Studies of widely diverse species of a protein are a powerful tool to gain information on the structure and function of the protein. We investigated the response of human, pig, shark and killifish cystic fibrosis trans-membrane conductance regulator (CFTR) to specific inhibitors of the channel: CFTRinh-172, GlyH-101, and glibenclamide. In several expression systems, including isolated perfusions of the rectal gland, primary cell cultures of rectal gland tubules and oocyte expression, we observed fundamental differences in the sensitivity to inhibition by these CFTR blockers. We hypothesized that lymph node involvement in minor salivary gland cancers is associated with clinical and pathological factors commonly available to the clinician after a typical initial workup. Our aim was to identify these factors using a dataset that allowed us to compile the largest series of minor salivary gland cancers in the published literature. Using this dataset we also aimed to characterize the distribution of histological types by primary site, identify the predictors of the use of external beam radiation therapy and neck dissection, and examine the effect of lymph node involvement on survival. Search For The Basolateral Potassium Channel In The Shark Rectal Gland: Functional And Molecular Identification Of A Task-1 Channel Coupled To Chloride Secretion, Conner Telles

In the shark rectal gland (SRG), apical Cl- secretion through CFTR channels is tightly coupled to a basolateral K+ conductance. The identity of this K+ conductive pathway is unknown. Studies were performed in the isolated perfused SRG with 16 K+ channel inhibitors at their IC50 and with acidic perfusate. During maximal chloride secretion stimulated by forskolin and IBMX, secretion was inhibited >90% by barium chloride, a non-selective inhibitor of K+ channels. Specific inhibitors of calcium sensitive, voltage sensitive, ATP sensitive, and inward rectifying K+ channels had no effect on chloride secretion. During the warmer months of the year the eastern Gulf of Maine features a plume of cold water which extends from the tidally well-mixed area adjacent to Grand Manan Island at the mouth of the Bay of Fundy southwest along the Maine coast to well beyond Penobscot Bay. Near Grand Manan Island the plume waters are cold (ca. 10°C), nearly vertically isothermal and carry high concentrations of dissolved inorganic nutrients. The analysis of temperature-salinity diagrams and hydrographic vertical section plots indicate that the deeper waters in Jordan Basin, of slope water origin, upwell and contribute a significant fraction of the …
