Improving Diabetes Self-Management Through A Partnership Between Clinical Providers And Community Educators In Appalachian Communities, Soghra Jarvandi, Lydia Hoskins, Karen Carver, Andrew May

Introduction: The burden of diabetes is an increasing public health challenge, especially in the Appalachian Region.

Purpose: This study assessed the impact of a community-based diabetes education program on glycemic control.

Methods: Health Extension for Diabetes (HED) is a 4-month diabetes education program offered by Extension educators in collaboration with clinical providers. Participants’ data were collected via phone interviews at baseline and after completing the program. Assessments included self-reported HbA1c, socio-demographics, diabetes knowledge, self-efficacy, and self-management behaviors. Data were analyzed by SAS software using paired analyses.

Results: Of the 105 participants in the program, 87 (83%) completed the post-assessment. Among …

Traumatic knee injury leads to posttraumatic osteoarthritis (PTOA) and significant skeletal muscle weakness, resulting in chronic disability. The current standard of care frequently fails to prevent musculoskeletal dysfunction, underscoring the need to identify therapeutic mechanisms of PTOA. Using an established preclinical anterior cruciate ligament (ACL) transection model of PTOA and leveraging an innovative SPiDER-senescence-associated β-galactosidase stain to discern senescent cells, we investigated cellular senescence at single-cell resolution and identified anti-inflammatory macrophages as a predominant contributor to the senescent cell burden in both muscle and knee joint after injury. 5-year experiments with herbicides were run in the mixture of alfalfa, red clover, timothy grass, and Italian ryegrass. On untreated plots, in the 5-year average 72 weed plants were found per 1 m2 , belonging to 66 weed species. Among them most numerous were: Stellaria media, Veronica agrestis, Poa annua, Lamium purpureum,Taraxacum officinale, and Capsella bursa pastoris. The most efficient herbicide was Etazin, killing 88 % of weeds. None of the applied herbicides controlled perennial weeds. Etazin was tolerated by alfalfa, but was toxic to the other three crops. With the transition to industrialized crop and animal production in the GDR, sprinkler irrigation of field forage plants and grassland, above all with sewage water and clean water +slurry mixtures, is becoming more and more important. It gets increasingly concentrated on field grass and grass/clover stands. The supple­mental water requirement is 210 to 270 mm/ year in case of clean-water sprinkling and up to 400 mm/year if sewage water is used. Introduction: Filtered cigars are a class of tobacco products that can be consumed similarly as conventional cigarettes. Here we have compared tobacco smoke condensates prepared from the 1C2 reference filtered cigar with those from the 1R6F reference cigarette with respect to effects on cell proliferation and viability and AhR-mediated gene expression. Methods: Tobacco smoke condensates were prepared using ISO, Health Canada Intense (HCI) or Cigar Smoking (CSR) regimens and certain HPHCs were measured. Cell walls of forage were studied for structural and chemical factors that limit digestion by rumen microorganisms. Tech­niques used to investigate digestion were histochemistry, electron microscopy, and anaerobic culturing of rumen populations on specific substrates. Certain tissues, such as the xylem cells, totally resisted degradation by rumen microorganisms; these tissues stained positive for lignin with acid phloroglucinol (Weisner test). Other supportive tissues, such as the sclerenchyma in blades, stained positive for lignin with chlorine-sulfite (and also the Maule test) and were also generally resistant to microbial digestion. Non-small cell lung cancer (NSCLC) remains a leading cause of cancer-related mortality worldwide, yet its molecular drivers are not fully defined. Emerging evidence highlights the importance of tumor-stroma interactions mediated by secreted glycoproteins. However, the mechanisms by which cancer cells regulate the secretion of these protumorigenic proteins remain largely unknown. Endoplasmic reticulum-resident (ER-resident) N-glycan-processing enzymes regulate proper protein folding, a prerequisite for glycoproteins to exit the ER and undergo secretion. By evaluating their prognostic significance in lung tumors and conducting functional screening in lung cancer cells, we identify α-glucosidase II (α-Glc II) as a key regulator of NSCLC progression. α-Glc …

Microcystin-LR (MC-LR) is a potent hepatotoxin that has been shown to cause liver damage even at doses lower than the established Low Observable Adverse Effect Level (LOAEL) of 200 μg/kg in animal models. We have previously observed that low-dose exposure to MC-LR in animals with diet-induced Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) and subsequent treatment with antioxidants like N-acetylcysteine (NAC) and the Na+/K+ ATPase-Src kinase inhibitor pNaKtide significantly alleviated hepatic infiltration of immune cells, downregulated markers of inflammation and hepatotoxicity, increased the breakdown of the toxin molecule, and restored phase I and II drug metabolism pathways, including …

Background/Objectives: Signaling mediators of PPARγ influence pathways involved in adipogenesis, lipid storage, inflammation, energy-related processes, and glucose utilization. Recent research indicates that PPARγ coregulators, recruited or released during ligand binding, govern specific gene pathways. It was recently discovered that Gαq, a heterotrimeric G protein subunit, also signals to PPARγ and may significantly affect adipogenesis and glucose sensitivity. The Dot/Icm type IV secretion system (T4SS) is essential for Legionella pneumophila infection, but its in situ architecture and mechanism remain incompletely understood. Using cryo-electron tomography, we performed subtomogram averaging and 3D classification to resolve structural heterogeneity within the complex. We identified multiple assembly states of the inner membrane complex, including a fully assembled form with a hexamer-of-dimers DotO ATPase and symmetry mismatches between subcomplexes. A composite in situ model revealed a central channel above the inner membrane, likely used for substrate secretion. A set of procedures for the selection of Rhizobium strains for enhanced dinitrogen fixation in forage legumes is presented. Although forage legumes are among the most efficient members of the Leguminosae regarding biological nitrogen fixation (BNF), they are generally neither productive nor persistent without effective nodulation by appropriate rhizobia. The following methods define a selection and evaluation process to obtain appropriate Rhizobium strains. Over 10 million patients undergoing non-cardiac surgery each year face major cardiovascular complications within 30 days, many due to destabilized atherosclerotic plaques. Reverse cholesterol transport (RCT), driven by HDL and Apoa1, protects against plaque progression, but the effects of surgical inflammation on this pathway remain unclear. Using an abdominal laparotomy model in ApoE−/− mice on a Western diet, we isolated the impact of surgical inflammation without confounding blood loss. Surgery acutely impaired RCT and cholesterol efflux, with inflammatory remodeling of HDL marked by elevated SAA1/2 and reduced Apoa1. Coal ash has historically been used as a convenient, on-site solidification material for brines generated from flue gas desulfurization at power plants. As alternate utilization scenarios are developed for coal ash, lignocellulosic (wood-based) sorbents can serve as a highly efficient substitute for the solidification process. Results will be shared for a variety of different brine types: thermally reduced, thermally reduced and neutralized, and reverse osmosis. Stantec recently supported commissioning a flue gas desulfurization (FGD) Wastewater Treatment System in accordance with the 2020 Environmental Protection Agency (EPA) Effluent Limitations Guidelines (ELG) Rule, which requires that coal fired power plants meet their more stringent requirements by December 31, 2025. With the addition of the 2024 EPA ELG Rule, coal fired power plants will now be expected to move to zero liquid discharge (ZLD) systems or permanently cease coal combustion by 2034. Employing a mixed-methods approach, the research integrates archival analysis with digital modeling to recreate the spatial context of the house. The treatment of water from a CCR unit is an important step in the closure strategy for surface impoundments to comply with 40 CFR §257.102(d)(2)(i) of the CCR rule.
