Assessment Of Anti-Poaching Effort In Ruaha National Park, Tanzania, C. L. Nahonyo
Assessment Of Anti-Poaching Effort In Ruaha National Park, Tanzania, C. L. Nahonyo
Tanzania Journal of Science
Anti-poaching effort in Ruaha National Park for the period between January 1996 to June 1999 was evaluated in terms of strength, frequency and duration of patrols, area covered and success of patrols. It was found that the park spent 72.4 US$ km-2 year-1 on law enforcement. There were 72 rangers each required to patrol on the average 143 km2. A total of 1,255 patrols were conducted; patrols ranged from 14 to 43 trips per month and each lasted between 1 to 25 days and most were foot and vehicle patrols. Patrol units had 2 to 11 rangers while the average …

Embryonic Stem Cell-Derived Hematopoietic Stem Cells, Yuan Wang, Frank Yates, Olaia Naveiras, Patricia Ernst, George Q. Daley
Dartmouth Scholarship
Despite two decades of studies documenting the in vitro blood-forming potential of murine embryonic stem cells (ESCs), achieving stable long-term blood engraftment of ESC-derived hematopoietic stem cells in irradiated mice has proven difficult. We have exploited the Cdx-Hox pathway, a genetic program important for blood development, to enhance the differentiation of ESCs along the hematopoietic lineage. Using an embryonic stem cell line engineered with tetracycline-inducible Cdx4 , we demonstrate that ectopic Cdx4 expression promotes hematopoietic mesoderm specification, increases hematopoietic progenitor formation, and, together with HoxB4, enhances multilineage hematopoietic engraftment of lethally irradiated adult mice. Clonal analysis of retroviral integration sites …

We examined the hypothesis that short-tailed shearwaters Puffinus tenuirostris aggregate to forage at the inner front of the SE Bering Sea because of enhanced production there. We tested this hypothesis by comparing primary production, the distribution of euphausiids and the distribution of shearwaters relative to the front during late spring and late summer/early fall of 1997, 1998 and 1999. We found enhanced primary production at the front and offshore of the front during summer but not during spring. Primary production varied between seasons and years. Major differences were related to anomalous conditions in 1997 and 1998. The density of euphausiids …

Multiple Stable Isotopic Analyses ([Delta]¹³C, [Delta]¹⁵N, [Delta]¹⁸O, And [Delta]D) Of The Boulder Patch, A High Arctic Kelp Community: Trophic And Temporal Perspectives, Casey William Debenham
Marine Biology
The Boulder Patch, a high Arctic kelp community, is a rarity in the Alaskan Beaufort Sea. Considered a biodiversity oasis, this area provides habitat for many organisms. Trophic relationships, spatial patterns, and isotopic changes over time were examined within the Boulder Patch using stable carbon and nitrogen isotopes. 394 samples, representing over 55 species were analyzed. Isotope values showed considerable variability in the food web base, particularly for the kelp Laminaria solidungula. Isotopic values for most animals fit their known feeding strategies. Little spatial variation was observed in isotope values, however temporal differences were found in L. solidungula isotope values …

dTDP-L-rhamnose, an important precursor of O-antigen, was prepared on a large scale from dTMP by executing an one-pot reaction in which six enzymes are involved. Two enzymes, dTDP-4-keto-6-deoxy-D-glucose 3,5-epimerase and dTDP-4-keto-rhamnose reductase, responsible for the conversion of dTDP-4-keto-6-deoxy- D-glucose to dTDP-L-rhamnose, were isolated from their putative sequences in the genome of Mesorhizobium loti, functionally expressed in Escherichia coli, and their enzymatic activities were identified. The two enzymes were combined with an enzymatic process for dTDP-4- keto-6-deoxy-D-glucose involving TMP kinase, acetate kinase, dTDP-glucose synthase, and dTDP-glucose 4,6- dehydratase, which allowed us to achieve a preparative scale synthesis of dTDP-L-rhamnose using dTMP …

Aromatase, the enzyme responsible for estrogen biosynthesis, is a particularly attractive target in the treatment of hormone-dependent breast cancer. The synthesis and biological evaluation of a series of 2-(4′-pyridylmethyl)thio, 7-alkyl- or aryl-substituted isoflavones as potential aromatase inhibitors are described. The isoflavone derivatives demonstrate IC50 values from 79 to 553 nM and compete with the endogenous substrate, androstenedione. Data supporting the ability of these analogs to suppress aromatase enzyme activity in the SK-BR-3 breast cancer cell line are also presented.

Host-choice experiments were carried out with rodent and bat ectoparasites on Ilha Grande, state of Rio de Janeiro, Brazil. We constructed experimental chambers that enclosed three different rodent or bat host species, and then introduced a selected set of ectoparasitic arthropods. When given the opportunity to choose among host species, the ectoparasites showed a strong tendency to select their primary hosts, and reject novel host species. These kinds of simple experiments can be valuable tools for assessing the ability of ectoparasites to locate and discern differences between host species, and make choices about which hosts to infest, and which hosts …
The Custody Battle Over Cryogenically Preserved Embryos After Divorce: Advocating For Infertile Women’S Rights, Cori S. Annapolen
The Custody Battle Over Cryogenically Preserved Embryos After Divorce: Advocating For Infertile Women’S Rights, Cori S. Annapolen
ExpressO
This paper focuses on the struggles that infertile women face to achieve motherhood because their rights are underrepresented in the American court system. It specifically centers on how the process of in vitro fertilization (IVF) helps infertile women conceive children, but then details the problems that increasing technology now causes for these women after they freeze embryos and then divorce. Because the courts of only four states have determined who gets custody of these embryos after a divorce, and because the divorce rate and the number of couples utilizing IVF are increasing, future states will likely be forced to answer …

Mapping Tampa Bay Cynoscion Nebulosus Spawning Habitat Using Passive Acoustic Surveys, Sarah Lyle Walters
USF Tampa Graduate Theses and Dissertations
Spotted seatrout, Cynoscion nebulosus, spawning locations as well as associated environmental variables were determined for Tampa Bay, Florida during the 2004 spawning season using a mobile hydrophone survey. Hydrophones, a type of underwater microphone, can be used to detect and record spawning sounds of soniferous fishes. During their spawning season in Tampa Bay which generally occurs between March and September, mature male spotted seatrout generate sounds associated with courtship in the crepuscular and evening periods by vibrating sonic muscles against the swim bladder. Active spawning sites can be located using hydrophones to find these calling males.

Directional transport of the phytohormone auxin is required for the establishment and maintenance of plant polarity, but the underlying molecular mechanisms have not been fully elucidated. Plant homologs of human multiple drug resistance/P-glycoproteins (MDR/PGPs) have been implicated in auxin transport, as defects in MDR1 (AtPGP19) and AtPGP1 result in reductions of growth and auxin transport in Arabidopsis (atpgp1, atpgp19), maize (brachytic2) and sorghum (dwarf3). Here we examine the localization, activity, substrate specificity and inhibitor sensitivity of AtPGP1. AtPGP1 exhibits non-polar plasma membrane localization at the shoot and root apices, as well as polar localization above the root apex. Protoplasts from …

The Optimal Discovery Procedure: A New Approach To Simultaneous Significance Testing, John D. Storey
The Optimal Discovery Procedure: A New Approach To Simultaneous Significance Testing, John D. Storey
UW Biostatistics Working Paper Series
Significance testing is one of the main objectives of statistics. The Neyman-Pearson lemma provides a simple rule for optimally testing a single hypothesis when the null and alternative distributions are known. This result has played a major role in the development of significance testing strategies that are used in practice. Most of the work extending single testing strategies to multiple tests has focused on formulating and estimating new types of significance measures, such as the false discovery rate. These methods tend to be based on p-values that are calculated from each test individually, ignoring information from the other tests. As …

Mixture Cure Survival Models With Dependent Censoring, Yi Li, Ram C. Tiwari, Subharup Guha
Mixture Cure Survival Models With Dependent Censoring, Yi Li, Ram C. Tiwari, Subharup Guha
Harvard University Biostatistics Working Paper Series
A number of authors have studies the mixture survival model to analyze survival data with nonnegligible cure fractions. A key assumption made by these authors is the independence between the survival time and the censoring time. To our knowledge, no one has studies the mixture cure model in the presence of dependent censoring. To account for such dependence, we propose a more general cure model which allows for dependent censoring. In particular, we derive the cure models from the perspective of competing risks and model the dependence between the censoring time and the survival time using a class of Archimedean …

The Optimal Discovery Procedure For Large-Scale Significance Testing, With Applications To Comparative Microarray Experiments, John D. Storey, James Y. Dai, Jeffrey T. Leek
UW Biostatistics Working Paper Series
As much of the focus of genetics and molecular biology has shifted toward the systems level, it has become increasingly important to accurately extract biologically relevant signal from thousands of related measurements. The common property among these high-dimensional biological studies is that the measured features have a rich and largely unknown underlying structure. One example of much recent interest is identifying differentially expressed genes in comparative microarray experiments. We propose a new approach aimed at optimally performing many hypothesis tests in a high-dimensional study. This approach estimates the Optimal Discovery Procedure (ODP), which has recently been introduced and theoretically shown …

Application Of A Multiple Testing Procedure Controlling The Proportion Of False Positives To Protein And Bacterial Data, Merrill D. Birkner, Alan E. Hubbard, Mark J. Van Der Laan
U.C. Berkeley Division of Biostatistics Working Paper Series
Simultaneously testing multiple hypotheses is important in high-dimensional biological studies. In these situations, one is often interested in controlling the Type-I error rate, such as the proportion of false positives to total rejections (TPPFP) at a specific level, alpha. This article will present an application of the E-Bayes/Bootstrap TPPFP procedure, presented in van der Laan et al. (2005), which controls the tail probability of the proportion of false positives (TPPFP), on two biological datasets. The two data applications include firstly, the application to a mass-spectrometry dataset of two leukemia subtypes, AML and ALL. The protein data measurements include intensity and …

The Kinetics Of Particulate Substrate Utilization By Bacterial Films, Joshua Boltz
The Kinetics Of Particulate Substrate Utilization By Bacterial Films, Joshua Boltz
LSU New Orleans Theses and Dissertations
There is a need to develop a mathematical expression capable of describing the removal of particulate chemical oxygen demand (PCOD) from wastewaters in biological film systems. In this context, organic particles that are maintained in suspension (i.e., not removed during normal settling) are the focus of experimentation, modeling, and discussion. The goal of this research project is to study the kinetics of PCOD removal from wastewaters by bacterial films, or biofilms. To achieve this objective, a bench-scale rotating disc biofilm reactor (RDBR) was operated using methanol (dissolved substrate), Min-U-Sil 10 (inorganic particulates), and Maizena corn starch (organic particulates) dissolved/suspended in …

Development Of Nicarbazin As A Reproductive Inhibitor For Resident Canada Geese, Kimberly S. Bynum, Christi A. Yonder, John D. Eisemann, John J. Johnston, Lowell A. Miller
Wildlife Damage Management Conference
Expanding populations of resident Canada geese that remain in suburban and urban areas year-round often result in increased conflicts with humans. Non-lethal and humane means are needed for managing the size of Canada goose flocks residing near or on airports, golf courses, industrial parks, government sites, and city parks. A side effect of nicarbazin, a veterinary drug used to control coccidiosis in chickens, is decreased egg production and hatching. Exploiting this side effect, studies of nicarbazin for reducing the hatchability of eggs from Canada geese were conducted. An initial study in Cotumix quail verified reduction in hatchability in a species …

Cell Wall Functions Required For Mating In Saccharomyces Cerevisiae, Stephen Dougherty
Cell Wall Functions Required For Mating In Saccharomyces Cerevisiae, Stephen Dougherty
Renée Crown University Honors Thesis Projects - All
In order for two haploid yeast cells to undergo normal fusion during mating, adhesive contacts are formed by a general class of fungal proteins termed “adhesins”. Within this family there is a subgroup of cell wall proteins termed “agglutinins” which are known to mediate aggregation, thereby increasing mating efficiency in liquid medium (Aga1p, Aga2p, Aga1p). In the absence of sufficient mating adhesive contacts we observed that cells undergo defective morphogenesis, apparently continuing to grow at their tips after fusing at the expense of laterally expanding their conjugation bridge. Based on these qualitative observations, I utilized differential interference microscopy to capture …
Conservation Of Castanea Dentata Genetic Resources Of The Southeastern United States, Mark Thomas Alexander
Conservation Of Castanea Dentata Genetic Resources Of The Southeastern United States, Mark Thomas Alexander
Masters Theses and Doctoral Dissertations
The ongoing effort by the American Chestnut Foundation to breed and reintroduce a blight resistant, locally adapted variety of American chestnut (Castanea dentata) requires a broad genetic base. To ensure adequate genetic diversity, genetic resources must be collected from throughout the native range of the species. Finding and breeding surviving C. dentata stump sprouts is problematic due to the relatively short life span of blooming stems and their often remote forest habitat. To facilitate more efficient location of surviving trees in the southeastern U.S., we used a Geographic Information System (GIS) to overlay soil, topography, and satellite imagery for the …

Current Primary Production Rates Of The Western Arctic Ocean Estimated By Stable Carbon And Nitrogen Isotope Tracers, Sang Heon Lee
Marine Biology
Currently, the environments in the Arctic are rapidly changing. These changes of climate and ice conditions may alter the quantity, quality, and timing of production of ice algae and phytoplankton in the Arctic Ocean. The objectives in this study were to detect any change in the carbon production between current and previous studies and lay the groundwork for the future monitoring of ecosystem response to climate change in the different regions of the western Arctic Ocean. As an Arctic ocean mostly covered by multi or first-year ice, the deep Canada Basin had generally low photosynthetic rates and the maximum rates …

We propose a unified and flexible framework for ensemble learning in the presence of censoring. For right-censored data, we introduce a random forest algorithm and a generic gradient boosting algorithm for the construction of prognostic models. The methodology is utilized for predicting the survival time of patients suffering from acute myeloid leukemia based on clinical and genetic covariates. Furthermore, we compare the diagnostic capabilities of the proposed censored data random forest and boosting methods applied to the recurrence free survival time of node positive breast cancer patients with previously published findings.
