Meta-analysis is a statistical approach that integrates data from multiple studies. By aggregating information, it enhances the power to detect the effects of interest and provides an estimate of the effect size with both accuracy and precision. Both fixed-effect and random-effect models are developed and widely used in biomedical research including clinical trials and genomic studies. In the case of rare events data, conventional meta-analysis methods that rely on large sample approximation may not be able to make reliable inferences. There have been various approaches proposed to deal with this situation, in particular, rare binary adverse events in clinical studies. …

Analysis of biological data for differentiation of organisms/cells within and across species or even the same organism is important to a wide variety of applications. This work considers three different biological data sets at the genome, proteome, and epigenome levels: respectively, DNA sequences, glycosalation data, and DNA methylation. We explore some statistical modeling approaches for handling these modern datasets, and provide a relevant set of experiments for explanation and illustration.

Determining risk to human populations due to natural disasters has been a topic of interest in the STEM fields for centuries. Earthquakes and the tsunamis they cause are of particular interest due to their repetition cycles. These cycles can last hundreds of years but we have only had modern measuring instruments for the last century or so which makes analysis difficult. In this document, we explore ways to improve upon an existing method for reconstructing earthquakes from historical accounts of tsunamis. Deconstructing Sex Differences In C-Reactive Protein Trends Over Time, Ashley W. Kranjac, Dinko Kranjac, Olivia Lounsbury

Objectives

Heightened inflammatory state, as measured by circulating C-reactive protein (CRP) levels, can promote inflammation-mediated disease risk. It is important to account for population fluctuation and sex variation in serum CRP concentrations on overall time trends.

Methods

Using the National Health and Nutrition Examination Survey data, we specify linear and algebraic decomposition models separately by sex to identify the drivers of the changing trends in the distribution of CRP values in the population.

Results

We found a nonsignificant overall increase in CRP, but a significant decrease among women and increase among men, over a 10-year period. We then used linear …

Recent years have witnessed a surge of problems lying at the intersection of statistics andneuroscience. In this thesis, we explore various statistical and computational problems that are inspired by neuroscience. This thesis consists of two main parts, each inspired by a different system in the brain. In the first part, we study problems related to the visual system. The radial velocity method has been widely used by astronomers since the 1990's for discovering extra-solar planets, often referred to as simply "exoplanets". This method involves estimating the radial velocity of a distant star over time using the stellar light, followed by modeling such radial velocity estimates as a function of time using Keplerian-orbital equations with parameters that describe the exoplanet. While a number of approaches exist for estimating the radial velocity from the stellar light, we introduce a new approach for this that uses Hermite-Gaussian functions to reduce the estimation to linear least-squares regression. Neural networks have been intensively studied as machine learning models and widely applied in various areas. This thesis investigates three problems related to the theory and application of neural networks. First, we analyze a learning scheme for neural networks that uses random weights in the backpropagation training algorithm, which is considered to be more biologically plausible than the standard training procedure. We establish theory that shows the convergence of the loss and the alignment between the forward weights of the network and the random weights used in the backward pass. With the rapid development of new data collection and acquisition techniques, high-dimensional data have emerged from various fields. Consequentially, new variable selection methods especially in ultra-high dimensional problems are demanding.

The first part of this dissertation focuses on developing a new Bayesian variable selection method for a differential expression analysis using raw NanoString nCounter data.
