Bioreactors are utilized in many industries, such as the food, alternative fuel, and pharmaceutical industries, to design and manufacture products. Unlike the similar chemical batch reactors utilized in many chemical processing facilities, bioreactor utilization requires engineers to consider a wider range of operating conditions and parameters. This results in a more complex reaction system and controls network. Bioethanol Production Using Saccharomyces Cerevisae Cultivated In Sugarcorn Juice, Thirumalai Nambi Thiruvengadathan

For the first time, juice extracted from sugarcorn, a new Canadian energy crop, was used for bioethanol production. Physical and chemical characteristics of sugarcorn juice (SCJ) were determined. SCJ contained a maximum of 145 g/L of carbohydrates, with sucrose, glucose and fructose together contributing 80%. Effect of autoclaving and carbon filtration on juice sugars were investigated.
Shake flask fermentations using Saccharomyces cerevisiae grown in yeast extract supplemented SCJ produced a maximum of 45.6 g/L ethanol in 72 h. Bioreactor studies using un-supplemented SCJ achieved 40 g/L ethanol in 26 h, yielding a maximum of 0.46 g ethanol/g fermentable sugars, representing …

The chemical diversity and biological activities of terpene and terpenoids have served in the development of new flavors, fragrances, medicines and pesticides. While terpenes are made predominantly by plants and microbes in small amounts and as components of complex mixtures, chemical synthesis of terpenes remains technically challenging, costly and inefficient. In this dissertation, methods to create new yeast lines possessing a dispensable mevalonate biosynthetic pathway wherein carbon flux can be diverted to build any chemical class of terpene product are described. We report a synthetic biology strategy for rapid genetic manipulation of natural product biosynthetic pathways. Based on DNA assembler, this method synthesizes the entire expression vector containing the target biosynthetic pathway and the genetic elements required for DNA maintenance and replication in various hosts in a single-step manner through yeast homologous recombination, offering unprecedented flexibility and versatility in pathway manipulations.

In addition to providing the molecular machinery for transcription and translation, recombinant microbial expression hosts maintain the critical genotype-phenotype link that is essential for high throughput screening and recovery of proteins encoded by plasmid libraries. It is known that Escherichia coli cells can be simultaneously transformed with multiple unique plasmids and thusly complicate recombinant library screening experiments. As a result of their potential to yield misleading results, bacterial multiple vector transformants have been thoroughly characterized in previous model studies.
