Mast cells secrete a variety of biological mediators crucial for innate and adaptive immunity. One such mediator is Tumor Necrosis Factor (TNF). TNF released from mast cells and other immune cells has been shown to elicit complex signaling pathways in target cells, resulting in the production of pro-inflammatory cytokines and/or immune cell recruitment. At the molecular level, TNF secretion/exocytosis is mediated by soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) and other regulatory proteins that work together to promote the fusion between secretory granules and the plasma membrane. One of the diseases caused by cellular regulation disturbance is cancer and mutations in EGF-related genes highly correlate with worse outcomes in cancer. The Epidermal Growth Factor (EGF) pathway is involved in the regulation of cell growth and differentiation. Hydrogels have been explored for many biomedical applications, including targeted, in situ drug delivery to avoid the negative side effects associated with systemic delivery. In our work, we are exploiting the high affinity, calcium-dependent binding between calmodulin and its target peptides to create a biomaterial for in situ, extracellular drug delivery. Genes were engineered to make two protomers, Calmodulin Collagen-Like Protein (CCLP) and Peptide Collagen-Like Protein (PCLP), that will spontaneously self-assemble in situ due to the high Ca2+ concentration in the extracellular space and provide tunable, targeted drug delivery in a biocompatible hydrogel. With the research on persons health and well being increasing within the past years, new questions have emerged on if certain environments can also benefit along with exercise. This study looked at the different psychological states of endurance runners as they run in urban environments and transition into rural settings. The results of this study showed significant differences between urban and rural environments. Also showed increased focus going downhill in nature and greater inward attention while moving uphill in nature. With the findings from this study, it explains why runners choose to run or train where they do.

This study investigated natural product inhibitors of blood coagulation Factor XII (FXII). FXII initiates the Kallikrein Kinin-System (KKS) within the intrinsic coagulation pathway. Increased expression of this system leads to proinflammatory and procoagulant activities. Inflammation occurs through downstream activation in the KKS. Following the activation of FXII-dependent pathway, bradykinin (BK) binds to the BK receptor 2 (B2) and produces inflammatory signaling. Abnormally increased BK can result in life threatening Hereditary Angioedema (HAE), characterized by severe reoccurring swelling. FXII is critical for pathogenic thrombosis, but is dispensable for hemostasis. This unique property of FXII makes it an attractive drug target. We …

Calmodulin is an intensely studied, intracellular, calcium-binding protein that is involved in diverse physiological processes. When bound to calcium, calmodulin undergoes a conformational change that allows it to bind specific target peptides with high affinity, one being the calmodulin binding region of myosin light chain kinase, herein called M13. This high affinity, calcium dependent association has made it a desirable choice for use in a biomaterial for drug delivery for non-steroidal anti- inflammatory drugs (NSAIDs). Background: Aquaporins (AQPs) are transmembrane water channels that facilitate osmotically-driven transmembrane water flux. AQP1 is abundantly expressed in vascular endothelium, where it functions in endothelial cell migration, wound healing, and cell volume regulation in response to mechanical stimuli. AQP1 protein abundance is enhanced in cultured human umbilical vein endothelial cells (HUVECs) in response to fluid shear stress. Statins, a pharmacological class of HMG-CoA reductase inhibitors, suppress the early development of vein graft intimal hyperplasia by improving endothelial cell function, most likely by affecting endothelial response to arterial shear stress.
