In this thesis, the upper-limit volume (gas plus partial tissue volume) as well as absolute volume (gas only) of lungs measured using 3He Magnetic Resonance (3He-MR) imaging is compared with that determined by micro-Computed Tomography (micro-CT) and xenon-enhanced micro-CT (Xe-CT) in normal rats. The upper-limit and absolute volumes obtained from He-MR and air-CT methods were not significantly different (P>0.05). Also, no significant difference was found between the volumes measured from He-MR and Xe-CT methods (P>0.05). The good agreement between the volumes measured from He-MR and air-CT suggests that He MR imaging can be used for quantitative analysis of …

The recent curricular reforms in mathematics education in Punjab, Pakistan and Ontario, Canada are studied in this work. The countries are first studied individually and then compared against each other. Using historical sociology, differences between traditional and modern education as well as the underlying theories of mathematics learning are incorporated to explain the shifts in the mathematics curricula in both the cases. Three genes encoding multidrug resistance proteins were identified in Trichoplusia ni larvae, named trnMDRl, trnMDR2 and trnMDR3. Their open reading frames were sequenced, and the translated sequences were compared to each other as well as to multidrug resistance proteins from other species. Quantification of their expression in various tissues of caterpillars fed on either artificial diet or organically- grown cabbage revealed upregulation of trnMDRl in Malpighian tubules, integument and fat body, trnMDR2 in fat body and trnMDR3 in brain in response to the plant diet. The acute respiratory distress syndrome (ARDS) carries a mortality rate of approximately 30% and is characterized by poor oxygenation, decreased lung compliance, and pulmonary edema. The first study in this thesis examined the combination of high frequency oscillatory ventilation (HFO) and exogenous surfactant treatment in an acid- induced lung injury model in rats. It was hypothesized that HFO and surfactant combined is superior to either intervention individually. The second study tested whether surfactant supplementation with a polymer called chitosan would improve surfactant treatment in the same acid model. Heart failure from myocardial infarct (MI) is the leading cause of death in North America. While stem cell therapy for MI has been examined in many animal models and in clinical triais, the ability of stem cells to regenerate damaged myocardium is controversial. I have examined the ability of bone marrow and skeletal muscle derived stem cells to differentiate into cardiac and skeletal muscle following injection into infarcted dog hearts. The purpose of this thesis is to reconsider the nature of ontology in contemporary political science, with the belief that such a move can be of great benefit to understanding changes in our era of globalization and terrorism. Unique individual vocal signatures are widespread among animals living in colonies such as the maternity roosts of Eptesicusfuscus. Such individual signatures have been identified in the echolocation calls of E. fuscus in the laboratory, but have not been demonstrated in the wild. By recording known wild E. fuscus as they emerged from their roosts at dusk, I tested the hypothesis that individual and/or roost signatures exist in the echolocation calls of wild E. fuscus. A simulation-based approach to assess seismic hazard and risk is developed and implemented. Its use together with the seismic hazard model proposed by the Geological Survey of Canada is presented. Sensitivity analyses investigating the effects of smoothing historical seismicity, epistemic uncertainty, attenuation relations, and time-dependent earthquake occurrence on seismic hazard are given. By using the developed lifecycle cost model of a structure and adopting the minimum expected lifecycle cost criterion, the optimal seismic design level for Canadian sites is investigated. Inflorescence bracteoles of Crataegus (Rosaceae) were investigated with the following objectives: i) to determine the taxonomie significance of bracteoles in Crataegus; and ii) to perform a pilot study on the functions of Crataegus bracteoles. The former was accomplished by a morphometric analysis using 25 bracteole characters from 75 species of Crataegus and 3 outgroup taxa. The latter was accomplished by experiments and observations on bracteole-ant interactions, exudate collection and analysis, and cell organelle staining. Understanding the commitments both an employee and his or her employer make to each other is important to organizations. I explore these commitments by examining the relation between employee commitments and employer psychological contracts (i.e., promises employers make to their employees). Traditionally, psychological contracts have been measured based on the contents of the contract (e.g., promotion opportunities), making it difficult to generalize findings across work arrangements. X-linked a-thalassaemia mental retardation (ATR-X) syndrome is characterized by severe cognitive delay, broad developmental abnormalities and a-thalassaemia. Disease-causing mutations in the ATRX gene give rise to a malfunctioning protein and result in aberrant DNA methylation at specific repeat sequences. ATRX is a SNF2 chromatin remodeling protein that targets α-globin gene expression, but additional targets remain largely elusive. Using a mouse model in which Atrx is conditionally deleted in the developing forebrain, I show that DNA methylation at repeat sequences is comparable to that found in ATR-X patients. Retinoic acid (RA) plays a crucial role in regulating cardiomyocyte differentiation and heart tube formation, but excess RA inhibits cardiomyocyte differentiation. We have used microarray technology and whole mount in situ hybridization to investigate the mechanism by which RA might alter cardiogenesis. Our results showed that RA inhibits cardiomyocyte differentiation at least partially through increasing follistatin gene expression and decreasing BMP signaling. Using cultured explants of the heart region, we demonstrated that addition of BMP4 and activin rescued the inhibitory effects of follistatin and RA on cardiomyocyte differentiation. Staphylococcus aureus infection is becoming a widespread threat in both hospitals and communities worldwide, resulting in substantial treatment costs and numerous patient deaths. Bacterial survival is largely dependant on the organism’s ability to scavenge iron from a host via specialized pathways. The recently identified cell walland membrane-associated Iron regulated surface determinant (Isd) proteins are one such pathway. Estimating the fixation probability of a beneficial mutation has a rich history in theoretical population genetics. However, fixation probabilities are extremely sensitive to assumptions regarding life history. We develop a “burst-death” life history model which assumes that generation times are exponentially distributed, but the number of offspring per individual is fixed. Using this model, we estimate the fixation probability for populations of constant size, and for populations which grow exponentially between periodic population bottlenecks. We then predict the optimal time at which to impose bottlenecks, maximizing the probability that beneficial mutations occur and are not ultimately lost. We find that …

Magnetic resonance imaging (MRI) can be used to track DC in vivo because it non-invasively combines critical anatomical detail and sensitivity. To identify cells using MRI it is necessary tolabel them with a contrast agent. DC were labelled with superparamagnetic iron oxide (SPIO) ∞ntrast agents for in vivo imaging. It was found that Feridex was taken up intracellularly and has minimal impact on the DC size and expression of cell maturation markers. The human DNA polymerase lambda (Pol λ) is a recently discovered member of the X-family DNA polymerases that has been shown to be involved in DNA repair and meiotic recombination, where it plays a gap filling role in these two processes. The protein’s primary sequence is organized into an N-terminal BRCAl-C terminai (BRCT) domain, a proline-rich domain and a C-terminus polymerase beta-like (Pol β-like) domain. Symbolic polynomials, whose exponents themselves are integer-valued multivariate polynomials, arise often in algorithm analysis. Unfortunately, modern computer algebra systems do not provide ample support for their algebraic structures. Basic operations involving symbolic polynomials are indeed trivial (addition, multiplication, derivatives); however, other crucial operations remain much more difficult, such as factorization and GCD. The time delay introduced by zone-2 distance relays limits the use of these devices for extra high voltage (EHV) applications since the fast fault clearance requirement is not achieved. The Lunge Test Difference Score (LTDS) measures ankle dorsiflexion ROM in weightbearing. Its reliability and validity were estimated in 53 participants (34.6 ± 13.9 years) with orthopaedic ankle injuries. The LTDS, the Lower Extremity Functional Scale, and the American Academy of Orthopaedic Surgeons Global Foot and Ankle Scale were recorded on three occasions. Test-retest reliability was determined from the results of the final two testing sessions. Correlations between the LTDS and the functional measures were used to validate the LTDS. Correlation between the Global Rating of Change scale and change in LTDS quantified sensitivity to change. The ICC2,1 was 0.94 …

Despite a growing body of research, a ∞mprehensive understanding of suicide remains elusive. The current dissertation involved the development of an integrated cognitive affective theory of suicidal thinking and behavior, and the initial assessment of components of this theoretical model in three separate, but related studies. In Study 1, the integrity of the model was assessed in a sample of 397 (83 male and 314 female) undergraduate students. The resulting fit indices (CFI=.87; GFI=.90; RMSEA=.09) of the structural equation modeling (SEM) analysis indicated an adequate fit between the hypothesized model and the data in the sample, with the model amounting …

The purpose of this dissertation was to describe athletes’ use of the functions of observational learning in various sport situations. A secondary purpose was to explore whether the Functions of Observational Learning Questionnaire (FOLQ; Cumming et al., 2005) could be used to examine naturalistic observational learning use with various groups of sport participants. To achieve these purposes, three studies were conducted. Study 1 examined the relationship between skill level, age, and use of the functions of observational learning in a sample of golfers.
