and plasma cortisol. Newby and Stevens’ (2008) paper ‘‘The effects of the acetic acid ‘pain’ test on feeding, swimming, and respiratory responses of rainbow trout (Oncorhynchus mykiss)’’ examines the effects of a noxious stimulus on the behaviour of trout in an attempt to replicate research conducted in my laboratory (Sneddon, 2003a; Sneddon, 2007; Sneddon et al., 2003a,b; Reilly et al., 2008). Retinal ganglion cells that express the photopigment melanopsin are intrinsically photosensitive (ipRGCs) and exhibit robust synaptically driven ON-responses to light, yet they will continue to depolarize in response to light when all synaptic input from rod and cone photoreceptors is removed. The light-evoked increase in firing of classical ganglion cells is determined by synaptic input from ON-bipolar cells in the proximal sublamina of the inner plexiform layer. OFF-bipolar cells synapse with ganglion cell dendrites in the distal sublamina of the inner plexiform layer. We developed a method for somatic cell nuclear transfer in zebrafish using laser-ablated metaphase II eggs as recipients, the micropyle for transfer of the nucleus and an egg activation protocol after nuclear reconstruction. We produced clones from cells of both embryonic and adult origins, although the latter did not give rise to live adult clones.

Growth Response Of Yearling Buffalo Male Calves To Different Dietary Energy Levels, Homayoun Mahmoudzadeh, Hassan Fazaeli

To evaluate different levels of energy for optimum growth in Iranian regional buffalo male calves, a completely randomized study was conducted, using 27 yearling buffalo male calves with initial live weight of 201 ± 14 kg. Three diets were formulated to provide 90%, 100%, and 110% energy level requirements equivalent to those of steers derived from NRC beef cattle recommendations and they were fed ad libitum for 2 consecutive 90 day trial periods where the crude protein was 11.2% in the first and 10.22% in the second period. Dry matter intake of the high energy diets was significantly (P < 0.05) lower than those with lower energy contents in both periods. At both stages, daily gain was higher (P < 0.05) and feed conversion ratio improved when the animals received NRC recommended energy diets. There were no significant differences between treatments for the carcass traits with exception of abdominal fat, which was significantly affected by the energy levels (P < 0.05). It can be concluded that the optimum growth rate of buffalo male calves may be obtained by providing the NRC beef cattle standard dietary metabolizable energy from yearling to 18 months of age.

In order to study the prevalence and etiology of subclinical mastitis a bacteriological survey on 16 Awassi dairy sheep flocks in southern Turkey was conducted. A total of 1458 milk samples from 729 Awassi ewes in mid-lactation were tested with the California mastitis test (CMT). Samples from 170 (11.7%) glands and 135 (18.5%) sheep had positive CMT results. Bacteria were isolated from 93 (6.4%) udder halves and 82 (11.2%) ewes. Positive CMT and bacteriological results were combined to define subclinical mastitis. The objective of this work was to study the protein patterns, plasmid profiles, and antibiotic susceptibility of Staphylococcus intermedius and Staphylococcus aureus isolates originating from mastitic mammary glands of dairy cattle in different parts of Konya province. A total of 114 Staphylococcus species were isolated and identified by conventional bacteriological methods from bovine mastitis. Of the total isolates 77 were identified as S. aureus and 37 as S. intermedius. Intra- and inter-species diversities in the coagulase-positive staphylococci were investigated by analysis of whole-cell protein profiles using sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). A wild boar (Sus scrofa) about 2 years old was brought to our parasitology laboratory from Orhaneli region, Bursa province. It was examined for ectoparasites and 32 lice collected from the animal's head were inspected and identified as Haematopinus suis. Five of them were found to be as nymphs, whereas 27 were adults. This paper is the first report of H. suis found in Bursa province, Turkey.

Phenotypic Characterisation Of Enterococcus Spp. From Femoral Head Necrosis Lesions Of Chickens, Nevi̇n Oktay, Seran Temelli̇, K. Tayfun Çarli

Two types of Enterococcus species were isolated from femoral head necrosis lesions of chickens. A total of 150 femoral articular tissues, of which 121 were from commercial broilers, 18 from broiler breeders, and 11 from layer breeders, were sampled. Out of 48 Enterococcus isolates of these clinical samples, 37 and 11 isolates were identified as Enterococcus faecalis and Enterococcus faecium, respectively. Additionally, 42 (28%) Escherichia coli and 33 (22%) Staphylococcus aureus isolates were obtained either alone or together with 1 of the 2 Enterococcus species from the same clinical samples, while no bacteria were detected from the remaining 56 samples. …

In order to evaluate heat tolerance, physiological constants of 32 cows were measured (Chinampo: n = 12; Holstein: n = 10; Jersey: n = 10). From June to December 2006, rectal temperature and respiratory rate were recorded every Monday at 0600 and 1700 h. Holstein rectal temperature was affected by heat most, followed by Jersey, and Chinampo (the most heat-resistant). Respiratory rate of the 3 breeds was affected by climatic factors, but the Holstein and Jersey cows registered values higher than those of Chinampo cows. With a temperature-humidity index (THI) above 72 the physiological variables of all 3 breeds increased. …
