Chlamydia trachomatis infections cause severe and irreversible damage that can lead to infertility and blindness in both males and females. Following infection of epithelial cells, Chlamydia induces production of reactive oxygen species (ROS). Unconventionally, Chlamydiae use ROS to their advantage by activating caspase-1, which contributes to chlamydial growth. NLRX1, a member of the Nod-like receptor family that translocates to the mitochondria, can augment ROS production from the mitochondria following Shigella flexneri infections. However, in general, ROS can also be produced by membrane-bound NADPH oxidases. Given the importance of ROS-induced caspase-1 activation in growth of the chlamydial vacuole, we investigated the …

Understanding emerging viruses is critical for disease monitoring and prediction; however, surveys of novel viruses are hindered by the lack of a universal assay for viruses. Viral metagenomics, consisting of viral particle purification and shotgun sequencing, is a powerful technique for discovering viruses in a wide variety of sample types. However, current protocols are not effective on tissue samples (e.g., lungs, livers and tumors), where they are hindered by the high amount of host nucleic acids which limits the percentage of sequences that originate from viruses. The aim of this study is the three-dimensional structural characterization of the genome packaging inside viral capsids via cryo-electron microscopy and three-dimensional reconstruction. The genome of some single-stranded viruses can be densely packaged within their capsid shells. Several stretches of the genome are known to adopt stable secondary structures, however, to date, little is known about the three-dimensional organization of the genome inside their capsid shells. Two techniques have been developed to facilitate the structural elucidation of genome packaging: the asymmetric random-model method, and the symmetry-mismatch, random model method. Background. Malnourished infants are small for age and weight. Objectives. Determine profiles in 24-hour energy metabolism in recovering malnourished infants and compare to similarly aged healthy controls. Methods. 10 malnourished infants (58.1 ± 5.9 cm, 7.7 ± 5.6 months) were healthy prior to spending 22 hours in the Enhanced Metabolic Testing Activity Chamber for measurement of EE (kcal/min), sleeping metabolic rate (SMR; kcal/min), respiratory quotient (RQ; VCO2/VO2), and physical activity (PA; oscillations in wt/min/kg body weight). Metabolic data were extrapolated to 24 hours (kcal/kg/d). Energy intake (kcal/kg/d) and the proportions (%) of carbohydrate, protein, and fat were calculated. Anthropometrics for …

We have previously shown that the C-glycoside analog of α-galactosylceramide (α-GalCer), α-C-GalCer, displays a superior inhibitory activity against the liver stages of the rodent malaria parasite Plasmodium yoelii than its parental glycolipid, α-GalCer. In this study, we demonstrate that NK cells, as well as IL-12, are a key contributor for the superior activity displayed by α-C-GalCer. Surprisingly, the diminished production of Th2 cytokines, including IL-4, by α-C-GalCer has no affect on its superior therapeutic activity relative to α-GalCer. Finally, we show that the in vivo administration of α-C-GalCer induces prolonged maturation of dendritic cells (DCs), as well as an enhanced …

To counter systemic risk of infection by parasitic wasps, Drosophila larvae activate humoral immunity in the fat body and mount a robust cellular response resulting in encapsulation of the wasp egg. Innate immune reactions are tightly regulated and are resolved within hours. To understand the mechanisms underlying activation and resolution of the egg encapsulation response and examine if failure of the latter develops into systemic inflammatory disease, we correlated parasitic wasp-induced changes in the Drosophila larva with systemic chronic conditions in sumoylation-deficient mutants. Pseudomonas aeruginosa is an opportunistic pathogen that causes life-long pneumonia in individuals with cystic fibrosis (CF). These long-term infections are maintained by bacterial biofilm formation in the CF lung. We have recently developed a model of P. aeruginosa biofilm formation on cultured CF airway epithelial cells. Using this model, we discovered that mutation of a putative magnesium transporter gene, called mgtE, led to increased cytotoxicity of P. aeruginosa toward epithelial cells. This altered toxicity appeared to be dependent upon expression of the type III secretion system (T3SS). In this study, we found that mutation of mgtE results in increased T3SS …

Disinfecting traps that have captured small mammals is one recommendation for preventing occurrence of hantavirus infection; however, the potential effects of disinfection on small mammal trappability have not been investigated thoroughly. We conducted an experiment to compare the effects of 2 disinfectants (Lysol® and household bleach) on the trappability of 4 small mammal species (Peromyscus maniculatus, Neotamias spp., and Spermophilus lateralis). Seasonal and non-seasonal Susceptible-Exposed-Infective-Recovered-Susceptible (SEIRS) models are formulated and analyzed. It is proved that the disease-free steady state of the non-seasonal model is locally asymptotically stable if Rv < 1, and disease invades if Rv > 1. For the seasonal SEIRS model, it is shown that the disease-free periodic solution is locally asymptotically stable when R̅v < 1, and I(t) is persistent with sustained oscillations when R̅v > 1. Numerical simulations indicate that the orbit representing I(t) decays when R̅v < 1 < Rv. The seasonal SEIRS model with routine and pulse vaccination is simulated, and results depict an unsustained decrease …

Many important antibiotics have become nearly obsolete due to the rise of antibiotic resistant pathogens. Rhodococcus, an actinomycete related to the prolific antibiotic producing genus Streptomyces, harbors over 30 genes for secondary metabolism that could be involved in antibiotic production. Several antibiotics have already been reported for Rhodococcus, suggesting the genus may be a good source for new inhibitory compounds. Fifty four soil bacteria were isolated using enrichment culture techniques (including 37 Rhodococcus) and screened for antibiotic producers. There has been a significant increase in the nation's female incarceration rate. During 2006, the number of women in prison increased by approximately 4.5 %. The increase of female prisoners from 2005 to 2006 was larger than the average growth rate of 2.9% from 2000 through 2005. Women ages 35 to 39 made up the largest percentage of female prisoners. At the end of 2006, females made up 7.2% of the prison population under State or Federal jurisdiction, up from 6.7% in 2000. The purpose of this study was to investigate relationships between four personality styles and two important indications of HIV risk behavior, at intake and 3-month follow-up, among a sample of adolescents participating in court-mandated substance abuse treatment in conjunction with an HIV prevention intervention. This study involved a secondary analysis of data from a NIDA funded project (1R01DA011875-01, R. Malow, PI). Predictor variables included levels of antisocial (unruly), dependent(submissive), avoidant (inhibited), and borderline (borderline tendency) personality styles drawn from scales of the Million Adolescent Clinical Inventory (MACI). This study examined the distinguishing physiological characteristics of the disgust reaction across different domains. According to an evolutionary analysis, disgust is a heterogeneous emotion with features that are specific to three distinct domains: pathogens, sex, and morality. Each domain is predicted to take as input information specific to the adaptive problem it evolved to solve and regulate behavior accordingly. The goal of the present study was to investigate whether there are any adaptive physiological differences associated with the disgust response across domains. Background Intravenous injection of mice with attenuated Plasmodium berghei sporozoites induces sterile immunity to challenge with viable sporozoites. Non-intravenous routes have been reported to yield poor immunity. Because intravenous immunization has been considered to be unacceptable for large scale vaccination of humans, assessment was made of the results of intradermal immunization of mice with Plasmodium yoelii, a rodent malaria parasite whose infectivity resembles that of human malaria. Capsular polysaccharides (CP) of serotypes 5 (CP5) and 8 (CP8) are major Staphylococcus aureus virulence factors. Previous studies have shown that salicylic acid (SAL), the main aspirin metabolite, affects the expression of certain bacterial virulence factors. In the present study, we found that S. aureus strain Reynolds (CP5) cultured with SAL was internalized by MAC-T cells in larger numbers than strain Reynolds organisms not exposed to SAL. In the early 1990’s, due to political circumstances at home, Somali immigrants and refugees began arriving in the state of Minnesota in large numbers. Over the past two decades, Somali immigrants have come to comprise one of the most populous ethnic groups in the Twin Cities, and are the largest Somali population in the world outside of Eastern Africa. Although quantitative data is unavailable, qualitative evidence and testimonies of healthcare professionals support the conclusion that Somali immigrants in Minnesota suffer from higher rates of diabetes than non-immigrant groups and than they likely did before migration.
