This study aims to answer a presented gap in the literature on how gateway community workers and businesses are directly impacted by federal government shutdowns. Three gateway communities were selected as case-study communities for this research based on a set list of criteria, including size, proximity to urban centers, geographic context, and the prominence of their associated park. The selected communities are Bar Harbor, Maine; Estes Park, Colorado; and Moab, Utah. As of 2023, outdoor recreation participation in the United States (U.S.) reached a 15-year high, generating $1.2 trillion in economic output and supporting 5 million jobs. Concurrently, the contiguous U.S. experienced earth surface temperatures averaging above the 20th-century norm for the 27th consecutive year. Climate change presents increasing challenges for outdoor recreation, particularly in the Northeast and North Central U.S., where seasonal shifts, extreme weather events, and resource degradation are projected to acutely impact recreation patterns and management needs. Indigenous water sovereignty has long been a contested problem within the continent of Turtle Island. In 2016, the Lakota Nation globalized the message Water is Life through their movement against the Dakota Access Pipeline, an oil infrastructure that would put one of the continent’s largest watersheds at risk. Fondly drawing upon excursions in coastal Maine with her grandnephew Roger Christie, Carson illustrates the powerful impact of simply accompanying children in nature and letting their curiosity and questioning guide the adventure. Spectroscopic imagers have been used to analyze the surface of Mars at varying spatial scales in several distinct modalities, each of which provides different information about surface geology and geochemistry. In this dissertation, we take a closer look at two such spectroscopic imagers: the Compact Reconnaissance Imaging Spectrometer for Mars (CRISM) on board the Mars Reconnaissance Orbiter and the Planetary Instrument for X-ray Lithochemistry (PIXL) on board the Mars 2020 rover Perseverance. This thesis juxtaposes a historical account of environmental toxicity in Maine against the popular notion of Maine as a “Vacationland.” Maine is famous globally for its pristine aesthetics, iconic coastline, and vast wilderness. Because of this, tourism is amongst the largest economic sectors for Maine, much different from the complex industrial history that once drove the economy and built a strong foundation for the state. Phytoplankton are abundant in both fresh and marine waters and can form harmful algal blooms (HABs) by producing potent toxins that have devastating effects on human, wildlife, and economic health. The HAB-forming diatom genus, Pseudo-nitzschia, occurs globally and approximately half of the 59 currently identified species can produce the neurotoxin, domoic acid (DA). DA accumulates in shellfish and finfish and can cause Amnesic Shellfish Poisoning (ASP) in humans and DA Poisoning (DAP) in several species of wildlife, resulting in severe neurological dysfunction and mortality. This paper broadly examines how rivers can be used as a pawn in war and, more specifically, how the Penobscot River, located in Penobscot County, Maine, influenced significant battles in America and the fabric of the developing nation due to its geographical location and geological development. This analysis focuses on one critical aspect of the Penobscot River that motivated attacks: the fur, fish and lumber trades, all of which were established due to the proximity of the coast and how the land developed during the Holocene era.

Privately owned water is the primary source of drinking water for 43 million Americans. Although residential or private wells are susceptible to a variety of contaminants, the Safe Drinking Water Act of 1974 positions individuals as responsible for the testing, remediation, and management of this water. Despite the elevated presence of arsenic in Maine, which is linked to various cancers, cardiovascular disease, and neurological damage, little is known about how private well owners perceive the safety and quality of their own water.

Across the country the number of search and rescue missions in national parks has been creeping up since 2018. While the number of missions being undertaken are up overall, some parks in particular have seen a dramatic spike in the number of people requiring rescue services, straining a patchwork, often volunteer-based search-and-rescue system. Some parks like Maine’s Acadia National Park, Zion National Park in Utah and the Grand Canyon National Park even experienced their busiest years to date in regards to the number of search and rescue calls they received.

The dogfish, Squalus acanthias is a marine cartilaginous elasmobranch found in the North Atlantic and Pacific oceans. Dogfish synthesize and excrete urea as a product of nitrogen metabolism. They also convert ammonia into urea and retain this urea, such that their plasma is isosmotic or slightly hyperosmotic to the surrounding seawater. To facilitate the regulation of body fluid and to maintain ionic concentrations and osmotic pressure, Dogfish use ion and solute transporters (e.g., NCC, UT-1) and aquaporin water channel proteins. Environmental stress disrupts cellular homeostasis leading to proteostasis disruptions which must be correct by rapid reprogramming of transcription and translation mediated by the translation machinery of which tRNA and aminoacyl tRNA synthetases are particularly important. Our aim in this study was to conduct a genome-wide RNAi kinase screen of a long-lived and stress-resistant rrt-1(gc47) (MC240) C. elegans mutant that constitutively, under naïve conditions, expresses a GFP reporter for gpdh-1, which is required for survival during hypertonic stress, but not expressed under naïve, non-stress conditions. Student loan levels in the United States have risen since their inception by the Federal Government in 1958 and they continue to rise today. Economists and the general public are at odds about what this means. The public narrative, as evidenced by news coverage, op-eds, social movements and polling data, suggest that student loans have become a “crisis” that negatively impacts the lives of young Americans. This thesis focuses on identification and development of novel small molecule therapeutics for blood cancer. The studies presented herein examine two classes of novel compounds with different mechanisms: tropolones as chemotherapies (Chapters II and III) and phosphoantigens as immunotherapies (Chapters IV-VII).

A library of α-tropolone derivatives was tested for anti-proliferative effects on a panel of hematological malignancies demonstrating preference towards cells of T cell lineage. Through investigating different compounds, some structure activity relationships for compound potency were determined. Osmoregulation is facilitated by using an assortment of ion and water channels to assist acclimation to changing conditions and to maintain cellular homeostasis. Euryhaline fish can be found in both seawater and freshwater (SW and FW) environments. Expression of ion and water channels have been experimentally demonstrated to change as a fish acclimates to different environmental conditions. Successful non-native species can reduce native species richness through both direct and indirect competition. Many invasive ants, such as the European fire ant (Myrmica rubra), are particularly successful invaders due to their ability to form multi-nest, multi-queen “supercolonies” that appear to displace native ant and non-ant invertebrate fauna in invaded regions. Pseudo-nitzschia is a pennate marine diatom that produces the neurotoxin domoic acid (DA). Domoic acid accumulates in the digestive tract of filter feeders and becomes concentrated at higher trophic levels. When DA concentrations reach critical levels, consumption of toxic shellfish can induce a condition known as Amnesic Shellfish Poisoning (ASP), which has been responsible for the deaths of marine mammals, seabirds and humans. The oxidative phosphorylation pathway (OxPhos) is the only pathway where proteins are encoded by both the mitochondrial and nuclear genomes. It is also responsible for creating the majority of ATP used by animal cells. The critical role this pathway plays in animal cells, and it’s reliance on two genomes makes it a perfect system to study the co-evolution of the mitochondrial and nuclear genomes. Three interviews with island health leaders, one community conversation with older islanders, and one pilot test provided information about what to include in the template.

Phytoplankton blooms follow predictable annual cycles in the Gulf of Maine, characterized by a large spring bloom and a smaller bloom in the fall. Marine phytoplankton form the foundation of primary production in Gulf of Maine waters, and thus community changes in composition, and abundance could have cascading effects on our coastal ecosystems.

Mercury (Hg) is a volatile element increasing in concentration in the environment as a result of anthropogenic emissions. Microorganisms can transform mercury into monomethylmercury (MMHg), the form of Hg that bioaccumulates, biomagnifies, and can harm humans and wildlife. Most studies of MMHg bioaccumulation in wildlife have focused on aquatic organisms due to consumption of fish being the primary route of human exposure to MMHg. In the early nineteenth century, a new form of human exhibitionism spread through eastern American cities. While public displays featuring live human beings had existed since the colonial era, these new shows specifically focused on Native Americans. This paper examines one such show, the Inuit Exhibition of 1820-1821, as a case study of this phenomena. Primarily through the use of contemporary newspaper accounts, this project argues that shows like the Inuit Exhibition occurred within a cultural context that legitimized the practice of human exhibitionism as a genuine, post-Enlightenment method of educating citizens about the natural world. Daphnia is a commonly used test organism in toxicology, ecology, and evolution. HR96 is a CAR/PXR/VDR ortholog in invertebrates, and a promiscuous endo- and xenobiotic nuclear receptor involved in acclimation to toxicants. Daphnia HR96 is activated by chemicals such as atrazine and linoleic acid (LA) (n-6 fatty acid), and inhibited by triclosan and docosahexaenoic acid (DHA) (n-3 fatty acid). HR96 is also a key regulator of Magro, Niemann Pick genes, and other genes involved in cholesterol and triacylglycerol uptake, metabolism, and transport. Marine calcifiers, especially those in their larval and juvenile stages, are thought to be most vulnerable to ocean acidification (OA) due to the effects of carbon dioxide (CO2) on growth and shell mineralogy. However, recent evidence is contradictory. We monitored molting activity, length and weight in early benthic phase juvenile American lobster Homarus americanus (Milne-Edwards 1837) under elevated CO2 conditions (500 µatm, 1100-1300 µatm, and 2000-2700 µatm) to determine how OA affects growth at this life stage. Large whales have historically been difficult to study and many aspects of their ecology remain unknown especially at the long -term population level. The ability to identify individual whales based on natural markings provides the opportunity to track individuals over time and space; this data may offer more insight into the ecology of whales than previously imagined. Although biomarkers are frequently used to assess sublethal effects of contaminants, a lack of mechanistic linkages to higher-level effects limits the predictive power of biomarkers. Bioenergetics has been proposed as a framework for linking cellular effects to whole-animal effects. We investigated sublethal effects of exposure to wastewater treatment facility effluent in freshwater mussels in situ, thereby capturing ecologically relevant exposure conditions. Our study focused on the energetic biomarker AMP-activated protein kinase (AMPK), while also considering more traditional biomarkers like heat shock proteins (HSP70), and antioxidant enzymes (i.e., superoxide dismutase (SOD), glutathione-S-transferase (GST)).
