Attenuated total internal reflectance Fourier transform infrared (ATR-FTIR) spectroscopy was used for the investigation of sorption of aqueous solutions of analytes into polymer coatings. A series of simple model polymers, such as poly(dimethylsiloxane), poly(epichlorhydrin), and poly(isobutylene), and films and analytes, such as aqueous solutions of ethylbenzene, xylenes, toluene, and nitrobenzene, were used to evaluate the use of ATR-FTIR spectroscopy as a screening tool for sensor development. The ratios of integrated infrared absorption bands provided a simple and efficient method for predicting trends in partition coefficients. Particle size distributions in diesel exhaust were studied using a thermal desorption technique to determine the volatile and non-volatile fractions. A thermal denuder was designed and characterized for optimal operating parameters and its influence on the exhaust chemistries studied using diesel engine exhaust. The study was conducted on a 1992 DDC Series 60 engine mounted on a heavy-duty direct current dynamometer and exercised over steady state and transient cycles. The particle size distributions were recorded upstream and downstream of the denuder maintained at different temperatures to determine the behavior of the volatile fraction. Detailed observations of stream, soil, and groundwater chemistry were used to determine the role of fire, permafrost and snowmelt processes on the fluxes of carbon, nitrogen and major solutes from interior Alaskan catchments. We examined an experimentally burned watershed and two reference watersheds that differ in permafrost coverage (high, 53%; medium-burn, 18%; and low, 4%) during the FROSTFIRE prescribed burn in July 1999. The fire elevated stream nitrate concentrations for a short period during the first post-fire storm, but nitrate declined thereafter, suggesting that less severe fires that leave an intact riparian zone may have only a short-term effect on …

Dissolved proteins are important components of high molecular weight marine dissolved organic matter (DOM). Little is known about the sources, transformations, and pathways of marine DOM, and <30% has been characterized at the molecular-level. Advances in proteomics technology have improved our ability to sequence proteins, making dissolved proteins a suitable target for molecular-level characterization. Dissolved proteins are important biomarkers in the oceans, and molecular-level analysis can provide valuable insight into their chemical characteristics, structural modifications, and potential sources. Molecular-level characterization of dissolved proteins in seawater requires a reproducible recovery method with efficient recovery yields and minimal sample losses. The use of surface blocking agents (SBA) to saturate sites of non-specific interactions reduced adsorptive sample losses during recovery of dissolved proteins from artificial seawater by tangential flow ultrafiltration. The SBA pre-treatment resulted in more reproducible recovery yields (∼55–60%) and enhanced initial recovery by 46%. Dissolved proteins recovered from deep and surface seawater samples were separated by gel electrophoresis, revealing the presence of a small number of discrete bands and significant amounts of unresolved background. A proteinase K assay determined that the bands and unresolved background were proteinaceous in nature. Enzymatic digests of the dissolved proteins were analyzed by reversed phase chromatography coupled on-line to a linear trapping quadrupole Fourier transform mass spectrometer (LTQ-FTMS). The peptide MS/MS spectra were searched against non-redundant databases by correlation algorithms, but no positive identifications were made. The peptide tandem mass spectra were directly interpreted by de novo sequencing to generate peptide tags that were used in database searches to develop protein homologue information. The similarity search results revealed two trends for specific classes of proteins observed (membrane/envelope proteins and enzymes) and identified conserved sequences for several multi-species protein homologues. These results demonstrate that there are several different classes of refractory proteins represented in the DOM pool and provide evidence for the presence of dissolved proteins other than bacterial outer membrane proteins. Additionally, an intact protein was extracted from a gel band and analyzed by direct infusion ESI FTMS. The resultant high resolution protein spectrum was deconvoluted to determine the accurate molecular weight of the protein.

We tested the hypothesis that the physical and chemical processes acting in sea grass habitats of the lower Chesapeake Bay are spatially structured and that dissolved elemental chemistry of sea grass-habitat surface waters have their own unique identity. We sampled surface waters from July to September 2001 in five sea grass habitats of the lower bay: Potomac, Rappahannock, York, Island (Tangier-Bloodsworth), and Eastern Shore. Dissolved Mg, Mn, Sr, and Ba concentrations were measured by sector field inductively coupled plasma-mass spectrometry. In the natural process of wet deposition, gas-water interfaces play an important role in the transport of chemical contaminants in the atmosphere via fog, rain and cloud drops. Evidences from several other works point out deviations in gas-liquid partitioning as predicted by Henry’s law. Uptake and mass transfer of benzene, naphthalene, and phenanthrene was chosen to study in a falling droplet train apparatus. Higher droplet-to-vapor partition constant (KDV) was noted for diameters less than 200ìm and was attributed to surface adsorption and accumulation. This study investigates the isotopic signatures of human skeletal remains that were recovered from several sites along the coast and inland in the north-central Patagonian region of Argentina. Human skeletal remains, dating from 2500 BP through the early historic period, are examined to determine the relative significance of terrestrial and aquatic food resources and subsequently, the extent to which coastal food resources were exploited by indigenous Argentinians. Carbon and nitrogen isotopes contained within human bone collagen and apatite are measured quantitatively to determine the relative significance of marine and terrestrial foods.
