The Na+/K+-ATPase (NKA) is a central regulator of cardiac ion homeostasis and a validated target for heart failure. Yet, cardiotonic steroids used in clinical practice are limited by a narrow therapeutic window and pro-arrhythmic effects. Here, we identify Ro 41-0960 as a non-steroidal inhibitor of NKA with a distinct mechanistic profile. The compound inhibits NKA activity with IC50 values of 17.9 ± 1.1 µM for purified enzyme and https://doi.org/10.3 ± 1.1 µM in microsomal preparations. This 1980 interview of Normal Cromwell was done by Pete Boughn at the University of Nebraska Medical Center in Omaha. He asked about Cromwell’s history but mostly about his time as director of the Eppley Cancer Center. The interview is reproduced with permission from the UNMC McGoogan Library.
Cromwell was born in 1913 in Terre Haute, Indiana. In 1935, he earned a bachelor’s degree in chemical engineering from the Rose-Hulman Institute of Technology in his hometown. Mycoplasma pneumoniae (MP) is a common pediatric respiratory pathogen that can rarely present with extrapulmonary manifestations such as hepatic involvement. Recognition of hepatic dysfunction in this context is important for accurate diagnosis and timely therapy. A five-year-old female with asthma presented with fever, cough, fatigue, pruritus, and scleral icterus. Laboratory evaluation revealed leukocytosis, elevated C-reactive protein, mild transaminitis, direct hyperbilirubinemia, elevated PT/PTT, and normocytic anemia. Infectious and hepatic workups were initially unrevealing. The patient showed gradual clinical improvement while receiving empiric ceftriaxone but had persistent leukocytosis. The variability in cathode compositions within recycled lithium-ion battery (LIB) feedstocks poses a significant challenge to efficient downstream refining processes. This study demonstrates the feasibility of using froth flotation with oleic acid as a collector to selectively separate lithium nickel-manganese-cobalt oxide (NMC) and lithium cobalt oxide (LCO) from lithium manganese oxide (LMO) materials. Laboratory-scale flotation tests achieved an 80% separation efficiency in a single stage, producing a froth product with >90% purity of NMC/LCO at approximately 90% yield. Punjab, India's primary rice and wheat production hub, has witnessed rapid expansion of paddy cultivation over the past two decades, driven by minimum support price incentives, changes in government policies, alignment of sowing with the monsoon season and the adoption of high-yielding varieties. This transition has intensified groundwater extraction and shortened the fallow period between rabi and kharif crop seasons, reducing the window between rice harvesting and wheat sowing, leading to widespread open-field burning of rice residue and recurrent post-monsoon air-quality deterioration across the Indo-Gangetic Plain. Single-grain pollen analysis is essential for advancing chemical palynology, which classifies morphologically similar taxa based on taxon-specific pollen and sporopollenin chemistry. Fourier-Transform Infrared (FT-IR) spectroscopy and microspectroscopy (μFT-IR) are widely used to differentiate morphologically similar grains but face limitations from Mie scattering and coarse spatial resolution. Here, we present the first application of Optical-PhotoThermal Infrared (O-PTIR) spectroscopy to chemically treated single pollen grains, using Molinia caerulea as a model taxon to fully test and evaluate the technical capabilities and limitations of this technique. O-PTIR achieves sub-micron “super-resolution” infrared spectroscopy through non-contact, non-destructive measurements by exploiting the photothermal effect. O-PTIR spectra …

"AI & Chemistry" is a four-week language-learning project based on the action-oriented approach. The project asks students to complete two main tasks: prepare an individual Ignite talk and a partnered podcast on any subtopic related to the use of AI in chemistry. Beyond language skills, the project additionally aims to encourage students’ collaboration with AI-powered tools and enhance their AI literacy. All project steps are detailed in an additional PDF file. The integration of the project is not restricted to a language course; it can be adapted for any university-level communicative course.

Mixed lineage leukemia (MLL) gene rearrangements cause ~75% of acute leukemia in infants and 5-10% in children and adults with poor clinical outcomes. Protein-protein interactions (PPI) between frequent MLL fusion partners AF9/ENL and AF4 or histone methyltransferase DOT1L are drug targets for MLL-rearranged (MLL-r) leukemia. Sixty-seven quinoxiline compounds were synthesized and tested for their ability to inhibit such PPIs. Compounds 16, 17, 59 and 63 were found to be potent inhibitors with IC50 values of 0.35-1.5 μM. Chemiluminescence is a phenomenon of light emission that occurs when molecules transition from an excited state to the ground state. This phenomenon is widely utilized in applications ranging from quantifying product yields in chemical reactions to biological imaging and atmospheric chemistry studies. It has also been used in many crucial applications in the scarce conditions of heat and electricity, such as light sources for astronomy discovery or deep-sea diving. The demands of emerging technologies, such as soft robotics, stretchable electronics, and next generation biomedical implants, has led to the development of diverse polymeric and hybrid organic/inorganic materials specifically tuned to meet these challenges. Biomass burning (BB) affects air quality and climate by releasing large quantities of gaseous and particulate pollutants into the atmosphere. Photochemical processing during daylight transforms these emissions, influencing their overall environmental impact. Accurately quantifying the photochemical drivers, namely actinic flux and photolysis frequencies, is crucial to constraining this chemistry. However, the complex radiative transfer within BB plumes presents a significant challenge for both direct observations and numerical models.

Therapies capable of resolving inflammatory lung disease resulting from high-consequence occupational/environmental hazards are lacking. This study seeks to determine the therapeutic potential of direct lung-delivered interleukin (IL)-10 following repeated lipopolysaccharide exposures. C57BL/6 mice were intratracheally instilled with LPS (10 μg) and treated with IL-10 (1 μg) or vehicle control for 3 days. Lung cell infiltrates were enumerated by flow cytometry. Lung sections were stained for myeloperoxidase (MPO), CCR2, vimentin, and post-translational protein citrullination (CIT) and malondialdehyde-acetaldehyde (MAA) modifications. Lung function testing and longitudinal in vivo micro-CT imaging were performed. Whole lungs were profiled using bulk RNA sequencing. IL-10 treatment reduced …

Environmentally persistent free radicals (EPFRs) are stable free radicals formed on particulate matter (PM) through processes such as combustion and pyrolysis. These free radicals are generated on transition metal oxide surfaces in the presence of aromatic precursors. Exposure to EPFRs occurs primarily via inhalation of PM deriving from combustion, traffic, industrial activities, and both indoor and outdoor burning. Other environmental factors that might generate EPFRs are radon, electronic and tobacco cigarettes. EPFRs exhibit unexpectedly long half-lives, ranging from several weeks to, in some cases, several years. In "DNA Sequence Context Modulates the Impact of a Cisplatin 1,2-GpG Intrastrand Cross-Link", Pilch, Todd, and Lippard found that the DNA sequence around the binding site influences how tightly cisplatin binds (Pilch, Todd, and Lippard).
Biophysical techniques like melting temperature and circular dichroism showed that some sequences stabilize DNA more strongly after drug binding (Pilch, Todd, and Lippard).
This explains why cisplatin is more effective in some tumor genomes (Todd and Lippard).

Science shows are often posited as a route to encourage young people to engage with science. Using a mixed methods approach, our research investigated the impact of attending a science show on high school students’ intentions to study science or pursue a career in science. We found that while a single visit to a science show has little impact on students’ aspirations, it can reinforce students’ confidence in their ability to succeed in science. In terms of design and performance, the best-received shows included ‘real-world’ content that students could identify as relevant to their lives.

Historically, it has been a challenge to simulate the experimentally observed cellular structures and marginal behavior of multi-dimensional, thin-channel hydrogen detonations in the presence of losses, even with detailed chemistry models. Very recently, a quasi-two-dimensional inviscid approach with a calorically perfect gas and two-step chemistry was pursued, where losses due to viscous boundary layers were modeled by the inclusion of an equivalent mass divergence in the lateral direction using Fay's source term formulation with Mirels’ compressible laminar boundary layer solutions. BACKGROUND: A phase I clinical study for patients with locally advanced H&N cancer with a new class of botanical drug APG-157 provided hints of potential synergy with immunotherapy. We sought to evaluate the efficacy of the combination of APG-157 and immune checkpoint inhibitors.
METHODS: CCL23, UM-SCC1 (human), and SCCVII (HPV-), MEER (HPV+) (murine) H&N cancer cell lines were utilized for in vitro and in vivo studies. We measured tumor growth by treating the mice with APG-157, anti-PD-1, and anti-CTLA-4 antibody combinations (8 groups). Chagas disease is one of the world’s neglected tropical diseases, caused by the human pathogenic protozoan parasite Trypanosoma cruzi. There is currently a lack of effective and tolerable clinically available therapeutics to treat this life-threatening illness and the discovery of modern alternative options is an urgent matter. T. cruzi glucokinase (TcGlcK) is a potential drug target because its product, D-glucose-6-phosphate, serves as a key metabolite in the pentose phosphate pathway, glycolysis, and gluconeogenesis. In 2019, we identified a novel cluster of TcGlcK inhibitors that also exhibited anti-T. cruzi efficacy called the 3-nitro-2-phenyl-2H-chromene analogues. In an ongoing effort to incorporate active learning and promote higher order learning outcomes in undergraduate organic chemistry, a hybrid (“flipped”) classroom structure has been used to facilitate a series of collaborative activities in the first two courses of the lower division organic chemistry sequence. An observational study of seven classes over a five-year period reveals there is a strong correlation between performance on the in-class activities and performance on the final exam across all classes; however, a significant number of students in these courses continue to struggle on both the in-class activities and final exam. Poly-ADP-Ribose Polymerase (PARP-1) is an overexpressed enzyme in several carcinomas; consequently, the design of PARP-1 inhibitors has acquired special attention. Hence, in the present study, three compounds (8–10) were produced through a Michael addition protocol, using phenylmethanethiol, 5-fluoro-2-mercaptobenzyl alcohol, and 4-mercaptophenylacetic acid, respectively, as nucleophiles and perezone as the substrate, expecting them to be convenient candidates that inhibit PARP-1. Use of a contraceptive (i.e., fertility control) is attractive for rodent management where lethal control is unwanted. Although population reduction is generally unachievable with small-scale, short-term contraceptive use, reduced juvenile recruitment is achievable. The injectable immunocontraceptive vaccine GonaCon (active ingredient: gonadotropin releasing hormone [GnRH]) was registered in 2022 by the United States Environmental Protection Agency (EPA) for controlling fertility of female prairie dogs (Cynomys, a type of ground squirrel) in urban/suburban settings.
