Retention Of Cellular Viability And Growth Potential In Cryopreserved Bovine Arterial Tissue: Implications For Clinical Use Of Cryopreserved Vascular Allografts, Kip G. Gardner

Segments of bovine calf aortas and pulmonary arteries were cryopreserved at -196° C for two weeks to 18 months and subsequently thawed. Cellular morphology, viability, and growth potential in these segments were then compared with the same attributes in segments from fresh (control) arteries. Scanning electron microscopy revealed no disruption of intimal surfaces attributable to cryopreservation. Endothelial cells were successfully cultured from all cryopreserved arterial segments. Endothelial monolayers grown from cryopreserved arteries had a cobblestone appearance and expressed Factor VIII antigen but not smooth muscle alpha-actin. Plating efficiency in primary cultures of endothelial cells was lower for cryopreserved arteries than …

Fucoidan is a polysaccharide commonly found in brown algae. It is a sulfated polymer of the monosaccharide L-fucose. Fucoidan is thought to function in water retention by algae. Commercially available fucoidan is extracted from a brown algae called Eurus vesiculosus.

This polysaccharide has immense potential as a biologically active agent: (i) it exhibits anticoagulant action, (ii) it has also been reported to block retroviral replication and (iii) it is capable of blocking sperm-egg binding in sea urchin, mouse and rabbit. A state-of-the-art computer code has been developed that incorporates a modified Runge-Kutta time integration scheme, Upwind numerical techniques, Multigrid acceleration, and Multi-block capabilities (RUMM). A three-dimensional thin-layer formulation of the Navier-Stokes equations is employed. For turbulent flow cases, the Baldwin-Lomax algebraic turbulence model is used. Two different upwind techniques are available, van Leer's flux-vector splitting and Roe's flux-difference splitting. Full approximation multigrid plus implicit residual and corrector smoothing were implemented to enhance the rate of convergence. The natural convective heat transfer and air movement in a Trombe wall solar passive system has been studied analytically and numerically. Three Trombe wall channel geometries including the parallel channel with axial inlet and exit, parallel channel with side vents and Trombe wall channel coupled to the room have been considered. Several models representing these Trombe wall geometries have been formulated. For the parallel channel with axial inlet and exit geometry, a momentum-integral method has been used to solve parabolic governing equations for two-dimensional laminar flow. This dissertation introduces a binary partitioned approach to statistical pattern classification which is applied to talker identification using neural networks. In recent years artificial neural networks have been shown to work exceptionally well for small but difficult pattern classification tasks. However, their application to large tasks (i.e., having more than ten to 20 categories) is limited by a dramatic increase in required training time. The time required to train a single network to perform N-way classification is nearly proportional to the exponential of N. In contrast, the binary partitioned approach requires training times on the order of N2. …

A comprehensive study of the inelastic stability of steel beam-columns and frames subjected to nonproportional loads is conducted. A set of materially nonlinear differential equations of equilibrium for planar and biaxially loaded beam-columns are first formulated including sidesway. The analysis includes the effect of flexible connections, and initial imperfections in the form of member crookedness and residual stresses. Also, hollow rectangular sections are adopted for the study. First, an iterative numerical solution procedure is formulated for sway beam-columns utilizing central finite-differences with second-order truncation errors. The purpose of this research was to determine the effects of cryopreservation on the physical, biochemical and cellular aspects of heart valve tissue. The present study demonstrated that radiolabeled proline accumulation provides a quantitative estimate of total cellular metabolic viability. Radiolabeled inulin was demonstrated to be a useful marker for extracellular tissue space. Assessment of metabolic viability of the cellular population of leaflet tissue as a function of procurement and processing revealed that proline accumulation declined with increasing warm and cold ischemic times. A new and efficient procedure for aerodynamic shape optimization is presented. The salient lineaments of this procedure are: (1) using a discrete sensitivity analysis approach to determine analytically the aerodynamic sensitivity coefficients; (2) obtaining the flowfield solution either by a computational fluid dynamics (CFD) analysis or, alternatively, by a flowfield extrapolation method which is based on a truncated Taylor's series; (3) defining the aerodynamic shape in such a way that it is not restricted to any class of surfaces and the optimizer automatically shapes the aerodynamic configuration to any arbitrary geometry; and (4) requiring no expertise other than that needed …

The relative performance and flow phenomena associated with several devices for controlling turbulent separated flow were investigated at low speeds. Relative performance of the devices was examined for flow over a curved, backward-facing ramp in a wind tunnel, and the flow phenomena were examined in a water tunnel using dye-flow visualization. Unsteady flows around rigid or flexible delta wings with and without oscillating leading-edge flaps are considered. These unsteady flow problems are categorized under two classes of problems. In the first class, the wing motion is prescribed a priori and in the second class, the wing motion is obtained as a part of the solution. The formulation of the first class includes either the unsteady Euler or unsteady Navier-Stokes equations for the fluid dynamics and the unsteady linearized Navier-displacement (ND) equations for the grid deformation.

The overall approach is to employ the finite-element method along with specified material models to account for material nonlinearity. This study includes two different applications: truss structures and slabs-on-grade.

Procedures for materially nonlinear analysis of truss structures, which apply for both determinate and indeterminate cases are proposed. The proposed procedures are simple and can handle a variety of different stress-strain functions, and the applied load does not have to be incremental. We discuss topics in the theory of nonlinear stochastic control, estimation, and decision via a probabilistic approach using measure transformations and martingale theory. First, we investigate the problem of estimating a diffusion process using coordinate transformations and measure transformations, both locally and globally; this is the analog of nonlinear coordinate and state feedback transformations used to obtain exact linearization in nonlinear deterministic control problems. The objective of this study was to develop a high-resolution-explicit-multi-block numerical algorithm, suitable for efficient computation of the three-dimensional, time-dependent Euler and Navier-Stokes equations. The resulting algorithm has employed a finite volume approach, using MUSCL-type differencing to obtain state variables at cell interface. Variable interpolations were written in the $\kappa$-scheme formulation. Inviscid fluxes were calculated via Roe's flux-difference splitting, and van Leer's flux-vector splitting techniques, which are considered state of the art. The viscous terms were discretized using a second-order, central-difference operator.

Scheduling is an essential factor influencing the efficiency of any production system. The effectiveness of the scheduling system depends upon the interaction of the human and machine. Thus, to effectively design the interface between the human and the machine, the human factors professional must understand scheduling behavior and the information requirements of the scheduling task. The present study modeled human scheduling behavior and determined the information requirements of the scheduling task. The study also compared alphanumeric, direct manipulation graphic, and equivalent interfaces to determine which interface best supports scheduling.
