Amyotrophic lateral sclerosis/frontotemporal dementia (ALS/FTD) is a progressive neurodegenerative disease spectrum characterized by motor dysfunction, cognitive decline, and death within a few years of clinical onset. Hallmark features of ALS/FTD include defects in nucleocytoplasmic transport (NCT) and abnormal cytoplasmic mislocalization and aggregation of the RNA-binding protein TDP-43, both of which have been shown to be exasperated by oxidative stress (OS). It remains unclear how these processes are connected and how they collectively contribute to neurodegeneration. In this study, I used an ecological and a cultural perspective to examine the first-year experiences of two cohorts of biology students exposed to multiple equity-minded High Impact Practices (HIPs) designed to promote their persistence and science identity development.
In chapter 1, I applied Graham et al.’s Persistence Framework (2013) and Bronfenbrenner’s Ecological Systems Theory (1977) to investigate the bridges and barriers to persistence that biology students face while experiencing either an equity minded learning community, a first-year seminar, a course-based research experience, and/or an active learning introductory biology course. Chapter One will detail the synthesis, purification, and characterization of DNA oligonucleotides containing one of fifteen modified nucleic acid bases. For each modification, relevant background information is provided to highlight its biological significance. This chapter primarily focuses on the development of oligonucleotide stocks for use in future biochemical and cellular assays. Several of these modifications are recognized substrates of AlkB-family enzymes, while others remain uncharacterized as potential substrates.1-4 The discovery of novel substrates for AlkB-family enzymes can be facilitated through comprehensive in vitro biochemical assays, contingent upon the availability of well-characterized substrate stocks. Undergraduate introductory human anatomy and human physiology courses are either taught as discipline-specific or integrated anatomy and physiology (A&P) sequences. These courses are critically important to future health care professionals yet a consensus is lacking on the optimal course approach. Our institution underwent a curricular revision to change pedagogical methodologies from discipline-specific human anatomy and human physiology to an integrated A&P I and II sequence. To enable natural and fluent human robot collaboration, it is critical for a robot to comprehend their human partners’ on-going actions, predict their behaviors in the near future, and plan its actions accordingly. Specifically, the capability of making early predictions can allow robots to determine the precise timing of turn-taking events and start planning and executing preparative tasks to take the turn. Such proactive behavior would save waiting time and increase efficiency and naturalness in collaborative tasks.

Heat stress is a global issue that crosses socioeconomic status. Heat stress leads to reduced worker capacity on seasonal scales, and weekly to sub-daily timescales, incapacitation, morbidity, and mortality. This dissertation focuses on 2 distinct parts: quantification methods of heat stress, and heat stress applications. Quantification methods of heat stress Chapters 1–3 focus on historical analysis of heat stress. How and why functional connectivity (FC), which captures the correlations among brain regions and/or networks, differs in various brain states has been incompletely understood. I review high-level background on this problem and how it relates to 1) the contributions of task-evoked activity, 2) white-matter fMRI, and 3) disease states in Chapter 1. In Chapter 2, based on the notion that brain activity during a task reflects an unknown mixture of spontaneous activity and task-evoked responses, we uncovered that the difference in FC between a task state (a naturalistic movie) and resting state only marginally (3-15%) reflects task-evoked connectivity. This dissertation is a study on the ways in which Virtual Reality can actively and reactively affect human physiology. The first manuscript describes the design of a highly generalized Virtual Reality biofeedback system. This system was developed such that, given proper design, it would be able to provide users with a highly responsive Virtual Environment to meet the criteria of embodiment, which will increase the user's responsiveness to stimuli. The system also functions as a biofeedback system that can be used for biomechanical as well as physiological feedback by using a generalized design paradigm.

One of the prominent causes of heart failure is atherosclerosis of the coronary arteries. Obstruction of blood flow within the coronary arteries results in inadequate perfusion, or ischemia, of the heart muscle. The let-7 family of microRNAs (miRs) regulates critical cell functions, including survival signaling, metabolic control and glucose utilization, which may be important during myocardial ischemia. MiR-let-7 expression is under tight temporal and spatial control through multiple redundant mechanisms. Intraperitoneal (IP) chemotherapy is a promising post-surgical therapy of ovarian cancer, with the full potential yet to be proven. To facilitate IP chemotherapy of ovarian cancer, we have developed a nanoparticle depot for IP chemotherapy consisted of paclitaxel (PTX) nanocrystals (PNC) and hyaluronic acid-based hydrogel (HA gel). PNC with a size of ~310 nm was produced by nonsolvent and temperature-induced crystallization. Dissolution kinetics of PNC could be determined by the light scattering method rather than the dialysis method due to drug reprecipitation caused by diffusion barrier. This dissertation describes an interdisciplinary research project that spans computer-vision, imaging of biological nanoparticles via cryogenic transmission electron microscopy (cryo-tem), and bio-informatics / proteomics. This research introduces a new method for reconstructing individual biological particles imaged via cryo-tem with typical accuracies of 88% to 98%. This individual particle reconstruction (IPR) technique is based on a hidden Markov model derived from the underlying biological structure. NMR spectroscopy is a powerful analytical tool for both qualitative and quantitative metabolite profiling analysis. However, accurate quantitative analysis of biological systems especially using one dimensional NMR has been challenging due to signal overlap. In contrast, the enhanced resolution and sensitivity offered by chemoselective isotope tags have enabled new and enhanced methods for detecting hundreds of quantifiable metabolites in biofluids using NMR spectroscopy or mass spectrometry. Microtubules are cytoskeletal protein polymers orchestrating a host of important cellular functions including, but not limited to, cell support, cell division, cell motility and cell transport. In this thesis, we construct a toy-model of the microtubule lattice composed of vector Ising spins representing tubulin molecules, the building block of microtubules. Nearest-neighbor and next-to-nearest neighbor interactions are considered within an anisotropic dielectric medium. As a consequence of the helical topology, we observe that certain spin orientations render the lattice frustrated with nearest neighbor ferroelectric and next-to-nearest neighbor antiferroelectric bonds. Aim: The aim of this study was to obtain descriptions of nurses’ perceptions and experiences of falling asleep behind the wheel on the drive home after a worked night shift.
Background: While the evidence suggests that the problem of drowsy driving and fall asleep driving is a strong contributor to vehicular accidents, little is known about nursing perceptions of the issue.

Myocardial transcriptional response to stress is critically dependent on availability of the histone acetyltransferase p300. p300 is a transcription coactivator which is rapidly induced by hemodynamic stress in the myocardium. This induction is both necessary and sufficient to drive myocardial hypertrophy in vivo. Although short-term elevation of p300 may be adaptive, sustained elevation of p300 increases the risk of heart failure. The mechanism of p300 induction during stress is unknown. According to an evolutionary analysis, disgust is a heterogeneous emotion with features that are specific to three distinct domains: pathogens, sex, and morality. Estrogen acts via a seven transmembrane receptor (7TMR), GPR30, to stimulate an HB-EGF autocrine loop. Here, evidence is provided that integrin α5β1 is a necessary transmembrane signaling intermediary required for matrix metalloproteinase (MMP)-mediated release of proHB-EGF by GPR30. Estrogen stimulation of human SKBR3 breast cancer cells, which lack ERα and ERβ, resulted in recruitment of integrin α5β1 to focal adhesions, formation of actin stress fibers, and assembly of fibronectin (FN) fibrils. Expression of a GPR30 mutant in SKBR3 cells selectively abrogated FN matrix assembly and EGFR tyrosine phosphorylation by estrogen. Determination of the age of a bloodstain can be used to establish the time when a crime was committed. As blood ages, deoxyhemoglobin (HbO 2) is converted into methemoglobin (MetHb), which is evident by the color change from red to brown. Fluctuations in temperature will cause blood to age at a non-constant rate. It has been shown that higher temperatures cause deoxyhemoglobin to oxidize more rapidly to methemoglobin.
