Neurochemical Changes Following Stroke: Role In Neuroprotection And Cardiac Susceptibility, Raymond Tak Cheung

The mechanisms mediating neuronal recovery and cardiovascular complications after ischemic stroke are not completely known. In these investigations, I examined changes in neuropeptides and a catecholamine synthesizing enzyme, tyrosine hydroxylase (TH), following middle cerebral artery occlusion (MCAO) in adult male Wistar rats.;Five days following MCAO, the immunoreactivity for TH and neuropeptide Y (NPY) increased in the insular cortex (IC) within the peri-infarct zone. NPY also increased ipsilaterally in the basolateral nucleus of the amygdala, whereas leucine-enkephalin, dynorphin and neurotensin increased ipsilaterally in the central nucleus of the amygdala. Anti-DNA antibodies are hallmarks of SLE both in human and inbred lupus prone mice. The role of these antibodies in disease pathogenesis, particularly in glomerulonephritis has been documented. It is now well accepted that the genetic potential to generate anti-DNA antibodies also exists in the normal B cell immune repertoire. This genetic potential has been revealed by detection of anti-DNA antibodies in serum of normal individuals and by immortalizing DNA reactive B cells as hybridomas and as transformed cell lines. Characterization Of Intestinal Epithelial Cells In Acute Intestinal Allograft Rejection, Xian-Chang Li

Intestinal transplantation (IT) remains highly experimental compared to other solid organ transplants because of poorly defined mechanisms of graft rejection. In this project, the role of luminal bacteria-derived endotoxin and inflammatory cytokines (IL-1, IL-6, TNF-{dollar}\alpha){dollar} in intestinal epithelial damage during acute graft rejection was examined. In a novel mouse model of IT, the acute intestinal rejection was associated with elevated levels of endotoxin, IL-6, and TNF-{dollar}\alpha{dollar} in the peripheral blood. The progressive increase in endotoxin and TNF-{dollar}\alpha{dollar} levels correlated well with the histologic severity of epithelial damage. Host and viral determinants influencing the outcome of coronavirus JHM infections of the CNS of rodents were investigated using intact animals and primary neural cell cultures. Two problems pertinent to the process of pathogenesis were addressed. The first was concerned with elucidating host factors responsible for controlling the age dependent nature for induction of the demyelinating form of disease in suckling rats. The Generation Of Cytotoxic Tumour Infiltrating Lymphocyte Clones Against Head And Neck Squamous Cell Carcinoma, Henry J. Lapointe

73 samples of H&NSCCa were processed for these studies. An enzymatic disaggregation protocol for the flow cytometric analysis of DNA content of tumour whole cell preparations was developed. Analysis of the tumour cell component without the need for incorporation of reference controls provided a more accurate assessment of DNA content of tumour cell populations. Six lines of H&NSCCa were established in vitro from fresh tumour samples. Tumour cell effects on PHA- or IL-2 stimulated proliferation of PBMNCs ranged from 61% and 45% suppression to 4% and 121% enhancement of the proliferative responses respectively. The innate ability of the immune system to prevent the onset of cancer involves an intricate network of defense known as the natural killer (NK) cell system. As reactive elements in tumour defense and essential components of adoptive immunotherapy, NK cells represent a relatively new field of study that holds immeasurable potential in our struggle against cancer. Although their anti-tumour potential has been realized, NK cells and many of their regulatory processes, remain a scientific challenge. Postmenopausal osteoporosis is a debilitating condition which affects 25% of Caucasian women. To date, neither prophylaxis nor treatment have met with any great success. Calcium supplementation (CaSuppl) has been widely advocated in the prevention and treatment of osteoporosis despite the paucity of scientific evidence suggesting that calcium has any beneficial effect. We have employed an oophorectomised rat model to investigate the effects of oestrogen deficiency on the skeleton and bone marrow.
